Effects of naloxone and diprenorphine on amphetamine-stimulated behavior in guinea pigs and rats.

Amphetamine (0.1-10 mg/kg), naloxone (0.1-10 mg/kg) and diprenorphine (0.03-10 mg/kg) were studied for their ability to modulate locomotor behavior in the guinea pig. Naloxone, administered alone, caused a non-significant decrease in locomotor activity and had a similar non-significant effect on amphetamine-stimulated activity. Diprenorphine induced a significant suppression of locomotor activity, the magnitude of which was inversely related to dose: smaller doses of diprenorphine caused a greater suppression of locomotor activity than larger doses. Two doses of diprenorphine (0.1 and 1.0 mg/k…

Effects of naloxone on heroin-, amphetamine- and caffeine-stimulated locomotor activity in the rat.

We investigated the effects of naloxone on the locomotor activating properties of heroin (0.25 mg/kg SC), d-amphetamine (0.25 mg/kg SC) and caffeine (7.5 mg/kg SC). Naloxone eliminated heroin-stimulated locomotion at doses approximately six times lower than those that blocked amphetamine-stimulated locomotion. Caffeine-induced locomotor activation was insensitive to naloxone at all doses tested. These results suggest that central opiate systems are differentially involved in the behavioral activation produced by heroin, amphetamine and caffeine.

Behavioural tolerance to amphetamine and other psychostimulants: the case for considering behavioural mechanisms.

An hypothesis is presented about the nature of behavioural tolerance in animals to stimulant drugs. It is suggested that, in many behavioural procedures, tolerance is due to behavioural adaptation to those drug effects which cause disruption of ongoing rewarded behaviour. This unitary hypothesis accounts for the available data on tolerance and cross-tolerance to stimulants more effectively than all of the other more conventional explanations which are based upon dispositional or functional concepts, the most common of which are described, evaluated, and found to be inadequate. Furthermore, it

Relative potency of amphetamine isomers in causing the serotonin behavioral syndrome in rats.

High doses of amphetamine evoke a behavioral syndrome in man which is similar to paranoid schizophrenia. The similarities between the behavioral effects of amphetamine and the symptoms of an endogenous behavior disorder suggest a mechanistic relationship between the two states. This study deals with a specific set of behavioral signs in the rat which are caused by relatively high doses of amphetamine and are mediated by serotonin receptor activation. Experiments on the mechanism by which amphetamine evokes this behavioral syndrome reveal that the l-isomer, like the d-isomer, acts indirectly, i…

Role of brain monoamines in the anorectic activity of mazindol and d-amphetamine in the rat.

The interaction of mazindol and d-amphetamine with brain monoamines was studied in rats. At each dose used, both compounds markedly counteracted the decrease of brain noradrenaline induced by 6-hydroxydopamine while only at high doses they did significantly reduce the effect of 6-hydroxydopamine on brain dopamine. Unlike d-amphetamine, mazindol significantly counteracted the decrease of brain serotonin induced by fenfluramine. The anorectic effect of mazindol and of d-amphetamine was markedly reduced by an electrolytic lesion at the level of the ventral noradrenergic bundle but not by an elect…

Simultaneous ultra-high-pressure liquid chromatography-tandem mass spectrometry determination of amphetamine and amphetamine-like stimulants, cocaine and its metabolites, and a cannabis metabolite in surface water and urban wastewater.

An ultra-high-pressure liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) method has been developed for the simultaneous quantific … [limit of quantification (LOQ) and 10LOQ]. Thus, surface water was spiked at 30 ng/L and 300 ng/L (amphetamine and amphetamine-like stimulants), 10 ng/L and 100 ng/L (cocaine and its metabolites), 300 ng/L and 3000 ng/L (tetrahydrocannabinol-COOH). Recovery experiments in effluent and influent wastewater were performed at spiking levels of three and fifteen …

Fusobacterium nucleatum enhances amphetamine-induced behavioral responses through a butyrate-driven epigenetic mechanism.

Amphetamines are psychostimulants that are commonly used to treat neuropsychiatric disorders and are prone to misuse. The pathogenesis of amphetamine use disorder (AUD) is associated with dysbiosis (an imbalance in the body's microbiome) and bacterially produced short-chain fatty acids (SCFAs), which are implicated in the gut-brain axis. Amphetamine exposure in both rats and humans increases the amount of intestinal Fusobacterium nucleatum, which releases SFCAs. Here, we found that colonization of gnotobiotic Drosophila melanogaster with F. nucleatum or supplementing the flies' diet with the S…

Amphetamine-Induced Psychosis Leading To Homicide, Suicidal Attempts, And Disorientation: A Case Report.

Amphetamine abuse leads to severe psychiatric and cardiovascular complications-psychosis, hallucinations and aggressive behavior. A 34-year-old male of five years noted history of amphetamine abuse and presented with amphetamine psychosis characterized by hallucinations, violent ideations and a recent suicide attempt is described in this case report. Previously under the influence of amphetamines, two years prior, the patient committed homicide. Patient had periods of very severe disorientation and agitation, history of suicidal ideation and violent thoughts toward others. Toxicology screening…

Opposite Contractile Effects of Amphetamine-Related Hallucinogenic Drugs in the Isolated Human Atrium.

The present study examined three hallucinogenic amphetamine derivatives, namely, 2,5-dimethoxy-4-iodoamphetamine (DOI) as well as 2,5-dimethoxy-4-methylamphetamine (DOM) and 4-methylmethcathinone (mephedrone). The objective of this study was to test the hypothesis that DOI, DOM, and mephedrone would increase the contractile force in isolated human atrial preparations in a manner similar to amphetamine. To this end, we measured contractile force under isometric conditions in electrically stimulated (1 Hz) human atrial preparations obtained during open surgery. DOI and DOM alone or in the presen…

Hypertensive Emergency Secondary to Combining Psilocybin Mushrooms, Extended Release Dextroamphetamine-Amphetamine, and Tranylcypromine.

Data on medication interactions with psychedelics are limited. Here we present what may be the first published report of a hypertensive emergency following the combination of psilocybin mushrooms with a monoamine oxidase inhibitor (MAOI). A 42-year-old man with treatment-resistant major depressive disorder took 1 g of Psilocybe cubensis mushrooms, while prescribed tranylcypromine, extended-release dextroamphetamine-amphetamine, and other medications. Approximately half an hour later, he developed severe hypertension with chest pain, palpitations, and headache. Upon hospital presentation, the e…

Cocaine and amphetamine regulated transcript (CART): a newly characterized neuropeptide in human prostate.

Cocaine and amphetamine regulated transcript (CART) is a somatostatin-like polypeptide. CART has been localized in the CNS, hypothalamo-pituitary-adrenocortical (HPA) axis, pancreatic islets and enteric nervous system. We investigated the cellular localization of CART in normal human prostate, benign prostatic hyperplasia, prostatic intraepithelial neoplasia and acinar adenocarcinoma. CART was assessed using immunohistochemistry (IHC) and in situ hybridization (ISH), and its gene expression was identified by RTqPCR. We found cellular expression of CART in both normal prostatic luminal secretor…
