Despite extensive research, the regulation of mitochondrial function is still not understood completely. Ample evidence shows that cytosolic Ca2+ has a strategic task in co-ordinating the cellular work load and the regeneration of ATP by mitochondria. Currently, the paradigmatic view is that Cacyt2+ taken up by the Ca2+ uniporter activates the matrix enzymes pyruvate dehydrogenase, alpha-ketoglutarate dehydrogenase and isocitrate dehydrogenase. However, we have recently found that Ca2+ regulates the glutamate-dependent state 3 respiration by the supply of glutamate to mitochondria via aralar,

Homeostatic adaptation to maternal calcium metabolism is a prerequisite for optimal delivery of sufficient calcium to the fetus and neonate during pregnancy and lactation, respectively. This article outlines the major adaptations known to occur and the physiological regulators likely to be principally involved. Importantly, different adaptive responses are used in pregnancy and lactation. The rarity of calcium disorders in pregnancy underscores the successful implementation of these adaptations in most women. For those few women with either pre-existing or pregnancy-acquired disorders of calci…

Non-invasive diagnostic techniques such as electron beam computed tomography and multislice spiral computed tomography are able to detect and quantify coronary calcifications. Several clinical studies have shown how the amount of coronary calcifications correlates to the coronary plaque burden. The detection of coronary calcium therefore provides a unique opportunity to identify and quantify coronary atherosclerosis in a subclinical stage. Measures of subclinical atherosclerosis may also help in recognizing factors related to atherosclerosis in asymptomatic populations. In addition, a signific…

BACKGROUND: Dietary potassium intake (K) lowers urinary calcium (Ca) excretion and, in short-term studies, may improve Ca balance.
PURPOSE: Our objective was to assess K effects on the Ca economy under steady-state conditions.
DESIGN: 8-day, inpatient metabolic studies of nitrogen, phosphorus, and Ca balance, combined with dual isotopic Ca tracer kinetics studies. Study diet matched to prestudy nutrient intakes.
SUBJECTS: 191 single women studied from 1-5 times at approximately 5-year intervals, for a total of 644 inpatient studies. Median age at time of study: 50.2 yrs; 301 studies were pe…

The causes of Darier disease (DD) and Hailey-Hailey disease (HHD) have eluded clinicians and scientists for more than 60 years. DD is characterized by loss of adhesion between suprabasal epidermal cells associated with abnormal keratinization, while loss of epidermal cell-to-cell adhesion is predominant in HHD. The genes for both conditions have recently been identified using candidate positional cloning approaches. The gene for DD (ATP2A2) encodes a calcium transport ATPase of the sarco (endo)plasmic reticulum (SERCA2) Verboomen et al. [1992: Biochem J 286(Pt 2):591-595], while the gene for H…

A condition of unbalanced minerals was found in soil and drinking water from three amyotrophic lateral sclerosis (ALS) foci on Guam, in the Kii Peninsula and in West New Guinea with a low concentration of calcium and magnesium coupled with a high concentration of aluminum and manganese. The current epidemiological studies in the Western Pacific including the Kii Peninsula of Japan, have suggested that environmental factors contribute to the pathogenetic process of ALS and parkinsonism-dementia (PD). Six Kii cases with ALS showed higher Ca and lower Mg contents in the central nervous system (CN…

The recent cloning of a G-protein-coupled, extracellular calcium [(Ca2+)e]-sensing receptor (CaRG) from the parathyroid, kidney and brain of several species has clarified the molecular mechanisms underlying Ca2+-sensing by parathyroid and other cell types. It has long been suspected that such a receptor existed on parathyroid cells, coupled to intracellular second messengers through guanine nucleotide regulatory (G) protein which is able to recognize and respond to (Ca2+)e. Recently, functional screening of a cDNA library constructed from bovine parathyroid mRNA led to the isolation of a 5.3-k…

The human calcium-sensing receptor (CaSR) is a 1078-amino-acid cell surface protein which is expressed in the parathyroids, thyroid cells and the kidney, and is a member of the family of G protein-coupled receptors. The CaSR allows regulation of parathyroid hormone (PTH) secretion and renal tubular calcium reabsorption in response to alterations in extracellular calcium concentrations. The human CaSR gene is located on chromosome 3q13.3-q21, and loss of function CaSR mutations have been reported in the hypercalcaemic disorders of familial benign (hypocalciuric) hypercalcaemia (FBH or FHH) and …

Postmenopausal bone loss is associated with a rise in the fasting urine calcium excretion which has been proposed to be a possible cause of increased bone resorption. The oophorectomized rat is widely used as a model of postmenopausal bone loss, but preliminary data suggest that urine calcium excretion is not increased following oophorectomy in this animal model. However the present detailed experimental protocol demonstrated that oophorectomy in adult rats increased the obligatory urine calcium excretion compared with ovary-intact rats (P < 0.001). Importantly urine hydroxyproline excretion w…

The calcium antagonists are effective in the treatment of essential hypertension. This quality, in addition to their ability to inhibit atherosclerotic lesion formation, may make them superior to other classes of antihypertensive agents in preventing vascular complications of mild to moderate hypertension. Preliminary results of clinical trials suggest that the calcium antagonists retard the progression of early atherosclerotic lesions in native coronary arteries and saphenous vein grafts, but have less effect on established lesions. Further study is needed to determine whether these agents ha…

Calcium deficiency causes osteoporosis in experimental animals because the skeleton is sacrificed for the preservation of the plasma (ionic) calcium and to meet obligatory calcium losses in the feces and urine. (Vitamin D deficiency, on the other hand, causes rickets and osteomalacia largely because of the loss of the calcemic action of vitamin D, which leads to hypocalcemia, secondary hyperparathyroidism, and hypophosphatemia.) The concept that human osteoporosis, particularly in postmenopausal women, results from negative calcium balance represents a working hypothesis that fits many, but no…

Serum calcium in humans is closely regulated, with a maximum excursion of values throughout the day of less than 6%. A number of hormonal controls serve to maintain the stability of the serum calcium value. The three most important hormones--PTH, 1,25-dihydroxyvitamin D, and calcitonin--act chiefly on three organs--intestine, bone, and kidney. A set of simple diagrams that summarize these control mechanisms is presented. Organ diagrams are used to indicate the normal fluxes of calcium (or other ions) and the sites of hormone action, while a feedback loop diagram summarizes the interactions amo…

In five patients with Guillain-Barré syndrome, clinical myokymia increased and myokymic burst amplification occurred when ionized Ca++ was lowered by hyperventilation. Myokymia decreased when ionized Ca++ was increased after IV infusion of CaCl2. These responses were absent or diminished in the four patients with myokymia due to brainstem lesions, suggesting that the blood-brain barrier impedes the effects of altered serum ionized Ca++ on axonal excitability. Altering serum Ca++ can distinguish peripheral and central myokymia.

The role of calcium in the metabolism of carbohydrates is rather complex. It has been shown particularly "in vitro" but also "in vivo", to act on beta cells and peripheral effectors. Cytosolic calcium is determinant for insulin secretion in beta cells. It may be increased by a transfer of extracellular calcium into the intra cellular medium or by a transfer of cytoplasmic organelles into the cytosol. The former is due to the action of glucose or its metabolites. The latter is dependent on the CAMP, probably modulated by the glucose or its metabolites. The action of calcium causes the calmoduli…
