Vascular calcification is a key pathological process contributing to cardiovascular mortality in chronic kidney disease (CKD). While HDAC2 is known to exert protective effects, its downstream mediators remain elusive. Here, we identify DLX2 as a critical pro-calcific driver and a target of HDAC2 in vascular smooth muscle cells (VSMCs). Transcriptomic analysis following HDAC2 overexpression revealed DLX2 as a key downregulated gene. Functional studies demonstrated that knockdown of DLX2 attenuated β-glycerophosphate (β-GP)-induced VSMC calcification, suppressed osteogenic transdifferentiation, …

Patients with chronic kidney disease (CKD) are at an increased risk for vascular calcification (VC), a strong predictor of cardiovascular events. Rodent models with elevated dietary levels of adenine and phosphate are commonly used to study CKD-induced VC; however, dietary composition, regimen duration, and reported outcomes vary widely across studies. Researchers customize high-adenine, high-phosphate (AdeHP) diets by adjusting phosphate levels and selecting specific phosphate salts as the phosphate source. In this observational study, we found that the selection and amount of phosphate salt …

Chronic kidney disease (CKD), a condition affecting over 850 million people worldwide, is a major global health issue. CKD leads to multimorbidities and disease complexity, expediting the aging process and increasing mortality rates-a phenomenon sometimes referred to as "renal senescence." The leading cause of death in patients with CKD is cardiovascular disease (CVD), accounting for one-third to one-half of all deaths, in stark contrast to cancer, which is the primary cause of death in the general population. While previous studies on kidney disease have focused extensively on urinary extrace

INTRODUCTION: Vascular calcification is prevalent in chronic kidney disease (CKD), but existing medical treatments fail to achieve satisfactory therapeutic effects. Vascular calcification is now recognized as an active multifactorial process involving diverse mechanisms. Alpha-ketoglutarate (AKG), an intermediate in tricarboxylic acid cycle, has been demonstrated to extend lifespan and ameliorate age-related osteoporosis. However, whether AKG inhibits vascular calcification remains unknown.
METHODS: Here, mineral deposition was studied with AKG treatment in rodent and human vascular smooth mu…

Colchicine (Col) is a traditional herbal medicine derived from the plant Colchicum autumnale. With the property of anti-inflammation, Col has demonstrated certain therapeutic effects in cardiovascular diseases. Vascular calcification is positively related to the morbidity and mortality of cardiovascular diseases. However, the specific cardiovascular conditions for which Col is effective remain unclear, particularly its impact on vascular calcification. In this study, we used high phosphate to induce calcium deposition in vascular smooth muscle cells and Vitamin D3 plus nicotine or 5/6 nephrect…

Total pharyngo-laryngo-esophagectomy (TPLE) with free jejunal transplantation (FJT) is the standard reconstructive procedure for hypopharyngeal cancer, typically utilizing the superior thyroid artery as the recipient vessel. However, patient-specific anatomical variations and comorbidities can significantly complicate this surgery. We present a unique case of a 68-year-old male with hypopharyngeal cancer who exhibited multiple challenges, including short stature (126 cm), low weight (35 kg), cervical spondylosis, and a history of vertebroplasty, highlighting the complexities inherent in such r

BACKGROUND: Systemic sclerosis (SSc) patients face an elevated risk of cardiovascular disease (CVD), even when classic cardiovascular risk factors are considered. Plasma dephosphorylated-uncarboxylated Matrix Gla-protein (dp-ucMGP), an inactive form of MGP, is associated with increased CVD risk. Smooth muscle cells, implicated in SSc's development, are the primary dp-ucMGP producers. This study assessed dp-ucMGP levels and initial CVD events in early-diagnosed SSc patients, investigating its potential as a CVD and all-cause mortality predictor over time.
METHODS: In a cohort of 87 SSc patient

Human cytomegalovirus (HCMV) is a widespread pathogen that causes lifelong latent infection in the majority of the world population. HCMV is associated with increased incidence and severity of many cardiovascular diseases including myocarditis, atherosclerosis, and transplant vasculopathy. Due to the species-restricted nature of cytomegalovirus infection, murine cytomegalovirus (MCMV) is a useful model that recapitulates many of the features of HCMV infection of the cardiovascular system. While in vivo MCMV studies are able to answer many questions regarding pathogenesis of infection, in vitro

OBJECTIVE: Anastomotic leakage (AL) is a catastrophic surgical complication affecting the prognosis of patients after colorectal surgery. We aimed to determine the value of the arterial calcification (AC) score in predicting AL.
METHODS: Medline and Embase were searched through November 2019. The odds ratio (OR) and 95% confidence interval (CI) were used to estimate the association between AC and AL after colorectal surgery. The fixed-effects model or random-effects model was adopted for data pooling. Subgroup analyses were conducted to assess the effect of different aortoiliac trajectories.

Vascular calcification (VC) is highly prevalent in patients with chronic kidney disease (CKD) and contributes to their high rate of cardiovascular mortality. Indoxyl sulfate (IS) is a representative protein-bound uremic toxin in CKD patients, which has been recognized as a major risk factor for VC. Recent studies have demonstrated that nuclear factor-kappa B (NK-κB) is highly activated in the chronic inflammation conditions of CKD patients and participated in the pathogenesis of VC. However, whether NK-κB is involved in the progression of IS-induced VC remains without elucidation. Here, we sho…

Although dietary phosphate restriction is important for treating hyperphosphatemia in patients with chronic kidney disease, it remains unclear whether a low-protein diet (LPD), which contains low phosphate, has beneficial effects on malnutrition, inflammation, and vascular calcification. The effects of LPD on inflammation, malnutrition, and vascular calcification were therefore assessed in rats. Rats were fed a normal diet or diets containing 0.3% adenine and low/normal protein and low/high phosphate. After 6 wk, serum and urinary biochemical parameters, systemic inflammation, and vascular cal…
