BACKGROUND: There is evidence that the rapid rise in Streptococcus pneumoniae (SP) antimicrobial resistance seen in other countries may have commenced in Australia. Streptococcus pneumoniae carriage and resistance levels are described for urban Northern Territory children in day care.
METHODS: A prospective cohort study was conducted of 250 children in nine Darwin day care centres between 24 March and 15 September 1997. Each fortnight nasopharyngeal swabs were collected from children, and parents were interviewed about medications administered.
RESULTS: Streptococcus pneumoniae was detected …

Streptococcus Pneumoniae infections are a common cause of bacteremia and community-acquired pneumonia in HIV-positive individuals. Bone and joint infections as a result of S. pneumoniae are extremely rare. We report the first case of S. pneumoniae septic arthritis and osteomyelitis in an HIV-positive adult patient. The patient was successfully treated with oral levofloxacin.

OBJECTIVE: To assess the rates of intermediate and high-level resistance to penicillin among Streptococcus pneumoniae isolates and to identify clonal relationship of isolates within the different serotypes by means of pulsed-field gel electrophoresis. METHODS: We studied all clinical isolates obtained between April 1995 and March 1996 from patients admitted to 10 hospitals in eastern France. Antibiotic susceptibility testing and serotyping were performed on all isolates. The genetic polymorphism of isolates susceptible, intermediately resistant and highly resistant to penicillin was studied by…

Seven patients with acute mastoiditis are reported. They were treated at Chiba Children's Hospital from 1991 to 1996. Patients included one 4-year-old child and 6 one-year-old infants. Except the 4-year-old girl whose body weight was 17 kg, all infants body weights were under 11 kgs. Chief complaints were ear discharge, retroauricular swelling and high fever (> 38 degrees C). Streptococcus pneumoniae was isolated in the 6 infants. One patient was diagnosed with penicillin-sensitive Streptococcus pneumoniae (PSSP), three patients were diagnosed with penicillin-insensitive Streptococcus pneumoni…

Pneumococcal meningitis in St. Petersburg in the period 1985-1991 occurred in 1.7-2.3 children per 100,000 annually. The most common serotypes among pneumococcal strains isolated from patients with meningitis were 19, 1, 6, 15, and 2, whereas, among the capsulated strains isolated from carriers, type 3 predominated. Only one third of strains from cases of meningitis were highly virulent for mice (types 1, 2, 3). Hyaluronidase was produced by all the 39 studied strains, 22 (84.6 +/- 7.1%) out of 26 strains from patients with otitis media, and only by 15 (11.5 +/- 2.8%) out of 130 strains isolat

Streptococcus pneumoniae is a leading cause of infectious disease-related illness and death in the United States, accounting for an estimated 3000 cases of meningitis, 50,000 cases of bacteremia, 50,000 cases of pneumonia, and 7 million cases of acute otitis media each year. Penicillin has been the antibiotic of choice for the treatment of infections caused by S. pneumoniae; since the mid-1980s, the prevalence of penicillin-resistant S. pneumoniae has increased substantially worldwide. In Canada, a strain of pneumococcus with reduced susceptibility to penicillin was first reported in 1974; bas…

Drug-resistant strains of Streptococcus pneumoniae have now been reported from all continents and have become the predominant pathogens in some areas; many strains are resistant to multiple agents. Because of the importance of pneumococci in the etiology of meningitis, the criteria used to assess susceptibility are conservative: strains with minimal inhibitory concentrations (MICs) of penicillin G of less than or equal to 0.06 microgram/mL are regarded as susceptible, those with MICs of 0.1-1 microgram/mL are considered intermediately resistant, and those with MICs of greater than 1 microgram/…

Five of six multiply drug-resistant clinical isolates of Streptococcus pneumoniae from South Africa demonstrated penicillin tolerance. In contrast to the common wild-type strains of pneumococci, treatment of the tolerant strains with penicillin above the minimum inhibitory concentration did not induce cell wall degradation, lysis, or leakage of intracellular components, and the rate of loss of viability was reduced compared with that of nontolerant strains. While these South African strains contained lower specific activity of autolytic enzyme than did nontolerant strains, the residual autolyt…

C-reactive protein (CRP), an acute-phase reactant which binds to phosphocholine (PC) in the pneumococcal cell wall, and anti-PC antibodies are protective against experimental pneumococcal bacteremia in mice. To determine the relative opsonic capacities of CRP and anti-PC compared with those of antibodies against pneumococcal capsular polysaccharides (anti-PCP), we correlated in vitro opsonic activity for serotype 7F Streptococcus pneumoniae with concentrations of CRP, anti-PC, and anti-type 7F PCP in human sera from 10 normal subjects and 38 patients with sickle cell (SS) disease, a high-risk …

