PURPOSE: Potential acute exposure to ionizing radiation in nuclear or radiological accidents presents complex mass casualty scenarios that demand prompt triage and treatment decisions. Due to delayed symptoms and varied response of radiation victims, there is an urgent need to develop robust biomarkers to assess the extent of injuries in individuals.
EXPERIMENTAL DESIGN: The transcription factor Nrf2 is the master of redox homeostasis and there is transcriptional evidence of Nrf2-dependent antioxidant response activation upon radiation. The biomarker potential of Nrf2-dependent downstream tar

A long-standing dogma in the radiation sciences is that energy from radiation must be deposited in the cell nucleus to elicit a biological effect. A number of non-targeted, delayed effects of ionizing radiation have been described that challenge this dogma and pose new challenges to evaluating potential hazards associated with radiation exposure. These effects include induced genomic instability and non-targeted bystander effects. The in vitro evidence for non-targeted effects in radiation biology will be reviewed, but the question as to how one extrapolates from these in vitro observations to

Dietary restriction (DR) has been extensively investigated for its capacity to extend lifespan and mitigate age-associated pathologies. In the context of ionizing radiation (IR), DR-implemented through caloric reduction, selective nutrient restriction, or fasting-based regimens-has emerged as a significant modulator of both cellular and systemic responses to radiation exposure. Growing evidence suggests that DR influences radiation outcomes in a highly context-dependent manner, exerting either protective or sensitizing effects depending on tissue type, metabolic status, timing of intervention,

Exposure to ionizing radiation can cause severe skin damage, leading to the development of cutaneous radiation syndrome. Wound healing of radiation-induced skin injuries proceeds in defined phases that depend on the intensity and type of radiation exposure. Skin damage caused by ionizing radiation can occur not only through accidental exposure, as in the case of the Chernobyl disaster, but also during radiotherapy of tumor patients. The extent of cell damage by ionizing radiation is greater in the presence of oxygen ("oxygen-effect"), most likely by the generation of reactive oxygen species (R

Ionizing radiation can perturb mitochondrial homeostasis and genomic stability, yet its developmental consequences remain insufficiently understood. We investigated how in utero X-ray exposure may affect mitochondrial DNA (mtDNA) regulation and developmental parameters in a mouse model. Pregnant C57BL/6N mice were exposed to 0 (sham-irradiated), 0.05, 0.2, 0.5 and 2 Gy X-rays at gestational day 8, corresponding to the onset of organogenesis. In maternal peripheral blood, mtDNA copy number (mtDNAcn) increased at 2 Gy, while the intact mtDNA ratio, defined as the degree of mtDNA homoplasy estima

Fractionated low dose ionizing radiation (LDIR) can induce various biological effects, which are key potential triggers for the development of radiation-induced lung injury (RILI). This study aims to investigate the effects of fractionated LDIR on early-stage cellular senescence, as well as on the levels of oxidation-antioxidation and DNA damage repair, in mouse lung tissue. C57BL/6J mice were exposed to fractionated LDIR, and subsequent assays were performed to evaluate the biological effects. Compared with the control group, mice in all dose groups showed the following changes: varying degre

Occupational exposure to ionizing radiation in radiology departments is a rare but important concern, given the potential long-term health effects and the regulatory framework surrounding radioprotection. This review analyzes the main risks associated with exposure to X-rays, including cancer, cataracts, radiodermatitis, pregnancy risks, and reduced fertility, as well as the specific contexts in which accidents occur. The majority of significant exposure incidents are reported in interventional radiology, where proximity to the radiation source and prolonged fluoroscopy times increase operator

BACKGROUND: Idiopathic scoliosis is a three-dimensional spinal deformity that typically develops during childhood or adolescence but may affect individuals across the lifespan. Regular monitoring is often necessary to detect progression and assess treatment effectiveness. Radiography remains the clinical gold standard; however, repeated ionizing radiation exposure is associated with increased cancer risks, highlighting the need for reliable, non-invasive, and radiation-free assessment methods. This systematic review and meta-analysis evaluated the diagnostic accuracy and criterion validity of

