Brain acetylcholinesterase activity controls systemic cytokine levels through the cholinergic anti-inflammatory pathway.

The excessive release of cytokines by the immune system contributes importantly to the pathogenesis of inflammatory diseases. Recent advances in understanding the biology of cytokine toxicity led to the discovery of the "cholinergic anti-inflammatory pathway," defined as neural signals transmitted via the vagus nerve that inhibit cytokine release through a mechanism that requires the alpha7 subunit-containing nicotinic acetylcholine receptor (alpha7nAChR). Vagus nerve regulation of peripheral functions is controlled by brain nuclei and neural networks, but despite considerable importance, litt

EDNRB/EDN3 and Hirschsprung disease type II.

The study of vertebrate pigmentary anomalies has greatly improved our understanding of melanocyte biology. One such disorder, Waardenburg syndrome (WS), is a mendelian trait characterized by hypopigmentation and sensorineural deafness. It is commonly subdivided into four types (WS1-4), defined by the presence or absence of additional symptoms. WS type 4 (WS4), or Shah-Waardenburg syndrome, is also known as Hirschsprung disease Type II (HSCR II) and is characterized by an absence of epidermal melanocytes and enteric ganglia. Mutations in the genes encoding the endothelin type-B receptor (EDNRB)

Kynurenine pathway in Parkinson's disease: pathophysiological roles and therapeutic interventions.

Several disrupted metabolic pathways contributed to the development of Parkinson's disease (PD). Progressive death of dopamine (DA) neurons in the substantia nigra pars compacta, abnormal aggregation of α-synuclein fibrils, and inflammation of the neural system are the hallmarks of PD. The kynurenine pathway (KP) becomes disrupted, and excitotoxic branches are activated by elevated levels of central inflammatory regulators in PD. This leads to a significant reduction in the neural protective metabolite, kynurenic acid (KYNA), and an increase in the neurotoxic metabolite, quinolinic acid (QUIN)

Neural oscillations and top-down connectivity are modulated by object-scene congruency.

The knowledge we have about how the world is structured is known to influence object recognition. One way this is demonstrated is through a congruency effect, where object recognition is faster and more accurate if items are presented in expected scene contexts. However, our understanding of the dynamic neural mechanisms that underlie congruency effects are under-explored. Using magnetoencephalography (MEG), we examine how the congruency between an object and a prior scene results in changes in the oscillatory activity in the brain, which regions underpin this effect, and whether congruency re

Role of spinal inhibitory mechanisms in whiplash injuries.

Whiplash injury, commonly encountered in road traffic accidents, is a major cause of morbidity. Its pathophysiology is not well understood, and diagnosis remains clinical. Imaging and electrophysiological methods have not provided objective diagnostic evidence. Availability of a sensitive and specific diagnostic method would be of high clinical interest. We studied 20 consecutive patients with chronic whiplash injury. Despite persistent symptoms, most had minimal neurological findings. Cutaneous silent period (CSP), a nociceptive spinal inhibitory electromyographic reflex, showed 90% sensitivi

Neuro-immune regulation of sepsis-associated delirium via the PBN-CeA-spleen axis.

BACKGROUND: Sepsis associated delirium (SAD) is the most prevalent manifestation of acute brain dysfunction in sepsis, yet its central regulatory mechanisms remain incompletely understood. As researchers have progressively explored the brain-spleen axis and neuroimmunity, this study aimed to investigate the role of the parabrachial nucleus (PBN)-central amygdala (CeA)-spleen axis in the pathogenesis of SAD. METHODS: In a mouse model of SAD induced by intraperitoneal injection of lipopolysaccharide (LPS, 10 mg/kg), we identified activation patterns in both the PBN and CeA using immunofluoresce

Toward a Genetic Signature of Resistance to Activity-Based Anorexia in Striatal Projecting Cortical Neurons.

OBJECTIVE: Converging evidence from neuroimaging studies and genome-wide association study (GWAS) suggests the involvement of prefrontal cortex (PFC) and striatum dysfunction in the pathophysiology of anorexia nervosa (AN). However, identifying the causal role of circuit-specific genes in the development of the AN-like phenotype remains challenging and requires the combination of novel molecular tools and preclinical models. METHODS: We used the activity-based anorexia (ABA) rat model in combination with a novel viral-based translating ribosome affinity purification (TRAP) technique to identi

Metformin-Loaded Schwann Cell Exosomes Reprogram Macrophages and Enhance Neurogenesis in Spinal Cord Injury Through PI3K/AKT Activation.

Spinal cord injury (SCI) creates a hostile microenvironment characterized by persistent inflammation and glial scarring, which severely limits endogenous neural regeneration. To address these multifactorial barriers, we developed a targeted nanotherapeutic system comprising glutathione-functionalized Schwann cell-derived exosomes loaded with metformin (Exos-GSH@Met). In vitro and in vivo evaluations showed that GSH functionalization enabled the exosomes to effectively cross the blood-spinal cord barrier and selectively accumulate in macrophages at the injury site. Pin1 Downregulation Is Involved in Excess Retinoic Acid-Induced Failure of Neural Tube Closure.

Neural tube defects (NTDs), which are caused by impaired embryonic neural tube closure, are one of the most serious and common birth defects. Peptidyl-prolyl cis/trans isomerase 1 (Pin1) is a prolyl isomerase that uniquely regulates cell signaling by manipulating protein conformation following phosphorylation, although its involvement in neuronal development remains unknown. In this study, we explored the involvement of Pin1 in NTDs and its potential mechanisms both in vitro and in vivo. The levels of Pin1 expression were reduced in NTD models induced by all-trans retinoic acid (Atra). Pin1 pl

High-frequency repetitive transcranial magnetic stimulation mitigates depression-like behaviors in CUMS-induced rats via FGF2/FGFR1/p-ERK signaling pathway.

