Suppression of Fusarium verticillioides in maize by Bacillus subtilis and Micrococcus luteus: evidence from biocontrol traits and GC-MS analysis.

A total of 204 rhizobacterial isolates from 39 maize rhizospheric soil samples were screened for antagonism against Fusarium verticillioides. Fifty-six isolates (27%) inhibited fungal growth in dual culture assays, with 27 cultures showing strong inhibition (>35%). The bacterial isolates were further evaluated for biocontrol related attributes, including ammonia, hydrogen cyanide, chitinase, β-1,4-glucanase, protease, amylase, and lipase production, along with plant growth-promoting traits such as phosphate, zinc, and iron solubilization and the production of indole-3-acetic acid (IAA) and gib

Improving nutritional quality and aflatoxin detoxification of peanut meal by co-fermentation with Weizmannia coagulans, Bacillus subtilis, and supplemented enzymes.

BACKGROUND: Peanut meal, a high-protein agricultural by-product, faces challenges as animal feed due to anti-nutritional factors, poor protein digestibility, aflatoxin contamination, and imbalanced amino acids. Microbial fermentation is one of the most effective methods to reduce anti-nutritional factors and enhance the nutritional value of peanut meal. Compared to single-strain fermentation, microbial-enzyme co-fermentation exhibits enhanced degradation efficiency, accelerates nutrient release, improves product safety by reducing mycotoxins and anti-nutritional factors, enhances sensory prope

Anaerobic fermentation of soybean meal by Bacillus subtilis ED-3-7 and its effect on the intestinal microbial community of chicken.

A strain named ED-3-7 with a high protease-producing ability was screened in a previous study. This strain can be used for the anaerobic fermentation of soybean meal (SBM) to degrade macromolecular antigen proteins and antinutritional factors. We here evaluated the nutritional quality of the anaerobic fermented SBM and its effects on the chicken intestinal microbial community. Crude protein and acid-soluble protein contents increased by 11.68% and 342.61%, glycinin and β-conglycinin decreased by 82.04% and 88.42%, urease content decreased by 90.10%, and the trypsin inhibitor content was lower

Investigating the Molecular Interactions of Quinoline Derivatives for Antibacterial Activity Against Bacillus subtilis: Computational Biology and In Vitro Study Interpretations.

Bacterial infections are evolving and one of the chief problems is emergence and prevalence of antibacterial resistance. Moreover, certain strains of Bacillus subtilis have become resistant to several antibiotics. To counteract this menace, the present work aimed to comprehend the antibacterial activity of synthesized two quinoline derivatives against Bacillus subtilis. Toxicity predictions via Protox II, SwissADME and T.E.S.T (Toxicity Estimation Software Tool) revealed that these derivatives were non-toxic and had little to no adverse effects. Molecular docking studies carried out in Schrodi

Effects of pH and Osmotic Changes on the Metabolic Expressions of Bacillus subtilis Strain 168 in Metabolite Pathways including Leucine Metabolism.

Bacillus subtilis is often exposed to diverse culture conditions with the aim of improving hygiene or food quality. This can lead to changes in the volatile metabolite profiles related to the quality of fermented foods. To comprehensively interpret the associated metabolic expressions, changes in intracellular primary and extracellular secondary volatile metabolites were investigated by exposing B. subtilis to an alkaline pH (BP, pH 8.0) and a high salt concentration (BS, 1 M). In particular, B. subtilis was cultured in a leucine-enriched medium to investigate the formation of leucine-derived

Development of Methods for Specific Capture of Biological Targets on Aluminum Substrates: Application to Bacillus subtilis Spore Detection as a Model for Anthrax.

Many (if not most) biosensors rely on functional silane coatings as a first step toward covalent immobilization of specific capture molecules. While methods for silanization of silica (SiO2) surfaces are very well developed, less has been done to develop and characterize silanization methods for alternative substrates, such as alumina (Al2O3). In particular, the behavior of Al2O3 coatings grown on aluminum under ambient conditions has not been studied. To address this issue, we have tested solution-phase deposition of two silanes on Al2O3 (3-aminopropyl triethoxysilane and 3-triethoxysilyl)pro

The exo-β-N-acetylmuramidase NamZ from Bacillus subtilis is the founding member of a family of exo-lytic peptidoglycan hexosaminidases.

