Cognitive components underpinning the development of model-based learning.

Reinforcement learning theory distinguishes "model-free" learning, which fosters reflexive repetition of previously rewarded actions, from "model-based" learning, which recruits a mental model of the environment to flexibly select goal-directed actions. Whereas model-free learning is evident across development, recruitment of model-based learning appears to increase with age. However, the cognitive processes underlying the development of model-based learning remain poorly characterized. Here, we examined whether age-related differences in cognitive processes underlying the construction and fle

Loss of peripheral vestibular input alters the statistics of head movement experienced during natural self-motion.

KEY POINTS: Sensory systems are adapted to the statistical structure of natural stimuli, thereby optimizing neural coding. Head motion during natural activities is first sensed and then processed by central vestibulo-motor pathways to influence subsequent behaviour, thereby establishing a feedback loop. To investigate the role of this vestibular feedback on the statistical structure of the head movements, we compared head movements in patients with unilateral vestibular loss and healthy controls. We show that the loss of vestibular feedback substantially alters the statistical structure of hea

Deep learning approaches using 2D and 3D convolutional neural networks for generating male pelvic synthetic computed tomography from magnetic resonance imaging.

PURPOSE: The improved soft tissue contrast of magnetic resonance imaging (MRI) compared to computed tomography (CT) makes it a useful imaging modality for radiotherapy treatment planning. Even when MR images are acquired for treatment planning, the standard clinical practice currently also requires a CT for dose calculation and x-ray-based patient positioning. This increases workloads, introduces uncertainty due to the required inter-modality image registrations, and involves unnecessary irradiation. While it would be beneficial to use exclusively MR images, a method needs to be employed to es

A python based algorithmic approach to optimize sulfonamide drugs via mathematical modeling.

This article explores the structural properties of eleven distinct chemical graphs that represent sulfonamide drugs using topological indices by developing python algorithm. To find significant relationships between the topological characteristics of these networks and the characteristics of the associated sulfonamide drugs. We use quantitative structure-property relationship (QSPR) approaches. In order to model and forecast these correlations and provide insights into the structure-activity relationships that are essential for drug design and optimization, linear regression is a vital tool. Cognition in mania and depression: psychological models and clinical implications.

Affective disorders, including bipolar disorder and major depressive disorder, are highly prevalent throughout the world and are extremely disabling. Diagnostic and Statistical Manual criteria and psychological models strongly implicate cognitive dysfunctions as being integral to our understanding of these disorders. We review the findings from studies that have used neurocognitive tests and functional imaging techniques to explore abnormal cognition in affective disorders. In particular, we highlight the evidence for cognitive dysfunctions that persist into full clinical remission, and the re

Real and perceived risks of medical radiation exposure.

There is almost no subject that elicits greater concern and less objective understanding than that of exposure of patients to radiation accompanying diagnostic radiologic procedures. One or more of the health effects (cancer, developmental abnormalties and genetic changes) attributed to low-level exposure to radiation are frequently misinterpreted as likely consequences rather than statistical risks following radiation exposure. Even less well understood is the small magnitude of the risks accompanying radiation exposure compared with those related to activities encountered during the daily ro

Tessellation analysis of glomerular spatial arrangement in mice with heritable renal hypoplasia.

Renal hypoplasia results from an insufficient kidney volume caused, in part, by a deficient number of glomeruli. The purpose of this study was to apply tessellation analysis to determine whether glomerular point patterns differed between adult normal (WT) and mutant (Br) mice with heritable renal hypoplasia and to delineate a spatial distribution accounting for the observed patterns. Kidneys from adult WT and Br mice were collected, processed with routine light histology and representative transverse sections were photographed. Cortical area and perimeter were calculated from traced tissue con

Pharmacokinetic/Pharmacodynamic Modeling of a Cell-Penetrating Peptide Phosphorodiamidate Morpholino Oligomer in mdx Mice.

PURPOSE: Peptide-conjugated phosphorodiamidate morpholino oligomers (PPMOs) have shown promise in treating Duchenne muscular dystrophy (DMD). We evaluated a semi-mechanistic pharmacokinetic (PK) and pharmacodynamic (PD) model to capture the relationship between plasma and muscle tissue exposure/response in mdx mice treated by mouse surrogate PPMO. METHODS: A single or repeated (every 4 weeks for 20 weeks) intravenous PPMO dose was administered to mdx mice (n = 6/timepoint). A PK/PD model was built to characterize data via sequential modeling. A 2-compartment model was used to describe plasma

Use of a marginal structural model to determine the effect of aspirin on cardiovascular mortality in the Physicians' Health Study.

The 1982-1988 aspirin component of the Physicians' Health Study, a randomized trial of aspirin and beta-carotene in primary prevention of cardiovascular disease and cancer among 22,071 US male physicians, was terminated early primarily because of a statistically extreme 44% reduction in first myocardial infarction, with inadequate precision and no apparent effect on the primary endpoint, cardiovascular death. Because of the demonstrated efficacy of aspirin in secondary prevention of cardiovascular death, nonfatal cardiovascular events may simultaneously be time-dependent confounders and interm

Digital Twins in Toxicology: Toward Predictive, Personalized and Real-Time Risk Assessment.

Toxicology has accumulated many predictive tools - physiologically based pharmacokinetic (PBPK) models, quantitative systems pharmacology (QSP) frameworks organ-on-chip systems, quantitative structure-activity relationship (QSAR) models, adverse outcome pathway (AOP) networks and machine-learning approaches. Yet predictive practice remains fragmented. Each tool captures a slice of biology; few capture the whole; none captures the patient. We argue that the concept of a digital twin - a continuously updated, mechanistically grounded, data-driven digital representation of an individual biologica

Information Leakage in Enzyme Substrate Prediction.

MOTIVATION: Enzymes are essential catalysts in many cellular processes. Understanding their interactions with small molecules, such as regulators, cofactors, and most importantly, substrates, is crucial for understanding the biochemical processes that occur in cells. Correctly interpreting the roles of small molecules that interact with enzymes is key to elucidating enzyme function. Recently, enzyme-small molecule interaction prediction has attracted growing interest from computational methods, especially deep learning. As a result, researchers have published several datasets and numerous mode

How to determine when data incompleteness compromises the correct training of data-enhanced mechanical models of haemodialysis? A data quality criterion.

OBJECTIVE: Hemodialysis is essential for managing end-stage renal disease, with rising patient numbers increasing the need for individualized treatment. To support this, hybrid models-integrating mechanistic frameworks with data-driven calibration-have increasingly replaced purely mechanistic or statistical approaches. These models rely on complete, high-quality clinical data; however, incomplete values-particularly hematological data-are common and may compromise model reliability. In this study, we evaluated the impact of incomplete data acquired during dialysis on the training and predictiv

Diffusion models vs. DCGANs for class-imbalanced lung cancer CT classification: A comparative study.

Effective lung cancer detection from CT scans remains critically challenged by class imbalance where benign and normal cases are underrepresented, leading to biased machine learning models with reduced sensitivity for minority classes and potentially missed diagnoses in cancer screening applications. We present a comprehensive comparative analysis of Diffusion Models and Deep Convolutional Generative Adversarial Networks (DCGANs), both incorporating modern architectural enhancements including spectral normalization, self-attention mechanisms, and conditional generation, for addressing class im
