Inhibitory Potential of Chromene Derivatives on Structural and Non-Structural Proteins of Dengue Virus.

Dengue fever is a mosquito-borne viral disease that has become a serious health issue across the globe. It is caused by a virus of the Flaviviridae family, and it comprises five different serotypes (DENV-1 to DENV-5). As there is no specific medicine or effective vaccine for controlling dengue fever, there is an urgent need to develop potential inhibitors against it. Traditionally, various natural products have been used to manage dengue fever and its co-morbid conditions. A detailed analysis of these plants revealed the presence of various chromene derivatives as the major phytochemicals. Ins

Dengue and Zika viruses: lessons learned from the similarities between these Aedes mosquito-vectored arboviruses.

The currently spreading arbovirus epidemic is having a severe impact on human health worldwide. The two most common flaviviruses, dengue virus (DENV) and Zika virus (ZIKV), are transmitted through the same viral vector, Aedes spp. mosquitoes. Since the discovery of DENV in 1943, this virus has been reported to cause around 390 million human infections per year, approximately 500,000 of which require hospitalization and over 20,000 of which are lethal. The present DENV epidemic is primarily concentrated in Southeast Asia. ZIKV, which was discovered in 1952, is another important arthropod-borne

A Video-Tracking Analysis-Based Behavioral Assay for Larvae of Anopheles pseudopunctipennis and Aedes aegypti (Diptera: Culicidae).

Aedes aegypti (L.) is the primary vector of dengue, yellow fever, Zika, and chikungunya viruses, whereas Anopheles pseudopunctipennis (Theobald) is the principal vector for malaria in Latin America. The larval stage of these mosquitoes occurs in very different development habitats, and the study of their respective behaviors could give us valuable information to improve larval control. The aim of this study was to set up a bioassay to study basic larval behaviors using a video-tracking software. Larvae of An. pseudopunctipennis came from two localities in Salta Province, Argentina, while Ae. a

Human infections with Plasmodium knowlesi--zoonotic malaria.

In 2004 a large focus of Plasmodium knowlesi malaria was reported in the human population in Sarawak, Malaysian Borneo. Plasmodium knowlesi, a parasite of the South-East Asian macaques (Macaca fascicularis and Macaca nemestrina), had entered the human population. Plasmodium knowlesi is transmitted by the leucosphyrus group of Anopheline mosquitoes and transmission is largely zoonotic and restricted to the jungle setting. Humans entering jungle transmission sites are at risk. Since 2004, human cases of P. knowlesi have been continuously reported in local communities and in travellers returning

Occurrence of immature forms of culicids (Insecta: Diptera) in the northeastern region of Brazil.

The Culicidae family is represented by approximately 3,610 species, among which many are involved in transmission of pathogens and parasites, thus presenting great medical-veterinary importance. In Brazil, the state of Pernambuco is considered an endemic area for many diseases vectored by mosquitoes, such as human and canine filariasis, caused by Wuchereria bancrofti and Dirofilaria immitis, respectively. The aim of this study was to evaluate the occurrence of immature forms of culicids, over an one-year period, in a specific area of northeastern region of Brazil. Culicids larvae were collecte

New infectious etiologies for posterior uveitis.

Emergent and resurgent arthropod vector-borne diseases are major causes of systemic morbidity and death and expanding worldwide. Among them, viral and bacterial agents including West Nile virus, Dengue fever, Chikungunya, Rift Valley fever, and rickettsioses have been recently associated with an array of ocular manifestations. These include anterior uveitis, retinitis, chorioretinitis, retinal vasculitis and optic nerve involvement. Proper clinical diagnosis of any of these infectious diseases is based on epidemiological data, history, systemic symptoms and signs, and the pattern of ocular inv

Complement receptor 1 and malaria.

Plasmodium falciparum malaria is an intracellular parasite that is transmitted by Anopheles mosquitoes. It is responsible for approximately 1 million deaths per year. Most deaths occur as a result of complications such as severe anaemia or cerebral malaria (coma). The complement receptor 1 is a key complement regulator found on the surface of red cells and most leucocytes. A growing body of evidence suggests that this molecule plays a critical role in the pathogenesis of P. falciparum malaria. Initial reports showed that CR1 enables the binding of infected red cells to uninfected red cells to

[Saint Louis encephalitis virus in Buenos Aires city during the outbreak of dengue in 2009].

We report the case of a male, 80-year-old resident in the City of Buenos Aires, with a diagnosis of St. Louis encephalitis (SLE) during a countrywide dengue outbreak, from January to May 2009. The patient had a chronic lymphocytic leukemia treated with chlorambucil, prostate cancer (hormone therapy and radiotherapy) and images consistent with bone metastases. Cerebrospinal fluid examination showed pleocytosis with a predominance of mononuclear cells and high protein concentration. Bacteria, fungi and mycobacteria cultures, as well as the PCR for herpes virus, HSV, CMV and EBV, were negative. W

Urban malaria in Accra, Ghana: transmission risk and influence of irrigated vegetable farms.

