Evaluation of a wipe surface sample method for collection of Bacillus spores from nonporous surfaces.

Polyester-rayon blend wipes were evaluated for efficiency of extraction and recovery of powdered Bacillus atrophaeus spores from stainless steel and painted wallboard surfaces. Method limits of detection were also estimated for both surfaces. The observed mean efficiency of polyester-rayon blend wipe recovery from stainless steel was 0.35 with a standard deviation of +/-0.12, and for painted wallboard it was 0.29 with a standard deviation of +/-0.15. Evaluation of a sonication extraction method for the polyester-rayon blend wipes produced a mean extraction efficiency of 0.93 with a standard de

Chemical, biological, radiological and nuclear terrorism: an introduction for occupational physicians.

BACKGROUND: Chemical, biological, radiological and nuclear terrorism poses considerable threat throughout the world. AIM: To provide occupational physicians with an understanding of this threat and its main forms and what action can be taken to counter this threat. METHODS: Presenters at a conference on chemical, biological, radiological and nuclear terrorism were asked to contribute their evidence-based opinions in order to produce a review article. RESULTS: This paper presents a summary of the different forms of chemical, biological, radiological and nuclear terrorism and the effective co

A miniature biochip system for detection of aerosolized Bacillus globigii spores.

The feasibility of using a novel detection scheme for the analysis of biological warfare agents is demonstrated using Bacillus globigii spores, a surrogate species for Bacillus anthracis. In this paper, a sensitive and selective enzyme-linked immunosorbent assay using a novel fluorogenic alkaline phosphatase substrate (dimethylacridinone phosphate) is combined with a compact biochip detection system, which includes a miniature diode laser for excitation. Detection of aerosolized spores was achieved by coupling the miniature system to a portable bioaerosol sampler, and the performance of the an

Bacillus anthracis evolution and epidemiology.

Bacillus anthracis is a pathogen that is widely distributed around the globe. However, this great distribution is not accompanied by great genetic diversity. Although subtle morphological and biochemical differences exist, the underlying genetic basis for this plasticity is not known. Indeed, very few single nucleotide differences have been detected among isolates and the only documented high variable sequences are associated with variable number tandem repeated (VNTR) sequences. The differences among the VNTRs has been used to suggest phylogenetic relationships among the worldwide isolates. T

Interim recommendations for protecting workers from exposure to Bacillus anthracis in work sites in which mail is handled or processed.

CDC has developed interim recommendations to assist personnel responsible for occupational health and safety in developing a comprehensive program to reduce potential cutaneous or inhalational exposures to Bacillus anthracis spores among workers in work sites in which mail is handled or processed. Such work sites include post offices, mail distribution/handling centers, bulk mail centers, air mail facilities, priority mail processing centers, public and private mail rooms, and other settings in which workers are responsible for handling and processing mail. The recommendations are based on the

[The characteristics of the interaction of Bacillus anthracis with host phagocytes in relation to the plasmid spectrum of the causative agent].

The study revealed that after the intraperitoneal inoculation of spores of B. anthracis strains with different plasmid composition into guinea pigs the active germination of the spores both outside and inside the cells of the host occurred as early as on hour 2 of their interaction with the macroorganism. The further fate of the infective agent and the character of its interaction with peritoneal exudate cells depended on the plasmid composition of the bacilli. Thus, the presence of toxin-formation plasmid PXO1 and capsule-formation plasmid PXO2 in B. anthracis permitted the successful adaptat

Effects of chemical and biological warfare agent decontaminants on trace survival: Impact on fingermarks deposited on paper.

Chemical and biological events present a challenging environment for forensic scientists to perform their work. This research is the follow-up of the previous article that investigated the impact of decontaminants on fingermarks deposited on glass. In the present study, the focus shifted to paper, a porous and sensitive substrate where fingermark residues are absorbed and cannot be optically seen, making their detection more difficult. Paper is also highly relevant in forensic casework, as demonstrated by the anthrax letter events. Here the impact of 16 decontaminants on fingermarks deposited …

Group 3 innate lymphocytes (ILC3s) upregulate IL-22 in response to elevated intracellular cAMP levels.

Group 3 innate lymphocytes (ILC3s) are important immune cells within mucosal tissues and protect against bacterial infections. They can be activated in response to the innate cytokines IL-23 or IL-1β, which rapidly increases their production of effector molecules that regulate barrier functions. Pathogens can subvert these anti-bacterial effects to evade mucosal defenses to infect the host. Bacillus anthracis, the causative agent of anthrax, produces two major toxins that can modulate the immune response. We have previously shown that lethal toxin downmodulates the function of ILC3s. On the ot…
