18 F-FDG PET/CT Five Days Post First Administration of Semaglutide : Side Effects in One Shot.

We present the case of a 60-year-old woman with metastatic breast cancer who underwent 18 F-FDG PET/CT for treatment response assessment. Beyond oncologic findings, the scan revealed abnormal gastric retention, gallbladder, colonic, and renal uptake patterns suggestive of Semaglutide-induced gastroparesis, cholecystitis, colitis, and possible acute kidney injury secondary to dehydration. Suspicions were confirmed by clinical evaluation. Symptoms resolved after supportive therapy and withholding of semaglutide. To our knowledge, this is the first report of multi-organ side effects seen on 18 F-…

Development of NAFLD-Specific Human Liver Organoid Models on a Microengineered Array Chip for Semaglutide Efficacy Evaluation.

Progressive non-alcoholic fatty liver disease (NAFLD) may culminate in severe complications, including fibrosis, cirrhosis and hepatocellular carcinoma, yet therapeutic breakthroughs remain elusive, necessitating novel pharmacological strategies. Semaglutide, a glucagon-like peptide-1 (GLP-1) receptor agonist clinically approved for type 2 diabetes and obesity management, has demonstrated pleiotropic effects in preclinical NAFLD models. In this study, we investigated semaglutide's therapeutic efficacy and mechanisms in a human liver organoids (hLOs) model of NAFLD. Utilising microengineered ar…

FABP4, marker of worse prognosis in cardiovascular disease, induces neutrophil's proatherogenic phenotype which is modulated by semaglutide.

Dysfunctional epicardial adiposity is a risk factor for coronary artery disease (CAD). Its genesis is associated with an upregulation of fatty acid binding protein 4 (FABP4) levels, which might exert paracrine inflammatory and atherogenic mechanisms on the cardiovascular system. We aimed to study the prognosis of patients with high systemic FABP4, its association with a neutrophil proatherogenic phenotype, involved mechanisms and its modulation by semaglutide, a glucagon-like peptide-1 receptor agonist (GLP-1ra). Higher plasma FABP4 levels than 50 ng/mL were associated with heart failure (HF) …

Spinal Cord Stimulation - Device Revision After Weight Loss in a Patient on Chronic Semaglutide - A Case Report.

BACKGROUND: Spinal cord stimulation (SCS) is an effective treatment option for patients suffering from chronic intractable pain of the trunk and/or limbs. One of the potential adverse effects of SCS is discomfort or pain at the site of the implantable pulse generator (IPG). It is a known phenomenon that patient weight loss may exacerbate or increase discomfort or pain at the IPG implantation site. OBJECTIVE: This case report aims to educate neuromodulation clinicians on the potential impact of glucagon-like peptide-1 receptor agonists (GLP-1RA) medications on patient weight loss and decreased

Semaglutide Treatment Effects on Liver Fat Content in Obese Subjects with Metabolic-Associated Steatotic Liver Disease (MASLD).

Background: Metabolic-dysfunction-associated steatotic liver disease (MASLD) represents a major clinical complication of obesity. Methods: In this study, we used magnetic resonance (MR) methods to determine the effect of obesity treatment with semaglutide, a GLP-1 receptor agonist, on the liver fat content and selected metabolic variables. We investigated whether treatment would affect the acute response of liver fat to glucose and fructose administration and whether it would affect the fatty acid profile of VLDL-triglycerides. Sixteen obese non-diabetic men underwent a 16-week dietary interve…

Semaglutide ameliorates cardiac remodeling in male mice by optimizing energy substrate utilization through the Creb5/NR4a1 axis.

Semaglutide, a glucagon-like peptide-1 receptor agonist, is clinically used as a glucose-lowering and weight loss medication due to its effects on energy metabolism. In heart failure, energy production is impaired due to altered mitochondrial function and increased glycolysis. However, the impact of semaglutide on cardiomyocyte metabolism under pressure overload remains unclear. Here we demonstrate that semaglutide improves cardiac function and reduces hypertrophy and fibrosis in a mouse model of pressure overload-induced heart failure. Semaglutide preserves mitochondrial structure and functio…

Comparative effectiveness of sleeve gastrectomy and semaglutide for weight loss and metabolic outcomes: a prospective non-randomized study.

BACKGROUND: Bariatric surgery and pharmacotherapy represent two major strategies for the treatment of obesity. Sleeve gastrectomy (SG) is the most widely performed surgical procedure and is supported by long-term evidence, while semaglutide, a glucagon-like peptide-1 receptor agonist (GLP-1 RA), has recently emerged as an effective medical therapy. However, prospective real-world studies directly comparing these interventions are limited. METHODS: We conducted a prospective, non-randomized trial at a single tertiary center (ChiCTR2300070632). Adults with obesity were allocated to SG (n = 36) …

Semaglutide use in a patient with a long-term ileostomy: A case report highlighting clinical risks and evidence gaps.

BACKGROUND: Glucagon-like peptide-1 receptor agonists (GLP-1 RAs), such as semaglutide, dulaglutide, liraglutide, and exenatide, are increasingly used to treat type 2 diabetes and support weight loss. These agents function by enhancing glucose-dependent insulin secretion, suppressing inappropriate glucagon secretion, slowing gastric emptying, and regulating appetite through central nervous system pathways. Since semaglutide's approval in 2017 and U.S. market availability in 2018, its use has expanded significantly. However, there remains limited literature evaluating its safety and efficacy in…
