Advantages of patient-specific cutting guides with disposable instrumentation in total knee arthroplasty: a case control study.

BACKGROUND: Total knee arthroplasty (TKA) is most frequently planned using conventional two-dimensional weight-bearing lower limb radiographs and is performed with conventional femoral and tibial cutting guides. Questions have been raised about the accuracy of conventional TKA instrumentation and planning for an anatomically standard or complex joint. Use of computed tomography (CT)-based three-dimensional (3D) templating and patient-specific cutting guides printed in 3D has shown improved postoperative lower limb alignment parameters. This case-control study compared costs and operative times

Novel strategy for multi-material 3D bioprinting of human stem cell based corneal stroma with heterogenous design.

Three-dimensional (3D) bioprinting offers an automated, customizable solution to manufacture highly detailed 3D tissue constructs and holds great promise for regenerative medicine to solve the severe global shortage of donor tissues and organs. However, uni-material 3D bioprinting is not sufficient for manufacturing heterogenous 3D constructs with native-like microstructures and thus, innovative multi-material solutions are required. Here, we developed a novel multi-material 3D bioprinting strategy for bioprinting human corneal stroma. The human cornea is the transparent outer layer of your ey

Integrated surgical management of forequarter lateral implosion injury: Technical considerations and early outcomes.

Concomitant ipsilateral fractures of the clavicle, scapula, and ribs, termed forequarter lateral implosion injury, represent a severe but underrecognized injury pattern resulting from high-energy lateral shoulder trauma. While the surgical indications for isolated chest wall and shoulder girdle injuries are well described, guidance on the integrated management of this combined injury complex remains limited. We describe a reproducible multidisciplinary approach for the concurrent surgical fixation of clavicle, scapula, and rib fractures, illustrated through two cases of forequarter lateral imp

Bioprinted human liver tissue model for evaluating Microcystin-LR-induced hepatotoxicity.

Microcystin-LR (MC-LR), a potent cyanobacterial hepatotoxin, poses increasing risks to environmental and human health, yet its toxicological effects in physiologically relevant human liver systems remain insufficiently understood. To address this gap, we developed a three-dimensional (3D) bioprinted human liver tissue model incorporating hepatocytes and hepatic stellate cells within a GelMA hydrogel. The printed constructs exhibited high structural fidelity, >85% cell viability, and progressive functional maturation over seven days, as demonstrated by increased ALB secretion, transporter activ

Gravitational forces and matrix stiffness modulate the invasiveness of breast cancer cells in bioprinted spheroids.

The progression of breast cancer is influenced by the stiffness of the extracellular matrix (ECM), which becomes stiffer as cancer advances due to increased collagen IV and laminin secretion by cancer-associated fibroblasts. Intriguingly, breast cancer cells cultivated in two-dimensions exhibit a less aggressive behavior when exposed to weightlessness, or microgravity conditions. This study aims to elucidate the interplay between matrix stiffness and microgravity on breast cancer progression. For this purpose, three-dimensional spheroids of breast cancer cell lines (MCF-7 and MDA-MB-231) were

Application of stem cell-derived exosomes in bone and joint diseases: Recent advances enabled by diverse carrier technologies.

Bone and joint diseases, such as osteoarthritis, exhibit significant pathological complexity. Current treatment modalities possess notable limitations, driving the development of cell-free regenerative treatment strategies centered on stem cell-derived exosomes, particularly mesenchymal stem cell-derived exosomes. Despite the promise of mesenchymal stem cell-derived exosomes, several challenges impede their clinical translation. These include rapid in vivo clearance of exosomes, insufficient targeting specificity, and the difficulty of dynamically regulating the pathological microenvironment w

Validation of an in vitro testing platform for intra-arterial thrombolytics on human thrombectomy retrieved clots.

BACKGROUND: Endovascular thrombectomy (EVT) is the standard treatment for acute ischemic stroke (AIS) due to large vessel occlusion. However, many patients fail to achieve good outcomes, especially without first pass reperfusion. Intra-arterial thrombolytics such as recombinant tissue plasminogen activator (rtPA) and tirofiban have shown potential as adjunctive therapies. The aim of this study was to compare the in vitro effects of rtPA, tirofiban, and their sequential combination on EVT retrieved clots. METHODS: Human clot obtained during EVT was tested in an in vitro model of cerebral circu
