Photoprotective effect of topically applied superoxide dismutase on sunburn reaction in comparison with sunscreen.

Photoprotective effect of topically applied superoxide dismutase (SOD) to guinea pig skin was compared with a commercially available sunscreen agent after a single exposure to UVB. While cutaneous SOD activity was remarkably decrease in non-treated control animals, both topical SOD and sunscreen agent significantly reduced the decrease in skin SOD activity after UVB irradiation. However, only the sunscreen agent successfully reduced erythema reaction 24h after irradiation but topical SOD failed. These findings suggest that topical SOD protects skin from photo-oxidative damage without affecting

The immunocytochemical localization of superoxide dismutase in the enterocytes of the avian intestine: the effect of vitamin D3.

Both light microscopical and electron microscopical immunocytochemical techniques were utilized to localize CuZn-superoxide dismutase (SOD) in the duodenum of normal, rachitic and vitamin-D3-replete chicks. This enzyme catalyses the dismutation of the superoxide anion, a toxic free radical generated during the normal aerobic metabolism of most respiring cells. Light microscopy showed no SOD activity associated with the duodenal enterocytes of normal and rachitic chicks. However, in rachitic animals subsequently treated with vitamin D, i.e. vitamin-D-replete chicks, intense immunoreactivity for

Auto-oxidation of dialuric acid, divicine and isouramil. Superoxide dependent and independent mechanisms.

The toxicity of dialuric acid to pancreatic beta cells, and the haemolytic action of divicine and isouramil involve auto-oxidation and redox cycling reactions. Divicine and isouramil are produced on hydrolysis of the fava bean glycosides, vicine and convicine. The mechanism of auto-oxidation of the three compounds as well as the acid hydrolysis product of vicine (provisionally assigned the structure 2-amino-4,5,6-trihydroxypyrimidine) has been studied. All four pyrimidines auto-oxidized rapidly at neutral pH, generating H2O2 by an O2-dependent chain mechanism. Superoxide dismutase inhibited th

Iron-mediated induction of sister-chromatid exchanges by hydrogen peroxide and superoxide anion.

When Chinese hamster fibroblasts were exposed to hydrogen peroxide or to a system consisting of xanthine oxidase and hypoxanthine, which generates superoxide anion plus hydrogen peroxide, sister-chromatid exchanges (SCEs) were formed in a dose-dependent manner. When the iron-complexing agent o-phenanthroline was present in the medium, however, the production of these SCEs was completely inhibited. This fact indicates that the Fenton reaction: Fe2+ + H2O2----OH0 + OH- + Fe3+ is responsible for the production of SCEs. When O2- and H2O2 were generated inside the cell by incubation with menadione,

Characterization of the O2-induced manganese-containing superoxide dismutase from Bacteroides fragilis.

A manganese-containing superoxide dismutase (MnSOD) has been isolated from extracts of O2-induced Bacteroides fragilis. The enzyme, Mr 43,000, was a dimer composed of noncovalently associated subunits of equal size. A preparation whose specific activity was 1760 U/mg had 1.1 g-atoms Mn, 0.3 g-atoms Fe, and 0.2 g-atoms Zn per mol dimer. Exposing the enzyme to 5 M guanidinium chloride, 20 mM 8-hydroxyquinoline abolished enzymatic activity. Dialysis of the denatured apoprotein in buffer containing either Fe (NH4)2(SO4)2 or MnCl2 restored O2-. scavenging activity. The iron-reconstituted enzyme was

[Catalase and superoxide dismutase activity in isogenous bacterial strains with different degrees of radioresistance].

Superoxide dismutase and catalase activity has been studied in isogenous strains of various radioresistance bacteria. In mutants Micrococcus radiodurans having defects in the systems of DNA repair the superoxide dismutase activity is lower than in cells of wild type. The changes of catalase and superoxide dismutase activity have not been revealed in investigated strains Escherichia coli differing in radioresistance. It has been concluded that the survival of bacteria exposed to ionizing radiation is determined by the effectiveness of DNA repair systems realiability of which depends on the cata

Chromosome instability in human and murine autoimmune disease: anticlastogenic effect of superoxide dismutase.

Increased chromosome breakage is observed in lymphocyte cultures from patients with so-called autoimmune diseases also in the animal model, the NZB mouse. A clastogenic agent was detected in the serum of patients and of NZB mice, that induces also chromosome breaks in cells of healthy individuals. In simultaneous cultures set up with or without superoxide dismutase in 5 patients with progressive systemic sclerosis, 5 patients with systemic lupus erythematosus and 5 patients with rheumatoid arthritis, highly significant differences in the incidence of chromosome breakage were observed. The aber

Superoxide Radical Mediated Branch and Crosslink Growth of FCC Kernel in Gold Nanoclusters.

