High Normal dietary casein attenuates bisphenol A-induced multiorgan toxicity in rats: Preservation of renal integrity, glucose homeostasis, and immune surveillance via modulation of oxidative stress, inflammation, and apoptosis.

Bisphenol A (BPA) causes multiorgan toxicity via oxidative stress, inflammation, and apoptosis, and the effect of nutritionally relevant elevation of casein intake is unknown. To evaluate whether a nutritionally relevant elevation of dietary casein (18% vs. 14%) attenuates BPA-induced multiorgan toxicity in rats, male Wistar rats were divided into four groups (n = 8/group) and treated for 35 days: control (14% casein), casein 18% alone, BPA (50 mg/kg/day +14% casein), and BPA + 18% casein. We measured oxidative and inflammatory markers, renal function, metabolic parameters, hematological param

Effects of intramammary infusions of casein hydrolysate, ethylene glycol-bis(β-aminoethyl ether)-N,N,N',N'-tetraacetic acid, and lactose at drying-off on mammary gland involution.

The transition from the lactation to the dry period in dairy cows is a period of high risk for acquiring new intramammary infections. This risk is reduced when involution of mammary glands is completed. Consequently, strategies that accelerate the involution process after drying-off could reduce the incidence of mastitis. The objective of this study was to assess the effect of 3 different treatments on mammary gland involution. Each quarter of 8 Holstein cows in late lactation was randomly assigned at drying-off to an intramammary infusion of casein hydrolysate (CNH; 70 mg), ethylene glycol-bi

The presence of the casein kinase II phosphorylation sites of Vpu enhances the CD4(+) T cell loss caused by the simian-human immunodeficiency virus SHIV(KU-lbMC33) in pig-tailed macaques.

The simian-human immunodeficiency virus (SHIV)/ macaque model for human immunodeficiency virus type 1 has become a useful tool to assess the role of Vpu in lentivirus pathogenesis. In this report, we have mutated the two phosphorylated serine residues of the HIV-1 Vpu to glycine residues and have reconstructed a SHIV expressing this nonphosphorylated Vpu (SHIV(S52,56G)). Expression studies revealed that this protein was localized to the same intracellular compartment as wild-type Vpu. To determine if this virus was pathogenic, four pig-tailed macaques were inoculated with SHIV(S52,56G) and vir

Delayed bacterial colonization of the gut alters the host immune response to oral sensitization against cow's milk proteins.

SCOPE: Cow's milk allergy is the most prevalent food allergy in infants whose immune system development is critically stimulated during postnatal gut colonization by commensal bacteria. Allergenic potential of cow's milk β-lactoglobulin (BLG) and caseins (CAS) was investigated in germ-free (GF) BALB/c mice and in GF mice conventionalized (CVd) at 6 weeks of age. METHODS AND RESULTS: Oral sensitization to cow's milk in the presence of cholera toxin led to higher BLG-specific IgE, IgG1, and IgG2a responses in GF mice than in conventional (CV) mice. No significant difference was observed for CAS…

Maternal malnutrition in the rat: effects on food intake and body weight.

The effects of dietary protein level on food intake and body weight were examined in adult female rats during a 35-day pre-mating period and during gestation and lactation. During the pre-mating period, no differences in daily food intake were observed among rats fed a 6% casein, 8% casein or 25% casein diet. However, during this period, rats fed the 6% casein diet gained significantly less weight than those with ad lib access to the 8% or 25% casein diets or than rats pair-fed the 25% casein diet in amounts equivalent to that consumed by rats in the 6% or 8% casein groups. Additionally, rats

Lactase-Treated A2 Milk as a Feasible Conventional Milk Alternative: Results of a Randomized Controlled Crossover Trial to Assess Tolerance, Gastrointestinal Distress, and Preference for Milks Varying in Casein Types and Lactose Content.

Background: Previous research indicates that gastrointestinal discomfort from milk consumption may be attributable to A1 β-casein, rather than lactose intolerance alone. A2 milk (free of A1 β-casein) consumption may result in fewer symptoms compared to conventional milk containing both A1/A2 β-casein. Objective: In this five-week, double-blind, double-crossover study, we assessed the physiological responses to doses escalating in volume of lactose-free conventional milk (Lactaid), A2 milk, and lactose-free A2 milk in fluid milk-avoiding participants. Methods: Each milk type was consumed over t

Determination of secretory IgA secretion and its contribution to total endogenous amino acid losses in broiler chickens fed a nitrogen-free diet or diets containing either casein or purified amino acids.

Secretory IgA (sIgA) secretion and its contributions to total endogenous amino acid (AA) losses were determined in broiler chickens. In Experiment 1, dietary treatments consisted of a nitrogen-free diet or a diet that contained 10% casein; both diets were provided to birds ad libitum. In Experiment 2, broilers received a nitrogen-free diet or diets containing 2.5, 5.0, 7.5, and 10% casein ad libitum. An additional diet was fed that contained a purified AA mixture that provided the same AA as the 10% casein diet. Another set of birds also received the 10% casein diet except birds were pair-fed

Subunit specific altered expression and activity of casein kinase 2 in the brain tissues resected from mesial temporal lobe epilepsy with hippocampal sclerosis patients & rodent temporal lobe epilepsy model.

