Suppression of niacin-induced vasodilation with an antagonist to prostaglandin D2 receptor subtype 1.

Niacin (nicotinic acid) reduces cardiovascular events in patients with dyslipidemia. However, symptoms associated with niacin-induced vasodilation (e.g., flushing) have limited its use. Laropiprant is a selective antagonist of the prostaglandin D(2) receptor subtype 1 (DP1), which may mediate niacin-induced vasodilation. The aim of this proof-of-concept study was to evaluate the effects of laropiprant (vs placebo) on niacin-induced cutaneous vasodilation. Coadministration of laropiprant 30, 100, and 300 mg with extended-release (ER) niacin significantly lowered flushing symptom scores (by appr…

Painful stimulation of the temple induces nausea, headache and extracranial vasodilation in migraine sufferers.

To determine whether painful stimulation of the temple would induce nausea, ice was applied to the temple for 30 s, three times at 4-min int … esumably due to anticipatory anxiety. In contrast, the ice application did not provoke extracranial vasodilation in either group after optokinetic stimulation. The findings show that susceptibility to nausea and stress-induced extracranial vascular hyper-reactivity are associated with the migraine predisposition. They also suggest that head pain might intensif …

Experimental model for comparative evaluation of pharmacologically induced vasodilation of arterial wall mechanical properties.

Arterial wall compliance (C) and distensibility coefficient (DC) are key factors of pathologic physiology, especially in arteries less than 2 mm in diameter. The aim of this study was to design an experimental model allowing comparative measurement of C and DC during pharmacologically induced vasodilation on small-diameter arteries. Both femoral arteries were exposed in eight New Zealand White rabbits. Diameter (d) and systolic/diastolic diameter changes (deltad) were measured simultaneously, and C and DC were calculated before and after topical application of 1 mL of 4% papaverine on the righ…

Muscarinic cholinergic and beta-adrenergic contribution to hindquarters vasodilation and cardiac responses to cocaine.

Cocaine produces a pressor response associated with an initial hindquarters vasoconstriction followed by a prolonged vasodilation in conscious rats. Propranolol pretreatment prevented the vasodilation and enhanced the pressor response, whereas atropine methylbromide pretreatment reduced the increase in systemic vascular resistance. We studied the role of selective muscarinic and beta-adrenoceptor antagonists on responses to cocaine in rats with an increase in systemic vascular resistance to cocaine (vascular responders). Arterial blood pressure and ascending aortic and distal descending aortic…

The in vitro reversal of histamine-induced vasodilation in the human internal mammary artery.

UNLABELLED: Anaphylactic shock therapy includes the use of catecholamines but they may not always be effective. Because vasodilation during anaphylaxis is a result of the endothelial release of multiple mediators, we investigated the effects of epinephrine, vasopressin, and inhibitors of nitric oxide and prostanoid pathways on histamine-induced relaxation in human internal mammary artery. The vessel segments were obtained intraoperatively and were suspended in organ chambers to record isometric tension. Norepinephrine (10(-6) M) was used to precontract the rings followed by histamine (10(-6.5)…

Hemodynamic effects of cannabinoids: coronary and cerebral vasodilation mediated by cannabinoid CB(1) receptors.

Activation of peripheral cannabinoid CB(1) receptors elicits hypotension. Using the radioactive microsphere technique, we examined the effec … vasodilators in the mesentery. We conclude that cannabinoids elicit profound coronary and cerebral vasodilation in vivo by direct activation of vascular cannabinoid CB(1) receptors, rather than via autoregulation, a decrease in sympathetic tone or, in the case of anandamide, the action of a non-cannabinoid metabolite. Differences between the hemodynamic profi …

Changes in coronary endothelial cell Ca2+ concentration during shear stress- and agonist-induced vasodilation.

Increases in intraluminal shear stress are thought to cause vasodilation of coronary arterioles by activation of Ca2+-dependent endothelial nitric oxide synthase followed by release of nitric oxide. We tested the hypothesis that endothelium-dependent vasodilation of isolated coronary arterioles to shear stress and agonists is necessarily preceded by an increase in endothelial cell Ca2+ concentration ([Ca2+]i). After selective loading of endothelium in isolated rabbit coronary arterioles with fura 2, simultaneous changes in diameter and [Ca2+]i were recorded. Vasodilations recorded in response …

Isoflurane and sevoflurane induce vasodilation of cerebral vessels via ATP-sensitive K+ channel activation.

