Stratification of surgical outcomes in trigeminal neuralgia using multimodal data.

Chronic pain remains a challenge for clinicians, with limited individualized predictive tools that can aid with diagnosis, disease course, or prediction of treatment outcomes. We hypothesized that a comprehensive analysis, encompassing a patient's complete pain-related clinical data, medical history and brain imaging, can identify key contributors linked to surgical outcomes and stratify specific outcome categories for trigeminal neuralgia (TN)-chronic facial pain syndrome. Using supervised and unsupervised machine learning approaches, we analysed data from 102 subjects with classical TN. Pre-

Profiling of Cardiogenic Shock: Incorporating Machine Learning Into Bedside Management.

Cardiogenic shock (CS) is a complex clinical syndrome with various etiologies and clinical presentations. Despite advances in therapeutic options, mortality remains high, and clinical trials in the field are complicated in part by the heterogeneity of CS patients. More individualized targeted therapeutic approaches might improve outcomes in CS, but their implementation remains challenging. The present review discusses current and emerging machine learning-based approaches, including unsupervised and supervised learning methods that use real-world clinical data to individualize therapeutic stra

Predicting in-hospital all-cause mortality in heart failure using machine learning.

BACKGROUND: The age of onset and causes of heart failure differ between high-income and low-and-middle-income countries (LMIC). Heart failure patients in LMIC also experience a higher mortality rate. Innovative ways that can risk stratify heart failure patients in this region are needed. The aim of this study was to demonstrate the utility of machine learning in predicting all-cause mortality in heart failure patients hospitalised in a tertiary academic centre. METHODS: Six supervised machine learning algorithms were trained to predict in-hospital all-cause mortality using data from 500 conse

A machine learning approach to identify patients at risk for long-term consequences after pulmonary embolism.

Pulmonary embolism (PE) can result in long-term sequelae, such as post-PE syndrome, including persistent dyspnea and chronic thromboembolic pulmonary hypertension (CTEPH). Existing prediction tools for severe post-PE complications lack sensitivity and specificity. This study aimed to develop a machine learning model to identify patients at risk for long-term consequences after PE. Using data from the RIETE registry, the largest prospective international PE registry, we developed supervised machine learning models to identify patients at increased risk of CTEPH and post-PE syndrome. Our approac

Testing regular expression searches and machine learning models to determine housing instability and low income status from primary care electronic medical record data in Toronto, Ontario.

BACKGROUND: Housing and income are important social determinants of health (SDoH). Primary care providers often do not have information about these determinants, which could be used to support equitable health system planning and care delivery. The aim of this study was to use primary care electronic medical record (EMR) data to test two approaches (machine learning and regular expression searches) to obtain information about patients' housing instability and low income status. METHODS: We used de-identified EMR data from the St. Michael's Hospital Academic Family Health Team (Toronto, Ontari

A novel optically gated thin-film transistor sensor for real-time chemical differentiation using machine-learning analysis.

We present an optically gated thin-film transistor sensor that distinguishes chemicals through illumination-induced transient electrical responses. The device consists of a p-type silicon substrate with native oxide and an amorphous Ge2Se3 photogating layer, and operates without reference electrodes or surface functionalization. Structured light-pulse sequences produce time-dependent electrical signatures whose features differ across chemical environments. These transient responses are analyzed using supervised machine-learning models to demonstrate real-time chemical differentiation in contro

Automated CLL cell population detection using a weakly supervised approach and CLL MRD flow cytometry data.

Minimal/measurable residual disease detection is routinely performed as part of post-diagnostic treatment plans for many types of cancer, for which multiparameter flow cytometry is one possible modality frequently used. We propose a machine learning approach for binary prediction of minimal residual disease status with flow cytometry data. Our method involves the projection of cells from the original feature space to a low-dimensional embedding in which cells are clustered by similarity, and the cluster-wise cell proportions are used for prediction as well as regression. This is a weakly super

Fusion of Supervised Feature Selection and Unsupervised Clustering for Multinuclei Classification of MER Signals in DBS for Parkinson's Disease.

In deep brain stimulation (DBS) surgery for Parkinson's disease (PD), the accurate intraoperative identification of key nuclei-such as the subthalamic nucleus (STN)-is critical to ensuring therapeutic efficacy. However, current approaches heavily depend on expert annotations, and classification tasks are typically confined to binary distinctions between STN and non-STN regions. This limitation hampers the real-time recognition of complex and diverse neuroanatomical structures during PD-related DBS procedures. To overcome this challenge, we propose a three-class classification strategy for micr

Decision making on vestibular schwannoma treatment: predictions based on machine-learning analysis.