High-frequency repetitive transcranial magnetic stimulation (rTMS) is a widely used and effective biological treatment for depression. Although previous studies have shown that astrocyte function may be modified by rTMS, the specific neurobiological mechanisms underlying its antidepressant action are not clear. Substantial evidence has accumulated indicating that neurotrophin dysfunction and neuronal apoptosis play a role in the development of depression. To evaluate this hypothesis, we applied a chronical unpredictable mild stress (CUMS) protocol to induce depression-like behaviors in rats, f

Sonic Hedgehog Signaling Affected by Promoter Hypermethylation Induces Aberrant Gli2 Expression in Spina Bifida.

GLI2 is a key mediator of the sonic hedgehog (Shh) signaling pathway and plays an important role in neural tube development during vertebrate embryogenesis; however, the role of gli2 in human folate-related neural tube defects remains unclear. In this study, we compared methylation status and polymorphisms of gli2 between spina bifida patients and a control group to explore the underlying mechanisms related to folate deficiency in spina bifida. No single nucleotide polymorphism was found to be significantly different between the two groups, although gli2 methylation levels were significantly i

Multiple pathways for establishment of poliovirus infection.

Poliovirus (PV) infects susceptible cells through poliovirus receptor (PVR), which functions to bind virus and to convert its conformation. To study early infection process of PV, infection systems were employed using in vitro cultured cells and in vivo neural pathway of PVR transgenic (Tg) mice. For in vitro study, mouse L cells were established expressing mouse high affinity Fc gamma receptor molecules, and used them as in vitro PV infection system. PV infection was mediated, albeit inefficiently, by mouse anti-PV monoclonal antibodies (mAbs; IgG2a subtypes) that did not show an activity to

Cerebellar activity predicts vocalization in fruit bats.

Echolocating bats exhibit remarkable auditory behaviors, enabled by adaptations both within and outside their auditory system. Yet research on echolocating bats has focused mostly on brain areas that belong to the classic ascending auditory pathway. This study provides direct evidence linking the cerebellum, an evolutionarily ancient and non-classic auditory structure, to vocalization and hearing. We report that in the fruit-eating bat Carollia perspicillata, external sounds can evoke cerebellar responses with latencies below 20 ms. Such fast responses are indicative of early inputs to the bat

Functional mapping of the auditory tract in rodent tinnitus model using manganese-enhanced magnetic resonance imaging.

Animal models of salicylate-induced tinnitus have demonstrated that salicylate modulates neuronal activity in several brain structures leading to neuronal hyperactivity in auditory and non-auditory brain areas. In addition, these animal tinnitus models indicate that tinnitus can be a perceptual consequence of altered spontaneous neural activity along the auditory pathway. Peripheral and/or central effects of salicylate can account for neuronal activity changes in salicylate-induced tinnitus. Because of this ambiguity, an in vivo imaging study would be able to address the peripheral and/or cent

A primary cilia-dependent etiology for midline facial disorders.

Human faces exhibit enormous variation. When pathological conditions are superimposed on normal variation, a nearly unbroken series of facial morphologies is produced. When viewed in full, this spectrum ranges from cyclopia and hypotelorism to hypertelorism and facial duplications. Decreased Hedgehog pathway activity causes holoprosencephaly and hypotelorism. Here, we show that excessive Hedgehog activity, caused by truncating the primary cilia on cranial neural crest cells, causes hypertelorism and frontonasal dysplasia (FND). Elimination of the intraflagellar transport protein Kif3a leads to

Invasion and spread of single glycoprotein deleted mutants of Aujeszky's disease virus (ADV) in the trigeminal nervous pathway of pigs after intranasal inoculation.

The purpose of the study was to evaluate the role which non-essential envelope glycoproteins play in the neuroinvasion and neural spread of ADV. The invasion and spread in the trigeminal nervous pathway with the Ka strain of ADV and its single deletion mutants Ka gI-, Ka gp63- and Ka gIII- were examined after intranasal inoculation in neonatal pigs by virus isolation and immunocytochemistry. Evaluation was performed in the nasal mucosa, trigeminal ganglion (1st neuronal level), ponsmedulla (2nd neuronal level) and thalamus-cerebellum (3rd neuronal level). The Ka gIII- mutant invaded up to the

Non-neuronal acetylcholine: from cellular signaling to clinical implications.

For over a century, acetylcholine (ACh) was viewed primarily as a wired neurotransmitter of cholinergic nerves. However, accumulating evidence supports the view of the non-neuronal cholinergic system (NNCS) as an evolutionarily ancient autocrine and paracrine network operating across peripheral tissues. Rather than an extension of neuronal signaling, non-neuronal ACh can be viewed as a tissue-embedded chemical communication system positioned at the interface of cellular homeostasis and systemic neural reflexes. This review frames the NNCS as a network of tissue-resident ACh-producing cells tha

Orientation-tuned surround suppression exhibits a unique laminar signature in the human primary visual cortex.

Spatial context modifies visual perception by enhancing novel and salient features over spatially redundant features in the underlying neural code of the primary visual cortex (V1). Although multiple intracortical pathways contribute to contextual modulation, their specific contributions to different types of contextual modulation are not fully understood. Leveraging the distinct laminar connectivity patterns of feedforward, feedback, and lateral pathways, we used ultra-high-resolution fMRI (7T T2*-weighted, 0.6 mm isotropic resolution) to infer their relative contributions to contextual modul
