Advances in the understanding of talaromycosis in HIV-negative patients (especially in children and patients with hematological malignancies): A comprehensive review.

Talaromyces marneffei (T. marneffei) stands out as the sole thermobiphasic fungus pathogenic to mammals, including humans, within the fungal community encompassing Ascomycota, Eurotium, Eurotiumles, Fungiaceae, and Cyanobacteria. Thriving as a saprophytic fungus in its natural habitat, it transitions into a pathogenic yeast phase at the mammalian physiological temperature of 37°C. Historically, talaromycosis has been predominantly associated with human immunodeficiency virus (HIV)/acquired immunodeficiency syndrome (AIDS), classified among the three primary opportunistic infections linked with…

Describing the intestinal microbiota of Holstein Fasciola-positive and -negative cattle from a hyperendemic area of fascioliasis in central Colombia.

The ability to identify compositional changes in the intestinal microbiota of parasitized hosts is important for understanding the physiolog … r detection analysis. Our results showed a reduction in the relative abundance of Bacteroidetes and Ascomycota in the Fasciola-positive samples, along with decreased relative abundances of the commensal taxa previously associated with fermentation and digestion processes. However, metabolomic approaches and functional analyzes of the intestinal microbiota ar …

The lichen symbiont Wadeana represents an independent origin of hysterothecial fungi in the Eurotiomycetes.

The genus Wadeana was erected almost fifty years ago to accommodate an unusual pair of lichen fungal symbionts with hysterothecial ascomata and polysporous asci associated with the algal genus Trentepohlia. The evolutionary position of Wadeana has long been unclear, and the genus has variably been treated as a dothideomycete, as incertae sedis or, recently, within the subclass Acarosporomycetidae of the Lecanoromycetes. Here we present multilocus evidence from metagenome-assembled genomes from the generitype, Wadeana dendrographa, that place it with strong support as sister to all genera curre

Dual biocontrol strategy: Fluorescent pseudomonads and methylotrophic bacteria in combating Corynespora leaf spot in blackgram.

Blackgram leaf spot, caused by Corynespora cassiicola, is a serious disease that significantly reduces global blackgram production. The non-commercial nature of the crop and growing nutritional significance in recent years made it necessary to implement an economical and environmentally responsible disease management strategy. In this line, twenty isolates each of fluorescent pseudomonads (FP) and facultative methylotrophs (FM) were isolated from the black gram phyllosphere. In vitro dual-culture assays demonstrated the potential of FPs (CBP-05, PPP-01) and FMs (PPM-01, ABM-04) to inhibit the

Global S-sulfenylation proteome analysis reveals a role of Fd-GOGAT in regulating GABA accumulation during Magnaporthe oryzae infection in rice.

Blast disease, caused by Magnaporthe oryzae, triggers complex redox signaling networks in rice involving reactive oxygen species (ROS) and reactive nitrogen species (RNS). These signaling molecules not only function as key regulators of plant immunity but also induce distinct cysteine-based post-translational modifications, namely S-sulfenylation and S-nitrosation, respectively, thereby modulating the activity, stability, and function of target proteins. While the role of S-nitrosation in rice immunity has been increasingly recognized, the contribution of S-sulfenylation remains largely unexpl

Toward in planta studies of persistent fungal viruses in a model plant.

A model plant, Nicotiana benthamiana, was examined as a host for persistent fungal viruses capable of crossing organismal kingdoms. Protoplasts of N. benthamiana were transfected with a mixture of virions of a betapartitivirus, Rosellinia necatrix partitivirus 18 (RnPV18), and an alphapartitivirus, RnPV19, and were then subjected to plantlet regeneration. Primary RT-PCR-based screening showed that nearly 100% of the resulting calli tested positive for RnPV18, whereas approximately 90% were positive for RnPV19. However, secondary screening performed at a later stage of tissue culture revealed t

Analysis of Secreted Proteins from Macrophomina phaseolina Isolated from Infected Glycine max Tissues.

Necrotrophic fungi such as Macrophomina phaseolina (MP) pose a growing threat to global food security, especially as climate change expands their ecological range. MP is a highly adaptable phytopathogen that secretes a diverse array of metabolites and proteins-collectively known as the secretome-which play critical roles in host colonization and disease progression. In this chapter, we describe a method to evaluate the secretome of MP stimulated with ground Glycine max roots and seedlings, to identify proteins potentially implicated in the infection process. Current methods for analyzing prote

Antibiotic resistance genes detected in lichens: insights from Cladonia stellaris.

BACKGROUND AND AIMS: Antibiotics are natural compounds produced by microorganisms that have long existed in ecosystems. However, the widespread clinical and agricultural use of antibiotics has intensified selective pressures on bacteria, leading to the proliferation of antibiotic resistance genes (ARGs). The increasing prevalence of these genetic elements in clinical and environmental settings now poses a major global health threat. While ARGs are well documented in anthropogenically influenced environments, their distribution and origins in remote ecosystems, such as boreal forests, remain po

Molecular insights into the antifungal activity of Bacillus velezensis JLU-53 against Cochliobolus heterostrophus in maize.

Maize (Zea mays L.), a globally cultivated staple grain and economically significant crop, is highly susceptible to Southern corn leaf blight (SCLB), a disease caused by Cochliobolus heterostrophus. This pathogen can lead to substantial losses in both yield and grain quality during the growing season. In this study, Bacillus velezensis strain JLU-53 was identified as a promising biocontrol agent against C. heterostrophus. JLU-53 exhibited broad-spectrum antifungal activity against several phytopathogens, including C. heterostrophus, Setosphaeria turcica, Rhizoctonia solani, Fusarium graminearu

Integrin ß4 is a receptor for emerging fungal pathogens from the genera Lomentospora and Scedosporium.

Lomentospora prolificans is an environmental fungus that can cause life-threatening infections when airborne conidia are inhaled. Airway epithelial cells are likely to be the first host cells to interact with L. prolificans during pulmonary infection; however, the fungal and host factors that govern this interaction are completely unknown. Herein, we combined whole fungal cell pulldowns of surface proteins from airway epithelial cells and liquid chromatography-mass spectrometry (LC-MS) to identify host proteins that could potentially serve as host receptors for the fungus. We provide evidence