Endo-β-N-acetylmuramidases, commonly known as lysozymes, are well-characterized antimicrobial enzymes that catalyze an endo-lytic cleavage of peptidoglycan; i.e., they hydrolyze the β-1,4-glycosidic bonds connecting N-acetylmuramic acid (MurNAc) and N-acetylglucosamine (GlcNAc). In contrast, little is known about exo-β-N-acetylmuramidases, which catalyze an exo-lytic cleavage of β-1,4-MurNAc entities from the non-reducing ends of peptidoglycan chains. Such an enzyme was identified earlier in the bacterium Bacillus subtilis, but the corresponding gene has remained unknown so far. We now report

An active β-lactamase is a part of an orchestrated cell wall stress resistance network of Bacillus subtilis and related rhizosphere species.

A hallmark of the Gram-positive bacteria, such as the soil-dwelling bacterium Bacillus subtilis, is their cell wall. Here, we report that d-leucine and flavomycin, biofilm inhibitors targeting the cell wall, activate the β-lactamase PenP. This β-lactamase contributes to ampicillin resistance in B. subtilis under all conditions tested. In contrast, both Spo0A, a master regulator of nutritional stress, and the general cell wall stress response, differentially contribute to β-lactam resistance under different conditions. To test whether β-lactam resistance and β-lactamase genes are widespread in

Effect of Bacillus Subtilis-based Direct-fed Microbials on Immune Status in Broiler Chickens Raised on Fresh or Used Litter.

Type of dietary direct-fed microbials (DFMs) or poultry litter could directly influence the composition of gut microbiota. Gut microbiota plays an important role in shaping the developing immune system and maintaining the homeostasis of the mature immune system in mammal and chickens. The present study was carried out to investigate the interaction among litter, DFMs and immunity in broiler chickens exposed to a field-simulated environment. Immune status of broiler chickens was assessed by serum antibodies against Eimeria spp. and Clostridium spp. and intestinal cytokine mRNA expression. The c

Lipid Compositions in Escherichia coli and Bacillus subtilis During Growth as Determined by MALDI-TOF and TOF/TOF Mass Spectrometry.

Lipids in Escherichia coli and Bacillus subtilis were analyzed by matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry and TOF/TOF tandem mass spectrometry. Lipids were extracted from bacterial cells using an equal volume mixture of dichloromethane, ethanol, and water, which formed a biphasic system with the lipids in the organic layer. The resulting mass spectra of the extracts from both bacteria showed a series of peaks corresponding to sodiated phospholipids - primarily phosphatidylethanolamines (PE) and phosphatidylglycerols (PG). The relative amounts of

Two evolutionarily closely related ABC transporters mediate the uptake of choline for synthesis of the osmoprotectant glycine betaine in Bacillus subtilis.

Biosynthesis of the compatible solute glycine betaine in Bacillus subtilis confers a considerable degree of osmotic tolerance and proceeds via a two-step oxidation process of choline, with glycine betaine aldehyde as the intermediate. We have exploited the sensitivity of B. subtilis strains defective in glycine betaine production against glycine betaine aldehyde to select for mutants resistant to this toxic intermediate. These strains were also defective in choline uptake, and genetic analysis proved that two mutations affecting different genetic loci (opuB and opuC) were required for these ph

Alkyl hydroperoxide reductase, catalase, MrgA, and superoxide dismutase are not involved in resistance of Bacillus subtilis spores to heat or oxidizing agents.

Only a single superoxide dismutase (SodA) was detected in Bacillus subtilis, and growing cells of a sodA mutant exhibited paraquat sensitivity as well as a growth defect and reduced survival at an elevated temperature. However, the sodA mutation had no effect on the heat or hydrogen peroxide resistance of wild-type spores or spores lacking the two major DNA protective alpha/beta-type small, acid-soluble, spore proteins (termed alpha(-)beta(-) spores). Spores also had only a single catalase (KatX), as the two catalases found in growing cells (KatA and KatB) were absent. While a katA mutation gr
