Toxicological impact assessment of the marine diatoms Skeletonema costatum and Navicula sp., using the anti-fouling agents CuSO4, DCOIT, and ECONEA.

Antifouling paints containing various chemical antifoulants are applied to prevent the attachment of marine organisms on submerged structures. However, these compounds can be released into the marine environment and adversely affect non-target marine species. This study compared the toxic effects of copper sulfate (CuSO₄), 4,5-dichloro-2-n-octyl-4-isothiazolin-3-one (DCOIT), and ECONEA alone and in combination on the periphytic diatom Navicula sp. and the planktonic diatom Skeletonema costatum. The 72 h EC50 values revealed a toxicity order of DCOIT>ECONEA>CuSO4. For Navicula sp., EC50 values

The influence of soil organic matter content on the toxicity of pesticides to the springtail Folsomia candida.

The European environmental risk assessment (ERA) of pesticides to soil invertebrates applies a correction factor (CF) of 2 to endpoints derived from toxicity tests with lipophilic pesticides (log Kow > 2) to correct for differences in organic matter (OM) content between artificial soil (AS) and natural soils. Because the applicability of this CF to springtails has never been assessed, this study investigated the influence of soil OM content on the toxicity of five pesticide active substances differing in lipophilicity to the springtail Folsomia candida. Toxicity tests following Organisation fo…

Testing Strategies for Metabolite-Mediated Neurotoxicity.

Compounds, which rely on metabolism to exhibit toxicity, pose a challenge for next-generation risk assessment (NGRA). Since many of the currently available non-animal new approach methods (NAMs) lack metabolic activity, their use may lead to an underestimation of the true hazard to humans (false negative predictions). We explored here strategies to deal with metabolite-mediated toxicity in assays for developmental neurotoxicity. First, we present an overview of substances that may serve as potential positive controls for metabolite-related neurotoxicity. Then, we demonstrate, using the MitoMet

Rapid fate of benzophenone in artificial soil and comprehensive toxicity assessment in Eisenia fetida.

Benzophenone (BP) is a widely used organic UV filter in personal care products. Despite its frequent detection in soil ecosystems, its toxicity to soil biota has not been extensively explored. This study investigates the effects of BP on earthworms (Eisenia fetida) exposed in artificial soil. Soil degradation kinetics followed a single first-order model at low (1.69 mg/kg) and medium (3.39 mg/kg) concentrations, and a biphasic double first-order in parallel model at high (16.9 mg/kg) concentration. The calculated DT50 (time to 50 % degradation) and DT90 (time to 90 % degradation) ranged from 0

Toxicity assessment and bioimaging potential of carbon dots synthesized from banana peel in zebrafish model.

Zebrafish serve as a pivotal model for bioimaging and toxicity assessments; however, the toxicity of banana peel-derived carbon dots in zebrafish has not been previously reported. The aim of this study was to assess the toxicity of carbon dots derived from banana peel in zebrafish, focusing on two types prepared through hydrothermal and pyrolysis methods. Banana peels were synthesized using hydrothermal and pyrolysis techniques and then compared for characteristics, bioimaging ability, and toxicity in zebrafish as an animal model. Pyrolysis-derived banana peel and hydrothermal-derived banana p

The use of weight-of-evidence approaches to characterize developmental toxicity risk for therapeutic monoclonal antibodies in humans without in vivo studies.

Regulatory guidance for global drug development relies on animal studies to evaluate safety risks for humans, including risk of reproductive toxicity. Weight-of-evidence approaches (WoE) are increasingly becoming acceptable to evaluate risk. A WoE for developmental risk of monoclonal antibodies (mAbs) was evaluated for its ability to retrospectively characterize risk and to determine the need for further in vivo testing based on the remaining uncertainty. Reproductive toxicity studies of 65 mAbs were reviewed and compared to the WoE. Developmental toxicities were absent in 52/65 (80%) mAbs. La

Proteomics Study of Benzene Metabolite Hydroquinone Induced Hematotoxicity in K562 Cells.

