The PARP inhibitor olaparib induces significant killing of ATM-deficient lymphoid tumor cells in vitro and in vivo.

The Ataxia Telangiectasia Mutated (ATM) gene is frequently inactivated in lymphoid malignancies such as chronic lymphocytic leukemia (CLL), T-prolymphocytic leukemia (T-PLL), and mantle cell lymphoma (MCL) and is associated with defective apoptosis in response to alkylating agents and purine analogues. ATM mutant cells exhibit impaired DNA double strand break repair. Poly (ADP-ribose) polymerase (PARP) inhibition that imposes the requirement for DNA double strand break repair should selectively sensitize ATM-deficient tumor cells to killing. We investigated in vitro sensitivity to the poly (AD

Misidentified LO2 and SGC-7901 cell lines challenge the claimed selectivity of ruthenium(II) polypyridyl complexes.

In this commentary, I express concerns about the misidentification of the LO2 and SGC-7901 cell lines used by Tian and his colleagues in their study of ruthenium(II) polypyridyl complexes with alleged anti-cancer properties. While these compounds appear to inhibit tumor growth by inducing cell cycle arrest, mitochondrial damage, apoptosis, ferroptosis, autophagy, and immunogenic cell death, the authors' claims of selectivity are undermined by their reliance on the LO2 line as a "normal liver cell". It is well established that the LO2 line is a HeLa derivative, rather than a benign hepatocyte c

The role of cell adhesion and migration in the in vitro invasiveness of mouse adrenal carcinoma cells.

Using the Matrigel invasion assay, we have examined the role of cell adhesion and migration in the invasiveness of two cell lines, DM and HSR, derived from the Y1 mouse adrenocortical tumor. The DM cells were metastatic and more invasive (10-fold) than the nonmetastatic HSR cells. The difference in invasiveness could not be ascribed to different levels of secreted type IV collagenolytic activity since HSR cells secreted higher levels of activity. Cells from the metastatic DM line showed greater motility to both laminin and fibronectin when compared to the HSR line. Furthermore, both Matrigel a

Cross-species oncolytic virotherapy in vitro: BoHV4EGFPΔTK as a novel candidate against canine osteosarcoma and histiocytic sarcoma.

Canine osteosarcoma (cOSA) and histiocytic sarcoma (cHS) are aggressive tumors characterized by poor prognoses and limited therapeutic options. Oncolytic viruses represent a promising treatment option for these tumors. Based on this hypothesis, this study aimed to evaluate direct and indirect oncolytic effects of Bovine Herpes virus type 1 (BoHV1) and a recombinant type 4 (BoHV4EGFPΔTK). Here we used cOSA, cHS and cutaneous fibroblast (FDOG) cell lines. BoHV1 and BoHV4EGFPΔTK can successfully infect cOSA and cHS cell lines with 1 multiplicity of infection (MOI). Notably, BoHV4EGFPΔTK displayed

[Burkitt's lymphoma with monoclonal pattern (IgG, kappa type) of cytoplasmic immunoglobulin--clinical report and review of Japanese literature].

A 52-year-old male was admitted to the hospital because of abdominal mass. Bone marrow examination revealed 26% blasts, which morphology was L3 in FAB criteria. Abdominal tumor was resected and histologic feature of the tumor was malignant lymphoma, small non-cleaved cell, Burkitt's. Lymphoma cells from the resected tumor were cultured and a cell line was established. Cytological studies of the original tumor cells and the cell line revealed that the lymphoma was negative both for EBNA and EBV DNA, and possessed t(8;14) (q24;q32) in chromosome analysis. Surface antigens were positive for HLA-D

Infrequent mutations of the TP53 gene and no amplification of the MDM2 gene in hepatoblastomas.

We have investigated the mutation of the TP53 gene in hepatoblastomas (HBLs) by using polymerase chain reaction-single strand conformation polymorphism and direct sequencing in 38 HBL tumor samples and in two HBL cell lines. We detected the TP53 gene mutation in an anaplastic hepatoblastoma cell line, but no aberration of the TP53 gene (exons 5-9) was found in tumor samples and in the other HBL cell line. The mutation of the cell line was a missense mutation from GAC (asparagine) to CAC (histidine) at codon 281, which was different from the G-to-T transversion of codon 249 that is frequently f

Evidence for an involvement of T4+ cytotoxic T cells in tumor immunity.