INTRODUCTION: Mesial temporal lobe epilepsy (MTLE), is associated with dysregulated excitatory-inhibitory balance in the brain. Numerous enzymes, protein kinases, that are modulated through phosphorylation, have been linked with key processes involved in the pathogenesis of epilepsy. Therefore, in this study, we determined the subunit specific expression and activity of multi-subunit casein Kinase 2 (CK2) which influences NMDARs through phosphorylation events, in MTS patients as well as pilocarpine model of TLE. METHODS: mRNA expression of CK2 (α, α', β) & NR2B was measured by real time PCR a

A method for improving the nutritional value of food proteins: covalent attachment of amino acids.

Casein was modified by use of a series of active N-hydroxy-succinimide esters of amino acids in order to study the effects of new covalently linked hydrophobic or hydrophilic groups on its physical and nutritional properties. Tryptophan was used to determine the best conditions for the chemical reaction and to study the stability of the newly formed amide linkage (isopeptide bond). Casein was also modified with glycine, alanine, methionine, N-acetyl-methionine and aspartic acid. In vitro hydrolysis studies using bovine chymotrypsin, pancreatine and rat bile-pancreatic juice indicated that dige

Curcumin Induces Macrophages Polarization to Alleviate Ulcerative Colitis by Regulating Macrophage Nuclear Morphology via Affecting the Interaction Between Enteric Glial Cells Glial-Derived Neurotrophic Factor and Casein Kinase 2.

Macrophages exhibit dynamic functional phenotype shifts under diverse influences, releasing either anti-inflammatory or pro-inflammatory cytokines and playing a pivotal role in ulcerative colitis (UC) pathogenesis. In recent years, the role of enteric glial cells (EGCs) in maintaining intestinal homeostasis has been increasingly recognized. In this study, we investigated whether the alleviation of UC by curcumin is associated with the regulation of EGCs-macrophage interactions, thereby affecting macrophage polarization and its specific mechanisms. Through animal experiments and flow cytometry,

Comparative effects of pea protein vs. dairy proteins (α-lactalbumin and casein) on aminoacidemia, exercise performance, gut microbiota, hormonal responses, neurocognitive function, mood, and metabolic-inflammatory responses in older adults.

This narrative review comprehensively contrasts the physiological impacts of plant-based protein (focusing on pea protein isolates and matrices) vs. dairy-derived proteins (including intact casein and whey protein variants, emphasizing the rapid-digesting α-lactalbumin fraction). We establish a structured framework to separately report evidence from preclinical animal models and clinical human trials, evaluating acute (< 24 h) and chronic (>7 days) horizons across three domains: (i) neurocognitive function/mood, (ii) gut microbiota/hormonal responses, and (iii) aminoacidemia/metabolic-inflamma

Quantitative Structure-Activity Relationship Modeling Coupled with Molecular Docking Analysis in Screening of Angiotensin I-Converting Enzyme Inhibitory Peptides from Qula Casein Hydrolysates Obtained by Two-Enzyme Combination Hydrolysis.

In this study, Qula casein derived from yak milk casein was hydrolyzed using a two-enzyme combination approach, and high angiotensin I-converting enzyme (ACE) inhibitory activity peptides were screened by quantitative structure-activity relationship (QSAR) modeling integrated with molecular docking analysis. Hydrolysates (<3 kDa) derived from combinations of thermolysin + alcalase and thermolysin + proteinase K demonstrated high ACE inhibitory activities. Peptide sequences in hydrolysates derived from these two combinations were identified by liquid chromatography-tandem mass spectrometry (LC-

Animal models for small-for-gestational-age (SGA) neonates and infants-at-risk (IAR).

In this study, the body weight changes seen in rat dams and brain and body weights seen in pups as sequelae of either an overt or a hidden form of chronic protein deprivation have been examined. In the overt model, the 6% casein-diet pups showed SGA brain and body weight deficits and irreversible central and peripheral chemical changes at birth. In contrast, the hidden form of prenatal deprivation did not result in brain and body weight deficits in the 8% casein-diet pups at birth, but their SGA-like irreversible peripheral and central chemical profiles categorizes them as IAR-neonates. Selective allergy to sheep's and goat's milk proteins.

BACKGROUND: Cow's milk proteins are amongst the most common causes of food allergy in infants, and caseins are probably the main allergens. The existence of a high degree of cross-reactivity between milk caseins from different animals has been reported. We describe a 2-year-old boy who experienced allergic reactions after eating and touching sheep's cheese, but who tolerated cow's milk and cow's milk dairy products. He had never ingested milk or milk derivatives from sheep or goat. METHODS: Skin prick tests were carried out using whey fractions of cow's milk, whole milk and casein from goat, …