OBJECTIVE: Hydroquinone (HQ), one of the phenolic metabolites of benzene, is widely recognized as an important participant in benzene-induced hematotoxicity. However, there are few relevant proteomics in HQ-induced hematotoxicity and the mechanism hasn't been fully understood yet. METHODS: In this study, we treated K562 cells with 40 μmol/L HQ for 72 h, examined and validated protein expression changes by Label-free proteomic analysis and Parallel reaction monitoring (PRM), and performed bioinformatics analysis to identify interaction networks. RESULTS: One hundred and eighty-seven upregulat

Joint effects of temperature and copper exposure on developmental and gene-expression responses of the marine copepod Tigriopus japonicus.

There is growing contamination of copper (Cu) in the marine environment, particularly after the ban of organotin compounds and the increase of the use of Cu-based antifouling paints. Although there are increasing research interests in temperature-dependent chemical toxicity to aquatic organisms, most existing studies focused on acute impacts of chemicals at high concentrations. This study aimed to investigate the interacting effect of temperature and copper exposure at environmentally relevant concentrations on survival and development in the marine copepod Tigriopus japonicus with a partial l

New supporting data to guide the use of evident toxicity in acute oral toxicity studies (OECD TG 420).

Currently there are three test guidelines (TG) for acute oral toxicity studies of substances or mixtures from the Organisation for Economic Co-operation and Development (OECD). TG 423 and TG 425 use lethality as an endpoint, while TG 420 replaces death with 'evident toxicity', defined as clear signs that exposure to a higher dose would result in death. However, the perceived subjectivity of 'evident toxicity' may be preventing wider use of TG 420. To address this, the UK National Centre for the Replacement, Refinement and Reduction of Animals in Research (NC3Rs) and the European Partnership fo

In vivo assessment of pathogens toxicity on Daphnia magna using fluorescent dye staining.

Daphnia has been widely used as an indicator species in aquatic biomonitoring for decades. Traditional toxicity assays based on lethality take a long time to assess, and the effect mode of contaminants is not clear. Because of the translucency of the Daphnia body and the application of fluorescent probes in cell staining, different intoxicated parts can be visualized. In this study, a double-staining method using two fluorescent dyes, Calcein AM (cell-permeant dye) and Propidium Iodide (cell-impermeant dye), was carried out on Daphnia magna exposed to six pathogens: Salmonella spp. (four strai

Toxicity assessment of metal mixtures to soil enzymes is influenced by metal dosing method.

Metals are present as mixtures in the environment, yet testing such complex mixture poses design and technical challenges. One possible solution is the use of fixed ratios, i.e. rays of increasing metal concentrations. But fixed ratios rays are compromised when soils dosed with metal salts are leached due to metal-soil selectivity rules. Two alternative metal forms, metal oxides and spinel minerals of quinary metal mixtures (Pb, Cu, Co, Ni, Zn), were evaluated for their toxicity to soil microorganisms measured by the activity of ammonia monooxygenases and acid-phosphatases in three soils. Is the test for irreversibility of tetanus toxoids still relevant?

Tetanus vaccines for human and veterinary use are based on toxoids resulting from a formaldehyde-mediated inactivation of tetanus neurotoxin (TeNT). Due to the high toxicity of TeNT, safety tests are mandatory for each batch of these toxoids. One of the tests addresses the irreversibility of inactivation: The toxoid is stored at 37 °C for 6 weeks and then subjected to in vivo toxicity testing. However, we found that TeNT solutions rapidly lose their activity at 37 °C. Accordingly, any active TeNT molecules arising in the toxoid due to reversion events may no longer be detectable after the 37 °

Assessing Cu impacts on freshwater diatoms: biochemical and metabolomic responses of Tabellaria flocculosa (Roth) Kützing.

Metals are a recognised threat to aquatic organisms but the impact of metals such as copper (Cu) on benthic freshwater diatoms is poorly understood, even if diatoms are commonly used as water quality indicators. Our study aimed to elucidate the cellular targets of Cu toxicity and the mechanisms cells resort to counteract toxicity and to increase tolerance to Cu. A concerted approach analysing the biochemical, physiological and metabolome alterations in diatom cells was conducted by exposing the freshwater diatom Tabellaria flocculosa to 0, 0.3, 6 and 10μgCu/L. Cu was already toxic to T. floccu

Lethal and sub-lethal effects of select macrocyclic lactones insecticides on forager worker honey bees under laboratory experimental conditions.