BACKGROUND: Activation of adenosine triphosphate-sensitive K+ channels causes cerebral vasodilation. To assess their contribution to volatile anesthetic-induced cerebral vasodilation, the effects of glibenclamide, an adenosine triphosphate-sensitive K+ channel blocker, on the cerebral vasodilation induced by isoflurane and sevoflurane were studied. METHODS: Pentobarbital-anesthetized dogs (n = 24) assigned to one of two groups were prepared for measurement of pial vessel diameter using a cranial window preparation. Each dog received three minimum alveolar concentrations (MAC; 0.5, 1, and 1.5 …

Isolated defect of adenosine-mediated coronary vasodilation: functional evidence for a new microangiopathic entity.

OBJECTIVES: The present study describes an isolated defect of the coronary vasodilation in response to adenosine in five patients examined for clinically suspected coronary microangiopathy. BACKGROUND: Coronary microangiopathies can be defined functionally as dysregulation of the microcirculatory vasomotion. METHODS: The five patients were compared with 24 control subjects. Coronary flow velocity was measured with an intracoronary Doppler guide wire (0.018 in. [0.046 cm], 12 MHz) at rest and during intracoronary administration of adenosine (80 micrograms/min and 160 micrograms/min over 3 min…

Cerebral vasodilation capacity: acute intracranial hypertension and supra- and infra-tentorial artery velocity recording.

This experiment is the first to compare cerebral vasomotor reactivity in the supra- and infra-tentorial regions in baseline conditions and during progressive acute intracranial hypertension. The increase in intracranial pressure was performed using liquid pressure transmission in two groups of 16 rabbits by elevating a saline infusion bottle connected to the subdural space. Cerebral microvessel dilation capacity was studied using acetazolamide arterial infusion during three stages of 20 min: at baseline conditions, with an intracranial pressure value equal to half the diastolic arterial pressu

Perivascular adipose tissue from female rats fed a high-fat diet impaired mesenteric artery vasodilation.

High-fat diet (HFD) is a cardiovascular risk factor that may disproportionately affect women compared to men. HFD alters perivascular adipose tissue (PVAT), which surrounds arteries and regulates vascular function. This study investigated the sex-specific effects of HFD on PVAT-mediated vascular dysfunction. Male and female Sprague-Dawley rats were fed a normal chow diet (NCD) or HFD for 16 weeks. Thoracic aortic PVAT was isolated, and PVAT adipokine expression and inflammatory markers were measured. We then assessed PVAT-conditioned media effects on vasodilation, endothelial oxidative stress,…

Short-term estradiol administration does not restore endothelin-B receptor-mediated vasodilation in postmenopausal women.

The endothelin-B receptor (ETBR) mediates vasodilation in young women, an effect that is absent in postmenopausal women. We have previously demonstrated that ETBR-mediated vasodilation is regulated by estradiol (E2) in young women; however, the impact of E2 on ETBR function in postmenopausal women remains unknown. Accordingly, the objective of this study was to test the hypothesis that E2 exposure restores ETBR-mediated dilation in postmenopausal women. Ten healthy postmenopausal women (55 ± 2 yr of age, 5 ± 3 years since menopause) completed the study. E2 was administered by transdermal patch…

β-Adrenergic receptors mediate sex differences in vasodilation but not sympathetic-mediated vasoconstriction during hypoxia.

β-Adrenergic receptors (ARs) mediate a portion of hypoxic vasodilation and blunt sympathetic vasoconstriction at rest. Vascular β-AR sensitivity may be greater in females relative to males; however, their sex-specific role during hypoxia has yet to be examined. We hypothesized β-AR blockade would blunt hypoxic vasodilation and augment sympathetic vasoconstriction during hypoxia, with effects greater in females relative to males. Ten female (26 ± 8 yr, 23 ± 3 kg/m2) and 13 male (28 ± 7 yr, 25 ± 2 kg/m2) adults completed two study visits randomized and blinded to oral placebo or propranolol (β-A…