Decision making on the treatment of vestibular schwannoma (VS) is mainly based on the symptoms, tumor size, patient's preference, and experience of the medical team. Here we provide objective tools to support the decision process by answering two questions: can a single checkup predict the need of active treatment?, and which attributes of VS development are important in decision making on active treatment? Using a machine-learning analysis of medical records of 93 patients, the objectives were addressed using two classification tasks: a time-independent case-based reasoning (CBR), where each

Improving respiratory disease detection through SSL-enhanced acoustic analysis and exercise-rest measurements.

BACKGROUND: Voice analysis has emerged as a promising non-invasive approach for monitoring respiratory and systemic health conditions. However, subtle physiological alterations are often difficult to capture using recordings collected at rest. In addition, combining traditional acoustic descriptors with modern self-supervised speech representations may provide complementary information for clinical voice analysis. OBJECTIVES: This study evaluates a generalized screening model integrating stress-induced acoustic analysis with machine learning. We investigate how physical exertion and the fusio

Understanding Mental Health Issues in Different Subdomains of Social Networking Services: Computational Analysis of Text-Based Reddit Posts.

BACKGROUND: Users increasingly use social networking services (SNSs) to share their feelings and emotions. For those with mental disorders, SNSs can also be used to seek advice on mental health issues. One available SNS is Reddit, in which users can freely discuss such matters on relevant health diagnostic subreddits. OBJECTIVE: In this study, we analyzed the distinctive linguistic characteristics in users' posts on specific mental disorder subreddits (depression, anxiety, bipolar disorder, borderline personality disorder, schizophrenia, autism, and mental health) and further validated their

Explainable AI-Assisted Label-Free Raman Biosensing Reveals Therapy-Associated Spectral Signatures in Melanoma Tumors.

Sensitive detection of treatment-associated Raman spectral alterations in tumor tissues remains challenging, particularly when such changes are not readily apparent from conventional morphological evaluation. Here, we developed a label-free Raman biosensing strategy combined with explainable machine learning to characterise treatment-associated spectral signatures in melanoma tumours. A B16-F10 melanoma-bearing mouse model was used to compare untreated and PBS-treated controls with cohorts receiving immune checkpoint blockade, anti-angiogenic intervention, or combination therapy. Machine learning-enabled multi-omics discovery of prognostic biomarkers and signaling targets in pancreatic cancer.

Pancreatic ductal adenocarcinoma (PDAC) remains difficult to subtype using single omics layers. We conducted an exploratory investigation integrating reverse-phase protein array (RPPA) and DNA methylation data from the cancer genome atlas (TCGA)- pancreatic adenocarcinoma (PAAD) to assess the feasibility of multi-omics subtyping, alongside a supervised machine learning analysis of a small gene expression omnibus (GEO) transcriptomic cohort (n = 26) to identify candidate diagnostic genes. RPPA-based K-means clustering suggested a weak, possible two-subtype structure (silhouette ≈ 0.16) that rem

Artificial intelligence for quality control of oscillometry measures.

BACKGROUND: The forced oscillation technique (FOT) allows non-invasive lung function testing during quiet breathing even without expert guidance. However, it still relies on an operator for excluding breaths with artefacts such as swallowing, glottis closure and coughing. This manual selection is operator-dependent and time-consuming. We evaluated supervised machine learning methods to exclude breaths with artefacts from data analysis automatically. METHODS: We collected 932 FOT measurements (Resmon Pro Full, Restech) from 155 patients (6-87 years) following the European Respiratory Society (

Data-driven supervised learning of a viral protease specificity landscape from deep sequencing and molecular simulations.

Biophysical interactions between proteins and peptides are key determinants of molecular recognition specificity landscapes. However, an understanding of how molecular structure and residue-level energetics at protein-peptide interfaces shape these landscapes remains elusive. We combine information from yeast-based library screening, next-generation sequencing, and structure-based modeling in a supervised machine learning approach to report the comprehensive sequence-energetics-function mapping of the specificity landscape of the hepatitis C virus (HCV) NS3/4A protease, whose function-site-spe
