Differentially regulated expression of 9-O-acetyl GD3 (CD60b) and 7-O-acetyl-GD3 (CD60c) during differentiation and maturation of human T and B lymphocytes.

GD3 (CD60a) and its 9-O-acetylated variant (CD60b) are intracellular regulators of apoptosis in T lymphocytes. Surface expressed 9-O-acetyl- and 7-O-acetyl-GD3 (CD60b and CD60c) may have a functional impact on activated T and B cells. In order to investigate the balance between surface and intracellular expression and synthesis and degradation of these glycosphingolipids in human lymphocytes of various differentiation stages, we analyzed (i) expression of GD3 molecules on native T and B cells and thymocytes by flow cytometry and (ii) activity and regulation of possible key enzymes for CD60a,b,

Characterization of the enzymatic 7-O-acetylation of sialic acids and evidence for enzymatic O-acetyl migration from C-7 to C-9 in bovine submandibular gland.

Microsomes prepared from bovine submandibular glands incubated with radioactive AcCoA incorporated acid-insoluble radioactivity, which was dependent on time, and the concentrations of AcCoA and proteins, and was inhibited by CoA in a concentration-dependent manner. Under the conditions used, the apparent Km for AcCoA was 1.63 microM with a Vmax of 21.9 pmol/mg protein.min. The radioactivity incorporated was mainly due to the O-acetylation of glycosidically bound Neu5Ac. The primary attachment site of O-acetyl groups was exclusively the hydroxyl at C-7 of Neu5Ac, the presence of an AcCoA:Neu5Ac…

Association between Glycoprotein Acetylation-Indicated Inflammation and Small HDL Particle Number in Coronary Artery Disease.

BACKGROUND: Glycoprotein acetylation (GlycA) is a nuclear magnetic resonance-derived biomarker of systemic inflammation. GlycA shows statistical interaction with small HDL particle number (s-HDL-P) in relation to coronary artery disease (CAD). This study examines the confounding effects of GlycA-related glycoproteins, hypertriglyceridemia, and s-HDL-P on CAD risk. METHODS: Plasma samples from 752 participants in the CATHGEN biorepository were size-fractionated by asymmetric flow field-flow fractionation and analyzed by targeted proteomics and lipidomics. Fraction concentrations were converted…

Lysine acetylation-mediated regulation of ferredoxin and ferredoxin reductase redox-active proteins in Haloferax volcanii.

Lysine acetylation is an evolutionarily conserved, post-translational modification that regulates metabolism and protein function, yet its role in archaeal electron transfer systems remains poorly understood. Here, we investigated lysine acetylation of the 2Fe-2S ferredoxin Hv Fdx (HVO_2995) and its flavin-dependent oxidoreductase Hv FdR (HVO_2345) partner in the halophilic archaeon Haloferax volcanii . Genetic and biochemical analyses established Hv Fdx as an essential 2Fe-2S ferredoxin with a midpoint redox potential of -385 mV. Lysine acetylation of Hv Fdx was found to occur primarily at K1…

HOXD10 expression in oral cancer is driven by histone acetylation rather than promoter DNA methylation.

OBJECTIVE: HOXD10 is implicated in the carcinogenesis and progression of various cancers, including oral cancer. However, its regulatory mec … expression. Two promoter regions were identified as active regulatory elements modulated by histone acetylation. Functionally, NaB impaired cell proliferation, migration and induced G2-M arrest. Survival analysis demonstrated modest prognostic value of HOXD10 alone but improved predictive accuracy for disease recurrence when integrated with tumor stage and gr …

Targeting NAT10 alleviates colonic senescence and elderly-onset colitis by disrupting N4-acetylation of DYRK1A.

Intestinal aging characterized by imbalance between cell senescence and mucosal self-renewal, increases susceptibility to the elderly-onset ulcerative colitis (UC), while the underlying mechanisms remain elusive. Here, we identify mRNA N4-acetylcytidine (ac4C) modification and its specific writer, N-acetyltransferase 10 (NAT10), as critical regulators of human colonic epithelial cell senescence. Knockdown of NAT10 significantly alleviates human colonic epithelial cell senescence in vitro and colonoid and intestinal aging in vivo in aged mice. Using ac4C-modified transcriptome sequencing, we re

HDAC5 depletion promotes hyper-acetylation of FOXA1 and potentiates HIF1α transcriptional activation in pancreatic cancer.

The biological significance of forkhead box A1 (FOXA1) in non-steroid-driven malignancies, such as pancreatic ductal adenocarcinoma (PDAC), has garnered increasing recognition. It assumes a pivotal role in regulating critical processes such as PDAC cell lineage, metabolism, and metastasis. However, its regulatory mechanisms remain elusive. Here, we demonstrate that histone deacetylase 5 (HDAC5) mediates the deacetylation of FOXA1 at lysine residue 270 (K270), leading to repression of FOXA1's global chromatin occupancy. In HDAC5-loss PDAC, K270 hyper-acetylated FOXA1 is reprogrammed to the tran…

N-terminal acetylation controls multiple functional aspects of the influenza A virus ribonuclease PA-X.