Cell-mediated immunity against cancer cells primarily involves class I major histocompatibility complex (MHC)-restricted cytotoxic T cells and natural killer (NK) cells. To investigate whether T4+ cytotoxic T cells also have a role in tumor-specific immunity, mice were immunized with a B cell lymphoma. T cell hybridomas were constructed from the immune spleen cells and analyzed for their cytotoxic ability against the immunizing lymphoma. A T4+, Lyt-1+ hybridoma cell line was developed (103L2) which specifically killed the immunizing tumor cells but not normal B cells or a range of other tumor

Patient-Derived Ovarian Cancer Spheroids Rely on PI3K-AKT Signaling Addiction for Cancer Stemness and Chemoresistance.

Ovarian cancer is the most lethal gynecological malignancy among women worldwide and is characterized by aggressiveness, cancer stemness, and frequent relapse due to resistance to platinum-based therapy. Ovarian cancer cells metastasize through ascites fluid as 3D spheroids which are more resistant to apoptosis and chemotherapeutic agents. However, the precise mechanism as an oncogenic addiction that makes 3D spheroids resistant to apoptosis and chemotherapeutic agents is not understood. To study the signaling addiction mechanism that occurs during cancer progression in patients, we developed

[Establishment and characterization of a human ureteral cancer cell line producing carbohydrate antigen 19-9 and carcinoembryonic antigen].

We established a new cell line (FU-UrC-1) derived from a human primary ureteral carcinoma xenografted in a nude mouse. This cell line exhibited epithelial characteristics and formed clusters in monolayer cultures. The cells were subcultured in vitro for more than 20 passages and had a doubling time of 53 hours. The modal number of chromosomes was 66. The cell line, which was xenografted again to nude mice, produced tumors essentially identical to the original tumor. Furthermore, the cultured cells expressed carbohydrate antigen 19-9 (CA19-9) and carcinoembryonic antigen (CEA) that were secrete

Cytopathological Study of the Circulating Tumor Cells filtered from the Cancer Patients' Blood using Hydrogel-based Cell Block Formation.

Circulating tumor cells have emerged as biomarkers for estimating the tumor burden and metastatic potential of cancer patients. However, to date, most of studies and applications of circulating tumor cells have been conducted and applied to epithelial cancers such as breast, colorectal, and prostate tumor. The only FDA-cleared method, CellSearch, makes use of antibody against epithelial surface protein expressed on CTCs, thus obstructing wide application for various cancers with non-epithelial and semi-epithelial characteristics including renal cell carcinoma. Due to rarity and ambiguity of CT

Rhabdoid tumors as a novel target for PSMA-directed CAR T cell therapy.

Rhabdoid tumor is an ultra-rare and highly aggressive pediatric malignancy with a poor prognosis and limited therapeutic options. To identify novel immunotherapeutic targets, transcriptomic data from the Cancer Cell Line Encyclopedia were analyzed, and we found that two rhabdoid tumor cell lines exhibit high expression of prostate-specific membrane antigen (PSMA), with levels comparable to well-established PSMA-positive prostate cancer cell lines. PSMA expression in rhabdoid tumors was subsequently validated in cell lines and in a subset of primary clinical rhabdoid tumor specimens. While PSMA

Sodium butyrate stimulates hCG and hCG-alpha production in a human choriocarcinoma cell line (BeWo).

To assess regulatory mechanism(s) of hormone production between ectopic and eutopic tumor cells, the effect of sodium butyrate on cell growth and hormone production was studied in BeWo, a trophoblastic cell line and in HeLa S3, a nontrophoblastic cell line. The addition of 2 mM sodium butyrate to HeLa S3 produced a marked increase in hCG-alpha both in the medium and in the cell extract, in contrast to its inhibitory effects on cell growth. Sodium butyrate had a similar effect on cell growth and hormone secretion in BeWo cells, as seen with an increase in release of hCG and hCG-alpha were also

Wogonin and related natural flavones overcome tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) protein resistance of tumors by down-regulation of c-FLIP protein and up-regulation of TRAIL receptor 2 expression.

Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) is a promising anticancer agent that kills various tumor cells without damaging normal tissues. However, many cancers remain resistant to TRAIL. To overcome TRAIL resistance, combination therapies using sensitizers of the TRAIL pathway would be an efficacious approach. To investigate potential sensitizers of TRAIL-induced apoptosis, we used TRAIL-resistant human T cell leukemia virus type 1 (HTLV-1)-associated adult T cell leukemia/lymphoma (ATL) cells as a model system. So far, HTLV-1-associated ATL is incurable by presently know

LASS2 enhances p53 protein stability and nuclear import to suppress liver cancer progression through interaction with MDM2/MDMX.

LASS2 functions as a tumor suppressor in hepatocellular carcinoma (HCC), the most common type of primary liver cancer, but the underlying mechanism of its action remains largely unknown. Moreover, details on its role and the downstream mechanisms in Cholangiocarcinoma (CCA) and hepatoblastoma (HB), are rarely reported. Herein, LASS2 overexpression was found to significantly inhibit proliferation, migration, invasion and induce apoptosis in hepatoma cells with wild-type (HB cell line HepG2) and mutated p53 (HCC cell line HCCLM3 and CCA cell line HuCCT1). Gene set enrichment analysis determined

Derivation of Genetically Defined Murine Hepatoblastoma Cell Lines with Angiogenic Potential.

Background/Objectives: Hepatoblastoma (HB), the most common pediatric liver cancer, often bears mutations in and/or otherwise deregulates the oncogenic transcription factors β-catenin (B), YAP (Y) and NRF2 (N). HB research is hampered by a paucity of established cell lines, particularly those possessing these molecular drivers. All combinations of B, Y and N (BY, BN, YN and BYN) are tumorigenic when overexpressed in murine livers, but it has not been possible to establish cell lines from primary tumors. Recently, we found that concurrent, in vivo Crispr-mediated targeting of the Cdkn2a tumor s

Mouse monoclonal antibody (FKH1) detecting human melanoma-associated antigens.

A mouse monoclonal antibody, FKH1, was produced to detect cytoplasmic melanoma-associated antigen. FKH1 was raised using cultured human melanoma cell line KHm-6 as an immunogen. Reactivity of this antibody was assessed by immunohistochemical techniques against cell lines and normal and neoplastic tissues. Positive reactions were seen against 5 human melanoma cell lines. It stained cytoplasm of melanoma cells in a diffuse and granular pattern in indirect immunofluorescence. Immunoelectron microscopy showed diffuse distribution of immunoreactant in the cytoplasm of KHm-1 cells excluding melanoso

Lymphokines modulate the growth and survival of thymic tumor cells containing a novel feline leukemia virus/Notch2 variant.

Tumorigenesis occurs through a multistep process initiated by genetic lesions and facilitated by endogenous and external growth/survival signals. In many malignancies, specific oncogenic mutations correlate with phenotypic characteristics, inferring lineage-specific pathogenic mechanisms. To characterize these relationships in a unique feline tumor, we studied primary cells and two-cell lines independently-derived from a thymic lymphoma that contained and actively expressed a novel feline leukemia virus (FeLV) recombinant with transduced host Notch2 sequences. All three tumor cell populations

Contribution of RIZ1 to regulation of proliferation and migration of a liver fluke-related cholangiocarcinoma cell.

PURPOSE: Retinoblastoma-interacting zinc finger gene (RIZ1) is a tumor suppressor gene which is highly inactivated by promoter hypermethylation in patients with liver fluke-related cholangiocarcinoma (CCA). Epigenetic aberration of this gene might withdraw the ability to restrain tumor cell proliferation and migration. We aimed to define the role of RIZ1 on cell proliferation and migration in CCA cell line. MATERIALS AND METHODS: Small interference RNA (siRNA) was used to knock down the expression of RIZ1 in a CCA-derived cell line in which cell proliferation and cell migration were performed

Impact of tumor necrosis factor-alpha and lysophosphatidic acid on the behavior of ovarian cancer cells in a three-dimensional collagen hydrogel.

BACKGROUND: Advanced ovarian cancer involves tumor cells proliferating within ascitic fluid, which is rich in chemokines, cytokines, growth factors, and phospholipids essential for tumor progression. Tumor necrosis factor-alpha (TNF-α) and lysophosphatidic acid (LPA) are critical components that modulate ovarian tumor cell behavior in vivo, though most in vitro studies have relied on cell monolayers that do not accurately represent the tumor microenvironment. We thus investigated TNF-α and LPA effects on ovarian tumor cells cultured in collagen scaffolds, which are a model more reflective of i
