CONTEXT: The tumor microenvironment (TME) includes diverse cellular components such as mesenchymal stem cells (MSCs) and immune cells, among others. MSC have been isolated from different tumors and they favor tumor cell growth; however, their role in pituitary tumors (PTs) remains unknown.
OBJECTIVE: Herein we report the presence of MSCs in 2 adrenocorticotropin (ACTH)-secreting PTs causing Cushing disease (MCU), 2 nonfunctioning adenomas of gonadotrope differentiation (MNF), and 2 nontumoral pituitary glands (MS).
METHODS: We have analyzed the transcriptomic profiles by RNA sequencing and c…

The study of the cellular and molecular microenvironment in B cell lymphoma, especially diffuse large B cell lymphoma (DLBCL), has led to prognostic and therapeutic algorithms that may improve patient outcomes. Emerging gene signature panels provide a granular understanding of DLBCL based on the immune tumor microenvironment (iTME). In addition, some gene signatures identify lymphomas that are more responsive to immune-based treatment, indicating that the iTME has a biological signature that could affect outcomes when targeted. In this issue of the JCI, Apollonio et al. report on fibroblastic …

Nerve terminals within the tumor microenvironment as potential pain-mitigating targets for local infiltration analgesia is relatively less explored. In this study, we examine the role of key analgesics administered as local infiltration analgesia in a model of cancer-induced bone pain (CIBP). CIBP was induced by administration of allogenic MRMT1 breast cancer cells in the proximal tibia of rats, and tumor mass characterized using radiogram, micro-CT, and histological analysis. In vitro responsiveness to key analgesics δ-opioid receptor agonist (DOPr), Ca2+ channel and TRPV1 antagonists was ass…

Immunotherapy, particularly immune checkpoint inhibitors, have become widely used in various settings across many different cancer types in recent years. Whilst patients are often treated on the basis of the primary cancer type and clinical stage, recent studies have highlighted disparity in response to immune checkpoint inhibitors at different sites of metastasis, and their impact on overall response and survival. Studies exploring the tumor immune microenvironment at different organ sites have provided insights into the immune-related mechanisms behind organ-specific patterns of response to

Cancer is a complex disease that includes the reprogramming of metabolic pathways by malignant proliferating cells, including those affecting the tumor microenvironment (TME). The "TME concept" was introduced in recognition of the roles played by factors other than tumor cells in cancer progression. In response to the hypoxic or semi-hypoxic characteristic of the TME, cancer cells generate a large amount of lactate via the metabolism of glucose and glutamine. Export of this newly generated lactate by the tumor cells together with H+ prevents intracellular acidification but acidifies the TME. I…

BACKGROUND: The role of flow cytometry in diagnosis and management of Hodgkin lymphoma (HL) remains limited. As knowledge emerges of the tumor microenvironment in this disease, various methods are being evaluated in its study. This study examines the microenvironment using flow cytometry to assess differences between subtypes and clinicopathologic correlates.
PROCEDURE: A retrospective cross-sectional study was performed analyzing the tumor immunophenotype, by flow cytometry, for 31 children with classical HL. Correlation was made with patient information, including outcome.
RESULTS: The mak…

BACKGROUND: Malignant peritoneal mesothelioma (MPM) is a rare cancer arising from mesothelial cells lining the peritoneal surface. Little is known about the tumor microenvironment in regulating MPM oncogenesis. The current study defined the chemokine/cytokine expression profile and inflammatory responses within the MPM microenvironment.
METHODS: Levels of 10 cytokines (Fractalkine, IFNγ, IL-6, IL-8, IP-10, MCP-1, MIP-1α, MIP-1β, TNFα, VEGF) in matched ascites and sera from 15 MPM patients were measured using Milliplex immunoassays. Sera from six normal control sera were included. Statistical …

Ovarian epithelial carcinomas are heterogeneous malignancies exhibiting great diversity in histological phenotypes as well as genetic and epigenetic aberrations. A general early event in tumorigenesis is regional dissemination into the peritoneal cavity. Initial spread to the peritoneum is made possible by cooperative signaling between a wide array of molecules constituting the tissue microenvironment in the coelomic epithelium. Changes in the activity of key microenvironmental components not constitutively expressed in normal tissue, including several disclosed adhesion molecules, growth fact