To counteract host antiviral responses, influenza A virus triggers a global reduction of cellular gene expression, a process termed "host shutoff." A key effector of influenza A virus host shutoff is the viral endoribonuclease PA-X, which degrades host mRNAs. While many of the molecular determinants of PA-X activity remain unknown, a previous study found that N-terminal acetylation of PA-X is required for its host shutoff activity upon ectopic expression. However, it remains unclear how this co-translational modification promotes PA-X activity. Here, we report that PA-X N-terminal acetylation …

NAT10 governs development through the Xrp1-mediated stress response rather than its RNA acetylation activity.

N4-acetylcytidine (ac4C) is installed by N-acetyltransferase 10 (NAT10) and represents the only known acetylation mark on messenger RNA. Since NAT10 also acetylates transfer RNA, ribosomal RNA (rRNA), and proteins, its in vivo molecular mechanisms remain elusive. Here, we demonstrate that knockdown of Drosophila NAT10 induces an eye-to-antenna transformation, c-Jun amino-terminal kinase activation, and cell apoptosis. NAT10 facilitates ac4C modification in rRNA, and its loss impairs rRNA processing and ribosomal assembly. Depletion of NAT10 activates the integrated stress response, ultimately …

Role of Nitric Oxide, Ethylene, Hydrogen Sulfide and Their Interplay With Histone Acetylation in Plants.

Histone acetylation is an essential epigenetic modification that regulates chromatin structure and gene expression by opposing the actions of histone acetyltransferases (HATs) and deacetylases (HDACs). Gas signaling molecules are endogenous gaseous compounds that can freely diffuse across biological membranes. They mediate interplant communications and plant signaling transductions, with representative members including nitric oxide (NO), ethylene (Eth), hydrogen sulfide (H2S), and methane (CH4). Multiple studies have revealed that gas signaling molecules and histone acetylation can interact t…

NAT10 promotes proliferation, invasion, and epithelial-mesenchymal transition in hepatocellular carcinoma through α-tubulin acetylation.

Hepatocellular carcinoma (HCC) remains a global health challenge due to its high metastatic potential and poor prognosis. This study investigated the role and underlying molecular mechanisms of N-acetyltransferase 10 (NAT10)-mediated α-tubulin acetylation in HCC progression. Using Huh-7 and SNU-449 cell lines, we established stable knockdown and overexpression models to systematically evaluate both gain- and loss-of-function effects. Functional assays revealed that NAT10 suppression significantly decreased α-tubulin acetylation, thereby inhibiting cell proliferation, invasion, and migration. C…

Reversion of aortic valve cells calcification by activation of Notch signalling via histone acetylation induction.

Calcification of the aortic valve is a prevalent cardiovascular pathology in the aging population. Traditionally linked to inflammation, lipid accumulation, and risk conditions, this disease remains poorly understood, and effective treatments to halt its progression are not yet available. We hypothesized that calcification of the human valve interstitial cells (VICs) is associated with cellular senescence and alterations in the epigenetic setup, like in arteries. To verify this hypothesis, we examined the epigenetic marks (DNA methylation; Histones H3/H4 acetylation/methylation), the senescenc…

Cystine import and oxidative catabolism fuel vascular growth and repair via nutrient-responsive histone acetylation.

Endothelial metabolism underpins tissue regeneration, health, and longevity. We uncover a nuclear oxidative catabolic pathway linking cystine to gene regulation. Cells preparing to proliferate upregulate the SLC7A11 transporter to import cystine, which is oxidatively catabolized by cystathionine-γ-lyase (CSE) in the nucleus. This generates acetyl units via pyruvate dehydrogenase, driving site-specific histone H3 acetylation and chromatin remodeling that sustain endothelial transcription and proliferation. Combined loss of SLC7A11 and CSE abolishes cystine oxidative and reductive metabolism and…

Acetylation of the Mitochondrial Chaperone GRP75 Governs ER-Mitochondrial Calcium Homeostasis and Hepatocyte Insulin Resistance.

Overnutrition exacerbates insulin resistance (IR) and is linked to excessive mitochondrial protein acetylation. However, the molecular mechanism by which mitochondrial protein acetylation influences hepatic IR remains incompletely elucidated. To investigate this biology, GCN5L1 liver knockout mice (LKO), which exhibit blunted mitochondrial protein acetylation are utilized. Interestingly, the hepatocytes of LKO mice exhibit impaired insulin signaling and exaggerated endoplasmic reticulum (ER) stress. To explore putative mechanisms, protein-interaction and acetyl-proteome analyses are conducted …