FCC (face-centered cubic) structures are found to be highly prevalent in both bulk Au and Au nanoclusters, but achieving precise tailoring or the formation of new stacking modes based on FCC structure has proved challenging. Here, we show a radical-induced structure transformation strategy using persulfates that results in the formation of three new nanoclusters: Au41(SR)25, Au46(SR)26, and Au49(SR)27 (SR: thiolate). The double helices of Au4 assemblies in their kernel structures are observed to adopt branch and cross-linked growth beyond that of linear growth in the previous FCC-kernelled str

Mutations in the superoxide-responsive regulator SoxR confer fluoroquinolone resistance in Stenotrophomonas maltophilia.

Stenotrophomonas maltophilia is a multidrug-resistant Gram-negative opportunistic pathogen. Its antimicrobial resistance arises from intrinsic traits and acquired mechanisms. Overexpression of drug efflux transporters is one of the key mechanisms mediating high-level antibiotic resistance. These transporters are regulated by transcriptional factors, including SoxR, a superoxide-responsive regulator containing an iron-sulfur [2Fe-2S] cluster. SoxR controls the multidrug efflux pumps MfsA and SmeVWX, as well as superoxide dismutase SodA1. To examine how soxR mutations affect antimicrobial resist

Superoxide dismutase in intervertebral disc degeneration: from pathophysiological mechanism to therapeutic strategies.

Reactive oxygen species (ROS) induce inflammation, senescence and various forms of cell death in nucleus pulposus cells. By promoting these reactions and their interplay, ROS significantly accelerate intervertebral disc degeneration (IDD). Conventional surgical interventions and analgesics can alleviate symptoms but fail to halt the disease progression mechanistically. Eliminating ROS may serve as a fundamental therapeutic approach to mitigate IDD. Within the endogenous antioxidant defense system for scavenging ROS, superoxide dismutase (SOD) serves as the first line of defense, playing an irr

In-Gel Zymography of Amyotrophic Lateral Sclerosis-Associated Variants of Superoxide Dismutase-1.

In-gel zymography allows the separation of protein via electrophoresis and subsequent measurement of enzymatic activity of enzymes from biological mixtures. The antioxidant enzyme superoxide dismutase 1 (SOD1) is an important cytosolic oxygen radical scavenging enzyme that has been implicated in a range of pathologies, including cancer, metabolic and neurodegenerative diseases, most notably amyotrophic lateral sclerosis (ALS). Here, we describe a method to detect and compare SOD1 activity from both cell and tissue samples. This method can be utilized to compare differences between ALS-associat

[Role of superoxide dismutase in acute pancreatitis: from antioxidant protection to gene regulation].

We present modern data on the role of superoxide dismutase (SOD) in antioxidant protection and gene regulation in acute pancreatitis. Antioxidant enzymes are essential in pathogenesis of numerous diseases. SOD is one of the key enzymes of antioxidant system. In this review, we analyzed activity of this enzyme depending on various factors, mechanisms and role in physiological and pathological processes, in particular, acute pancreatitis. SOD is significantly less active in patients with severe acute pancreatitis accompanied by renal failure, severe circulatory disorders and high mortality. Ther

Differential expression of various isoforms of superoxide dismutase in the cells of the human exocrine pancreas.

OBJECTIVES: Superoxide dismutase (SOD) is an enzyme that plays a crucial role in protecting cells from oxidative damage. Our study aims to address the lack of papers simultaneously analyzing the immunoreactivity of all three distinct isoforms of SOD in human exocrine pancreas cells. BACKGROUND: Superoxide dismutases (SODs) facilitate the conversion of superoxide radicals into less harmful substances. By neutralizing superoxide radicals, SODs help prevent the formation of highly reactive and destructive species that can adversely affect manifold cellular components. METHODS: The study analyze

Molecular Engineering of Pure Superoxide Radical Photogenerator for Hypoxia-Tolerant Tumor Theranostics.

Photodynamic therapy (PDT) is a promising cancer treatment, but limited oxygen supply in tumors (hypoxia) can hinder its effectiveness. This is because traditional PDT relies on Type-II reactions that require oxygen. Type-I photosensitizers (PSs) offer a promising approach to overcome the limitations of tumor photodynamic therapy (PDT) in hypoxic environments. To leverage the advantages of Type-I PDT, the design and evaluation of a series of Type-I PSs for developing pure Type-1 PSs, by incorporating benzene, thiophene, or bithiophene into the donor-acceptor molecular skeleton are reported. Uric acid/superoxide dismutase can predict progression of gestational hypertension to preeclampsia.

INTRODUCTION: Preeclampsia (PE), at early onset, is likely to be diagnosed as gestational hypertension (GH). Some cases of GH rapidly progress to PE within a short period of time, increasing the mortality rate of pregnant women and adverse events in neonates during the peripartum period. Oxidative stress participates in the occurrence and progression of PE. However, it is unknown whether the progression of GH to PE can be predicted. METHODS: A total of 1548 patients diagnosed with PE (649 cases) or GH (899 cases) from January 2016 to June 2022 were selected as the study subjects. The 1548 pat
