Comparative genomics of parasitoid lifestyle as exemplified by Mermithidae and Nematomorpha.

Mermithidae and Nematomorpha are parasitoids united by the commonalities in their lifestyle. Immature stages infect arthropod hosts, can manipulate their host to induce water-seeking behavior, and both have a final free-living non-feeding reproducing adult stage, often killing their host upon emergence. Some species are of great economic importance, being evaluated as biological control agents against mosquito vectors responsible for diseases like malaria, but with scarce genomic resources currently available. This study aimed to investigate the genetic features of the genomes of both Mermithi

A Cross-Sectional Study of Rift Valley Fever Exposure in Humans and Livestock in Southwestern Uganda Using a One Health Approach: Evidence of Elevated Seroprevalence Outside Recognized Outbreak Periods.

Rift Valley fever (RVF) transmission has intensified in southwestern Uganda since 2016. To quantify human and livestock exposure and associated risks, we conducted a cross-sectional serosurvey in Isingiro, Kabale and Rubanda districts between October and November 2023. A total of 766 humans and 2383 livestock were sampled and tested for RVF antibodies using ELISA, with structured questionnaires capturing demographic, behavioral and environmental data. Human seroprevalence was 11.5% (88/766), varying by district (13.8% Isingiro, 11.8% Rubanda, 6.8% Kabale; p = 0.04). Independent predictors from

Global Assessment of current and future chikungunya virus transmission risk using optimized maxent modeling.

Chikungunya virus (CHIKV) primarily spreads through an urban transmission cycle involving humans and two principal mosquito vectors, Ae. aegypti and Ae. albopictus, especially in densely populated areas, posing significant public health challenges as climate change alters vector habitats. This study applied an optimized Maximum Entropy (MaxEnt) model incorporating CHIKV and primary vector occurrence data (2005-2020) and filtered environmental predictors within a hierarchical framework to project CHIKV transmission risk under current and 96 future scenarios. Under current climate conditions, ar

Impact of environmental factors on the bionomics of Anopheles mosquito vectors of zoonotic malaria: A narrative review.

The increasing burden of non-human primates (NHP) malaria, driven primarily by Plasmodium knowlesi, poses a growing public health threat in many countries across Southeast Asia. Compounding this challenge, the emergence of other NHP Plasmodium species infecting humans, including P. cynomolgi, P. inui, and P. fieldi, introduces additional complexity to malaria elimination efforts, particularly in countries like Malaysia, Thailand, and Vietnam. A complex interplay among human populations, vector dynamics, and environmental factors influences the transmission and prevalence of this disease. This

Dengue and chikungunya viruses among febrile travelers entering Iran (2015-2021): Evidence of multiple introductions from different countries.

OBJECTIVES: Recent outbreaks of dengue, chikungunya, and Zika have underscored the growing threat to public health. This study retrospectively describes the presence of these viruses among febrile travelers entering Iran between 2015 and 2021. Although Iran had no local transmission until 2024, several factors increased the risk of outbreaks, including the presence of mosquito vectors, favorable climate, and inadequate traveler control measures. Climate change and increased humidity have further increased this risk. METHODS: A multiplex real-time reverse transcription-polymerase chain reactio

A Multiplex Serological Assay for the Detection of Antibody Responses to Arboviruses.

Arthropod borne viruses (arboviruses) are a group of mosquito-borne pathogens that are globally distributed. Since the 2016 outbreak of Zika virus (ZIKV) in the Americas that brought these infections to public awareness, arboviruses continue to impose a considerable public health burden. Their emergence in previously non-endemic regions is a cause of public health concern. The surveillance and control of arboviral infections is a current priority for the World Health Organization (WHO), which launched a global strategic plan to fight arboviral diseases in 2024. The multiplex® technology has re

Fluid management in children with severe dengue: a narrative review.

Dengue is a mosquito-borne arboviral infection of increasing public health importance. Globally, children account for a significant proportion of infections. No pathogen-specific treatment currently exists, and the current approach to reducing disease burden is focused on preventative strategies such as vector control, epidemiological interventions, and vaccination in selected populations. Once infected, the mainstay of treatment is supportive, of which appropriate fluid management is a cornerstone. The timely provision of fluid boluses has historically been central to the management of septic

Green Nano-Biotechnology: A New Sustainable Paradigm to Control Dengue Infection.