Canine cutaneous mast cell tumors (MCTs) exhibit heterogeneous biological behaviors, with increasing evidence implicating the tumor immune microenvironment (TIME) in disease progression. This study evaluated biopsy-confirmed cases of canine MCTs to investigate associations between tumor grade and host factors, and to characterize grade-related differences in immune marker expression. Tumors were histologically graded by the Kiupel system and analyzed using immunohistochemistry for CD4, CD8, CD25, CD57, MHC I, MHC II, PD-1, PD-L1, and mast cell tryptase. Quantitative assessments included positi

Efferocytosis plays a critical role in maintaining tissue homeostasis and controlling inflammation. However, in the tumor microenvironment, this process is hijacked to promote immune suppression. Treatments like radiotherapy and chemotherapy lead to widespread tumor cell death, which triggers efferocytosis and drives macrophages into an immunosuppressive state, allowing the tumor to evade the immune system and resist treatment. Nonetheless, the way efferocytic signals alter macrophage function and promote protumor effects during the recognition, engulfment, and digestion of apoptotic cells rem…

Thyroid cancer (TC) is the most common malignant tumor of the endocrine system. Although most cases have a favorable prognosis, some patients may be resistant to treatment or exhibit aggressive behavior. The tumor microenvironment (TME) network, composed of stromal cells, immune cells, vascular cells, and cancer cells, has become a key factor in the development of TC. The TME affects the biological behavior of TC through different immune states. TC cells can suppress antitumor immune response by promoting an immunosuppressive microenvironment, such as through the recruitment of tumor-associate…

Chimeric antigen receptor (CAR)-T cell therapy has achieved remarkable success in hematological malignancies but remains largely ineffective in solid tumors. Accumulating evidence indicates that this limitation reflects not only suboptimal antigen recognition but also the inability of therapeutic cells to reach and function within the suppressive tumor microenvironment. This review synthesizes recent advances in CAR-T cell bioengineering aimed at overcoming physical barriers, immunosuppressive signaling, metabolic stress, and cellular exhaustion that characterize solid tumors, which are a lead…

Protein lactylation, a novel post-translational modification (lysine lactylation, Kla) driven by the oncometabolite lactate, has emerged as a critical epigenetic mechanism that directly links cellular metabolic state to gene regulation. Within the tumor microenvironment (TME), lactate accumulation resulting from the Warburg effect provides abundant substrate for lactylation, positioning this modification as a central hub in cancer biology. This review systematically elucidates the dual role of lactylation in driving tumor progression. Intrinsically, lactylation promotes tumor cell malignancy b…

BACKGROUND: Immunotherapy targeting immune checkpoint proteins (ICPs) has transformed cancer care, yet current treatments focus on a narrow set of inhibitory ICPs and benefit only a subset of patients. The co-stimulatory pair OX40-OX40L, implicated in inflammation and autoimmunity, also plays roles in cancer immunity. We previously showed that high OX40L mRNA expression in melanoma correlates with favorable prognosis and improved responses to PD-1 blockade. However, the protein-level expression and functions of OX40L in melanoma remain poorly defined.
METHODS: Formalin-fixed paraffin-embedded

Nitric oxide (NO) is a widespread signaling molecule which has far-reaching effects in cellular physiology and pathophysiology, especially in cancer biology. Its actions are concentration-dependent where low concentrations facilitate tumor development and high concentrations cause cytotoxicity. NO alters several cancer hallmarks, affecting the initiation, progression, immune evasion, and therapeutic responses of tumors via cGMP-dependent and -independent pathways. Various cell types in the tumor microenvironment (TME) produce NO in a concentration gradient creating a strong concentration gradi…

Tumors co-opt normal wound-healing programs to shape their immune microenvironment, yet how distinct immune states arise and influence therapy remains unclear. Here, we synthesize emerging evidence that tumors become locked in either proinflammatory or pro-resolution phases, defined by neutrophil- or macrophage-dominated ecosystems. These states not only remodel local tissues but also systemically reprogram hematopoiesis, creating self-reinforcing immune circuits that drive progression, metastasis, and treatment resistance. We highlight recent advances linking epithelial-mesenchymal plasticity

Hypoxia and acidity represent two distinct properties of tumor microenvironments (TMEs). Tumor acidity was initially proposed to be a "by-product" of hypoxia, which results from abnormal vasculature and excessive oxygen demands. It is now recognized that acidity has its unique function in TMEs, as it can exacerbate abnormal cellular energy metabolism, inhibit cellular carcinogen metabolism, impede DNA damage repair mechanisms, and suppress immune cell activation, all of which play crucial roles during carcinogenesis. Acidic TMEs are also major contributors to the invasion and metastasis of tum…