The specific capsular polysaccharide of Streptococcus pneumoniae type 9N (American type 9) contains D-glucose, D-glucuronic acid, 2-acetamido-2-deoxy-D-glucose, and 2-acetamido-2-deoxy-D-mannose in the molar ratio of 2:1:1:1. Accumulated data from spectroscopic (13C and 1H nuclear magnetic resonance) and methylation analyses of the native and specifically degraded polysaccharide indicated that it was linear and composed of the following pentasaccharide repeating unit; -4)-alpha-D-GlcpA-(1 leads to 3)-alpha-D-Glcp-(1 leads to 3)-beta-D-ManpNAc-(1 leads to 4)-alpha-D-Glcp-(1 leads to 4)-beta-D -…

Infections caused by Streptococcus pneumoniae (Spn, pneumococcus), manifesting as pneumonia, meningitis, and otitis media, represent a leading cause of severe morbidity and mortality globally. Escalating antibiotic resistance and serotype replacement by conjugate vaccines underscore the need for serotype-independent therapies. Hypothiocyanous acid (HOSCN), a reactive oxidant naturally produced by the host innate immune system, is formed through the interaction of hydrogen peroxide, thiocyanate anions (SCN-), and peroxidase enzymes. HOSCN is produced at mucosal surfaces, including the respirato…

Background. Infections caused by Streptococcus pneumoniae are a public health problem worldwide, and penicillin-non-susceptible isolates are a priority for the World Health Organization, which requires further research and development of new antibiotics and vaccines.Aim. To describe the global pneumococcal sequence clusters (GPSCs) among penicillin non-susceptible S. pneumoniae isolates obtained from patients with invasive pneumococcal disease in Colombia after the introduction of PCV10 (Synflorix, GlaxoSmithKline) to generate data on the genetic structure of pneumococcal populations with diff…

PURPOSE: Streptococcus pneumoniae (S. pneumoniae) causes disease in multiple body systems, including at the ocular surface and in the lungs. S. pneumoniae IgA1 protease cleaves and inactivates human IgA1, which is present in high concentrations at mucosal surfaces. Mice lack IgA1, but studies have shown that the gene encoding IgA1 protease is required for virulence in pneumonia in mice. This study aimed to determine if the gene is required for virulence in the mouse keratitis model.
METHODS: S. pneumoniae D39 or the isogenic IgA1 protease mutant, D39Δiga, was applied to the scratched ocular s…

BACKGROUND: Streptococcus pneumoniae is a Gram-positive, facultative anaerobic bacterium and a leading cause of community-acquired pneumonia, meningitis, bacteremia, and otitis media, responsible for an estimated 1.6 million deaths annually. Despite widespread polysaccharide and conjugate vaccine programs and decades of antibiotic therapy, pneumococcal disease continues to pose major public health challenges. These challenges are increasingly understood to reflect the organism's sophisticated capacity for adaptive recombination, allowing it to exploit every selective pressure applied through v…

Synergy between influenza A virus (IAV) and Streptococcus pneumoniae is a long-recognized and clinically important problem. Recent work has demonstrated that IAV particles can directly bind to the bacterial surface and that bacterial-viral complexes exhibit enhanced bacterial colonization and invasive disease, increased viral environmental survival leading to increased efficacy of airborne transmission, and enhanced vaccine response to both pathogens over simultaneous co-infection without direct interactions. However, the molecule(s) responsible for mediating the direct interaction are yet to …

Streptococcus pneumoniae is a major respiratory pathogen and a commensal bacterium of the nasopharynx. However, it remains unclear how commensal S. pneumoniae switches to a pathogenic form. Recently, a 3D human lung tissue model was established and adopted as a bacterial infection model. Here, we analysed the effects of commensal bacteria on the pathogenicity of S. pneumoniae in human tissues using a model constructed by interstitial layer of human fibroblasts mixed with collagen and an epithelial layer of layered human lung epithelial cells. When S. pneumoniae infected to the model, severe in…

Streptococcus pneumoniae (SPN) is a significant pathogen causing pneumonia and meningitis, particularly in vulnerable populations like children and the elderly. Available pneumonia vaccines have limitations since they only cover particular serotypes and have high production costs. The emergence of antibiotic-resistant SPN strains further underscores the need for a new, cost-effective, broad-spectrum vaccine. Two potential vaccine candidates, CbpA and PspA, were identified, and their B-cell, CTL, and HTL epitopes were predicted and connected with suitable linkers, adjivant and PADRE sequence. T…

Bacteria have evolved complex transcriptional regulatory networks, as well as many diverse regulatory strategies at the RNA level, to enable … -acting regulatory RNAs (including several riboswitches and a T-box), in the opportunistic pathogen Streptococcus pneumoniae TIGR4. By evaluating native locus mutants of each regulator that result in constitutively active or repressed expression, we establish that growth defects in planktonic culture are associated with constitutive repression of gene expression, while co …