Dengue is a growing mosquito-borne viral disease prevalent in 128 countries, while 3.9 billion people are at high risk of acquiring the infection. With no specific treatment available, the only way to mitigate the risk of dengue infection is through controlling of vector, i.e., Aedes aegypti. Nanotechnology-based prevention strategies like biopesticides with nanoformulation are now getting popular for preventing dengue fever. Metal nanoparticles (NPs) synthesized by an eco-friendly process, through extracts of medicinal plants have indicated potential anti-dengue applications. Green synthesis

A Rat Model of Prenatal Zika Virus Infection and Associated Long-Term Outcomes.

Zika virus (ZIKV) is a mosquito-borne flavivirus that became widely recognized due to the epidemic in Brazil in 2015. Since then, there has been nearly a 20-fold increase in the incidence of microcephaly and birth defects seen among women giving birth in Brazil, leading the Centers for Disease Control and Prevention (CDC) to officially declare a causal link between prenatal ZIKV infection and the serious brain abnormalities seen in affected infants. Here, we used a unique rat model of prenatal ZIKV infection to study three possible long-term outcomes of congenital ZIKV infection: (1) behavior,

Potential Anti-Malarial Agents from Endophytic Fungi: A Review.

Malaria is one of the major infectious diseases and foremost cause of mortality and morbidity in many subtropical and tropical regions. In the last years, the situation has become worst in many ways, due to increase in the parasites resistance to various available antimalarial agents. Furthermore, malaria`s control is beginning to be more sophisticated by the parallel spread of mosquito vector`s resistance to the available insecticides. Recently, there is a wide consensus to seek for target specific, safe, affordable, and effective new antimalarial agents, which can compete with synthetic ones

Mapping the potential distributions of etiological agent, vectors, and reservoirs of Japanese Encephalitis in Asia and Australia.

Japanese encephalitis virus (JEV) is a substantial cause of viral encephalitis, morbidity, and mortality in South-East Asia and the Western Pacific. World Health Organization recognized Japanese Encephalitis (JE) as a public health priority in demands to initiate active vaccination programs. Recently, the geographic distribution of JEV has apparently expanded into other areas in the Pacific islands and northern Australia; however, major gaps exist in knowledge in regard to its current distribution. Here, we mapped the potential distribution of mosquito vectors of JEV (Culex tritaeniorhynchus,

Discovery of novel dengue virus entry inhibitors via a structure-based approach.

Dengue is a mosquito-borne virus that has become a major public health concern worldwide in recent years. However, the current treatment for dengue disease is only supportive therapy, and no specific antivirals are available to control the infections. Therefore, the need for safe and effective antiviral drugs against this virus is of utmost importance. Entry of the dengue virus (DENV) into a host cell is mediated by its major envelope protein, E. The crystal structure of the E protein reveals a hydrophobic pocket occupied by the detergent n-octyl-β-d-glucoside (β-OG) lying at a hinge region be

Spatial quantification of the world population potentially exposed to Zika virus.

BACKGROUND: Zika virus is an emerging Flaviviridae virus, which has spread rapidly in the last few years. It has raised concern because it has been associated with fetus microcephaly when pregnant women are infected. The main vector is the mosquito Aedes aegypti , distributed in tropical areas. METHODS: Niche modelling techniques were used to estimate the potential distribution area of A. aegypti. This was overlapped with human population density, determining areas of potential transmission risk worldwide. Afterwards, we quantified the population at risk according to risk level. RESULTS: The

Chemoprophylaxis with sporozoite immunization in P. knowlesi rhesus monkeys confers protection and elicits sporozoite-specific memory T cells in the liver.

Whole malaria sporozoite vaccine regimens are promising new strategies, and some candidates have demonstrated high rates of durable clinical protection associated with memory T cell responses. Little is known about the anatomical distribution of memory T cells following whole sporozoite vaccines, and immunization of nonhuman primates can be used as a relevant model for humans. We conducted a chemoprophylaxis with sporozoite (CPS) immunization in P. knowlesi rhesus monkeys and challenged via mosquito bites. Half of CPS immunized animals developed complete protection, with a marked delay in para

Trypsin inhibitor from Leucaena leucocephala seeds delays and disrupts the development of Aedes aegypti, a multiple-disease vector.

BACKGROUND: Nowadays, the Aedes aegypti mosquito represents a serious public health issue in view of the large outbreaks of the arboviral diseases zika, dengue, chikungunya and yellow fever. This holometabolous insect has midgut digestive enzymes that are trypsin- and chymotrypsin-like proteins. Protease inhibitors are able to bind to proteolytic enzymes and promote a blockage in digestion and nutrition, leading to death. Thus, we investigated the effect of trypsin inhibitor of Leucaena leucocephala (LTI) seeds on egg hatching, larval development and digestive midgut proteases. RESULTS: LTI w