The one-carbon metabolism enzyme bifunctional methylenetetrahydrofolate dehydrogenase/cyclohydrolase 2 (MTHFD2) is among the most overexpressed proteins across tumors and is widely recognized as a promising anticancer target. While MTHFD2 is mainly described as a mitochondrial protein, a new nuclear function is emerging. Here, we observe that nuclear MTHFD2 protein levels and association with chromatin increase following ionizing radiation (IR) in an ataxia telangiectasia mutated (ATM)- and DNA-dependent protein kinase (DNA-PK)-dependent manner. Furthermore, repair of IR-induced DNA double-str

PURPOSE: After irradiation, double-stranded DNA leaked into the cytoplasm activates the cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) pathway, leading to the production of type I interferon (IFNI). In this study, we sought to probe the effect of ionizing radiation on activity of cGAS-STING-IFNI pathway in normoxic or hypoxic glioma cells and explore a more effective method to activate the signaling pathway, thereby activating the anti-tumor immune response and improving the therapeutic effect of radiotherapy for glioma.

Nanohydrogel particles of polyethylene glycol (PEG), gelatin (GEL), and PEG-GEL mixtures (MIXs) were synthesized with a high electron beam and 60Co gamma-ray radiation. The relatively novel technique of Asymmetrical Flow Field Flow Fractionation (AF4 or AFFFF) coupled to a Multi-Angle Laser Light Scattering (MALLS) detector was mainly used to determine the hydrodynamic diameter (Dh) of the radiation-synthesized PEG, GEL, and PEG-GEL nanohydrogel particles. Our approach to achieving nanohydrogel particles is to enhance the intracrosslinking reactions and decrease the intercrosslinking reactions

Hypertrophic cardiomyopathy (HC) is characterized by the enlargement of individual cardiomyocytes, which is a typical pathophysiological process that occurs in various cardiovascular diseases. Ionizing radiation (IR) is an important independent risk factor for hypertrophic cardiomyopathy, but the underlying molecular mechanism is still unclear. In the present study, we aimed to clarify the role of IR in promoting cardiac hypertrophy and investigate the mechanism by which the SUMO2-mediated SUMOylation of SH3GLB1 affects mitophagy in IR-induced cardiac hypertrophy. In vivo, IR promoted cardiac

Ionizing radiation has dramatic effects on living organisms, causing damage to proteins, DNA, and other cellular components. γ radiation produces reactive oxygen species (ROS) that damage biological macromolecules. Protein modification due to interactions with hydroxyl radical is one of the most common deleterious effects of radiation. The human eye lens is particularly vulnerable to the effects of ionizing radiation, as it is metabolically inactive and its proteins are not recycled after early development. Therefore, radiation damage accumulates and eventually can lead to cataract formation.

Research is an integral part of radiological and nuclear activities; thus, workers may receive different levels of occupational exposure. In the present study, occupational external exposure of workers was studied at the largest radiological and nuclear research institute of Iran from 2015-2020. The activities included research reactors, nuclear fuel cycle, development on radiopharmaceuticals and radioisotopes, particle accelerators, plasma and fusion, and nuclear agriculture. The average doses of the monitored and exposed workers were calculated to be 0.095 and 0.575 mSv, respectively, which

Currently, our knowledge of how different cell types in a tissue microenvironment respond to low and high linear energy transfer (LET) radiation is highly restricted. In this study, a comparative analysis was performed on γ-ray-induced DNA damage and repair in primary human melanocytes and keratinocytes isolated from 3 donors. Our study demonstrates a modest interindividual variability in both melanocytes and keratinocytes in terms of both spontaneous and ionizing radiation (IR)-induced 53BP1 foci formation and persistence. Melanocytes, in general, showed a slightly elevated (1.66-2.79 folds m

PURPOSE: Precise molecular and cellular mechanisms of radiation-induced dermatitis are incompletely understood. Histone variant H2A.J is associated with cellular senescence and modulates senescence-associated secretory phenotype (SASP) after DNA-damaging insults, such as ionizing radiation (IR). Using ex vivo irradiated cultured foreskin, H2A.J was analyzed as a biomarker of radiation-induced senescence, potentially initiating the inflammatory cascade of radiation-induced skin injury.
METHODS: Human foreskin explants were collected from young donors, irradiated ex vivo with 10 Gy, and culture