Selective insecticide application is one important strategy for more precisely targeting harmful insects while avoiding or mitigating collateral damage to beneficial insects like honey bees. Recently, macrocyclic lactone-class insecticides have been introduced into the market place as selective bio-insecticides for controlling many arthropod pests, but how to target this selectivity only to harmful insects has yet to be achieved. In this study, the authors investigated the acute toxicity of fourmacrocyclic lactone insecticides (commercialized as abamectin, emamectin benzoate, spinetoram, and s

Assessment of methods used to determine the safety of the topical insect repellent N,N-diethyl-m-toluamide (DEET).

N,N-diethyl-m-toluamide (DEET) has been registered for commercial use as an insect repellent for over five decades, and is used widely across the world. Concerns over the safety of DEET first emerged during the 1980s after reports of encephalopathy following DEET exposure, particularly in children. However, the role of DEET in either the illness or deaths was and remains purely speculative. In response to these cases a number of reviews and investigations of DEET safety were carried out. Here we examine the methods used and information available to determine the safety of DEET in humans. Anima

A toxicological and dermatological assessment of aryl alkyl alcohols when used as fragrance ingredients.

The aryl alkyl alcohol (AAA) fragrance ingredients are a diverse group of chemical structures with similar metabolic and toxicity profiles. The AAA fragrances demonstrate low acute and subchronic dermal and oral toxicity. No carcinogenicity in rats or mice was observed in 2-year chronic testing of benzyl alcohol or α-methylbenzyl alcohol; the latter did induce species and gender-specific renal adenomas in male rats at the high dose. There was no to little genotoxicity, mutagenicity, or clastogenicity in the mutagenic in vitro bacterial assays, and in vitro mammalian cell assays. All in vivo mi

Imaging methods for determining uptake and toxicity of carbon nanotubes in vitro and in vivo.

Demand for carbon nanotubes (CNTs) is increasing rapidly in electrical, mechanical, and health and medical applications due to their thermal, electrical conductive and other properties. The continued commercial up-scaling of CNT production and application needs to be accompanied by an understanding of the occupational health, public safety and environmental implications of these materials. An increasing volume of literature on the toxicity of CNTs is being published; however, the results of these studies are frequently inconclusive. Due to the enormous number of permutations of nanoparticle sh

Metallothionein in the freshwater gastropod Melanopsis dufouri chronically exposed to cadmium: a methodological approach.

Previous studies have demonstrated that the use of differential pulse polarography (DPP) for metallothionein (MT) determination in marine gastropod tissues, particularly the digestive gland, requires taking into account the presence of heat-stable high molecular weight compounds that exhibit polarographic signal. In the present paper, similar compounds were identified in tissues from the freshwater snail Melanopsis dufouri which also interfere with MT determination by DPP and, due to their silver binding capacity, also interfere in the silver assay for MT quantification. Ultrafiltration seems

Toxicity of haloacetic acids to freshwater algae.

Haloacetic acids (HAA), such as trichloroacetic acid (TCA), are commonly occurring by-products from disinfection and bleaching processes using sodium hypochlorite. Currently, the lowest no observed effect concentration (NOEC) for TCA is reported to be 8.7microgL(-1), which was derived from a toxicity study conducted in 1981 on Chlorella pyrenoidosa. The purity of the test material was not documented and it is unknown if other halogenated impurities or co-formulants were present. However, this NOEC is used to derive a predicted no effect concentration, which is used in various regulatory risk a

Final report on the safety assessment of Trichloroethane.

Trichloroethane functions in cosmetics as a solvent. Although Trichloroethane has been reported to the Food and Drug Administration (FDA) to be used in cosmetic products, an industry survey found that it is not in current use in the cosmetic industry. Trichloroethane is considered a Class I ozone-depleting substance by the Environmental Protection Agency (EPA) and its use is prohibited in the United States, unless considered essential. The FDA has stated that Trichloroethane's use in cosmetics is considered nonessential. Trichloroethane is detected by gas chromatography, gas chromatography-mas