Urban malaria transmission in sub-Saharan Africa is often underestimated, emphasizing the need for research on vector distribution, abundance, behavior, and infectivity in cities. Urban malaria transmission has been on the rise across sub-Saharan Africa since 2003. Also, monitoring Anopheles populations is crucial for developing effective interventions. This study examined the biting and resting habits, distribution, abundance, and Plasmodium infection of malaria vectors in Accra. Adult malaria vectors were collected using human landing catch (HLC) and Prokopack aspiration (PPK) at ten sites i

Malaria transmission in the coastal zone and in the Centre of Côte d'Ivoire during the dry season.

BACKGROUND: Malaria remains a threat in sub-Saharan Africa, particularly in Côte d'Ivoire, where it is endemic and represents the leading cause of hospital consultations, morbidity and mortality. The strong climatic variations that exist between coastal and savannah areas of Côte d'Ivoire suggest that vector control interventions should be scheduled according to the eco-epidemiological diversity. This study evaluates bioecological parameters of vectors and malaria transmission in two health districts, one coastal and one central of Côte d'Ivoire. METHODS: The study was conducted in the coasta

Antiviral Wolbachia strains associate with Aedes aegypti endoplasmic reticulum membranes and induce lipid droplet formation to restrict dengue virus replication.

Wolbachia are a genus of insect endosymbiotic bacteria which includes strains wMel and wAlbB that are being utilized as a biocontrol tool to reduce the incidence of Aedes aegypti-transmitted viral diseases like dengue. However, the precise mechanisms underpinning the antiviral activity of these Wolbachia strains are not well defined. Here, we generated a panel of Ae. aegypti-derived cell lines infected with antiviral strains wMel and wAlbB or the non-antiviral Wolbachia strain wPip to understand host cell morphological changes specifically induced by antiviral strains. Antiviral strains were f

Stochastic Bayesian Runge-Kutta method for dengue dynamic mapping.

Dengue Hemorrhagic Fever (DHF) is still a threat to humanity that cause death and disability due to changes in environmental and socioeconomic conditions, especially in tropical areas. A critical assessment of the models and methods is necessary. The vital role of stochastic processes of infectious disease research in Bayesian statistical models helps provide an explicit framework for understanding disease transmission dynamics between hosts (humans) and vectors (mosquitoes). This research presents a Bayesian stochastic process for the cross-infection SIR-SI model as a differential equation (s

Efficacy of afoxolaner (NexGard®) in preventing the transmission of Leishmania infantum and Dirofilaria immitis to sheltered dogs in a highly endemic area.

BACKGROUND: Leishmania infantum and Dirofilaria immitis are among the most important canine vector-borne pathogens (CVBPs) of zoonotic concern in Europe. In endemic areas for both of these CVBPs, the use of systemic ectoparasiticides, such as afoxolaner (NexGard®; Boehringer Ingelheim Animal Health), may have the potential for controlling these infections. The aim of this study was to assess, for the first time, the insecticidal efficacy of NexGard® in decreasing the transmission of D. immitis and L. infantum to sheltered dogs living in a hyperendemic area, compared to the year before treatmen

Magnitude of Antibody Cross-Reactivity in Medically Important Mosquito-Borne Flaviviruses: A Systematic Review.

INTRODUCTION: Flaviviruses are a genus of enveloped single-stranded RNA viruses that include dengue virus (DENV), yellow fever virus, West Nile virus (WNV), Japanese encephalitis virus, and Zika virus. Nowadays, diverse serological assays are available to diagnose flaviviruses. However, infection with flaviviruses induces cross-reactive antibodies, which are a challenge for serological diagnosis. OBJECTIVE: This systematic review aimed to assess the magnitude of medically important mosquito-borne flavivirus-induced antibody cross-reactivity and its influence on serological test outcomes. MET

Involvement of Anopheles nili in Plasmodium falciparum transmission in North Benin.

BACKGROUND: Several studies carried out in Benin have shown the involvement of Anopheles gambiae sensu lato (s.l.), the Anopheles funestus group in malaria transmission, but none of them reported the contribution of the Anopheles nili group to the transmission of this disease. The current study investigated the question through an entomological cross-sectional survey performed in Northern Benin. METHODS: Mosquito samplings were performed in September and October 2017 in 4 villages located in two districts: Bambaba and Wodara (Kérou district) and, Péhunco 2 and Béké (Péhunco district). West Nile virus infection. What you need to know about this emerging threat.

The most important aspects to remember about West Nile virus infection are summed up in the following points: West Nile virus is transmitted by bites from infected Culex species mosquitoes. Most infections do not result in serious illness. Encephalitis and serious morbidity and mortality are more likely to occur in elderly persons than in any other age-group. No effective antiviral treatment is available. Avoiding contact with mosquitoes is the key preventive measure.

Assessment of antibody responses to Anopheles SG6-P1 and Aedes N-term 34 kDa salivary peptides: a randomised human-challenge trial of controlled exposures to vector bites.

BACKGROUND: Human antibodies against mosquito salivary proteins are proposed as proxy biomarkers of exposure to vector bites. This trial sought to characterise the boosting and decay dynamics of antibodies against Anopheles SG6-P1 and Aedes N-term 34kDa salivary peptides in a human challenge model of controlled exposure to the main Southeast Asian malaria and global dengue vectors. METHODS: In this single-centre, open-label, randomised, exploratory factorial trial, healthy volunteers aged 18-60 years with no history of recent travel to rural areas were recruited in Mae Sot, Thailand (ClincalT
