Evaluating eight smoking metrics for modelling survival in non-small cell lung cancer.

INTRODUCTION: Smoking is a strong modifiable prognostic factor for lung cancer survival. We compared eight smoking metrics to determine which metric best models the relationship between smoking exposure with overall survival (OS) and lung cancer-specific survival (LCSS). These metrics included cigarettes-per-day, smoking duration, pack-years, square-root pack-years, logcig-years, the comprehensive smoking index, age-of-initiation, and years-since-quit. MATERIALS/METHODS: This retrospective, pooled analysis included 25 International Lung Cancer Consortium studies between June 1, 1983-December

UL45 of Human Cytomegalovirus Promotes Macrophage Survival and Modulates RIPK3-Associated Cell Death.

Programmed necrotic cell death (necroptosis) is a key antiviral defense pathway, yet its regulation during human cytomegalovirus (HCMV) infection remains incompletely defined in physiologically relevant cell types. The M45 protein of murine cytomegalovirus, a homologue of UL45, inhibits necroptosis and contributes to MCMV dissemination, providing a rationale for examining UL45 in HCMV. Previous studies of UL45 have largely relied only on fibroblasts or immortalized cell lines, which lack the necroptotic machinery necessary to reveal viral anti-necroptotic functions. To investigate the role of

Retinal ganglion cell survival and functional maturation in transiently vascularized human retinal organoids.

Retinal organoids are widely used to model human retinal development and disease, but their utility is limited by the absence of vascular networks and stable axonal projections, which contribute to retinal ganglion cell degeneration and loss of function. To address these challenges, we incorporated stem cell-derived endothelial cells to induce transient vascular-like networks and used microfluidic devices to stabilize axonal growth. The resulting organoids showed reduced hypoxia, increased size, and decreased apoptosis, indicating improved long-term survival and maturation of retinal ganglion

STAT3/STAT5 Mutations Predict Shorter Overall Survival in Patients with Plasma Cell Myeloma.

INTRODUCTION: Plasma cell myeloma is a heterogeneous hematologic malignancy characterized by clonal expansion of plasma cells. While RAS-MAPK pathway mutations are known high-risk features, the clinical relevance of STAT3/STAT5 mutations remains unclear. METHOD: We analyzed 16 myeloma patients with STAT3/STAT5 mutations identified by next-generation sequencing (9 STAT3, 5 STAT5A, 2 STAT5B) and compared them with 32 mutation-negative controls. RESULTS: STAT3/STAT5 mutations were present at initial diagnosis in all eight patients with available initial sequencing data, suggesting these are ear

Host-cell dependent epigenetic profiles associated with survival outcomes in T. gondii infection.

Toxoplasmosis is a widespread parasitic disease affecting roughly one-third of the global population. In immunocompromised individuals or during pregnancy, infection can result in severe complications. Following primary infection, Toxoplasma gondii forms dormant bradyzoite cysts in tissues such as the brain and eyes. These cysts can rupture, particularly in immunocompromised hosts, releasing active parasites and triggering recrudescence. Efforts to experimentally induce and study bradyzoite cyst recrudescence have been hindered by the limited capacity of cell culture adapted strains to form ti

The TENT5A-ATXN2 axis modulates germline and somatic cell survival during heat stress.

Stress conditions elicit the formation of different kinds of stress granules (SGs). Although male germ cells are particularly sensitive to heat stress, the composition of heat-induced SGs in the testis remains poorly characterized. Here, we show that ATXN2 is a main component of heat-induced SGs in male germ cells and identify a unique population of ATXN2-positive SGs in leptotene and zygotene spermatocytes lacking classical SG markers. The terminal nucleotidyltransferase 5A (TENT5A) is a cytoplasmic poly(A) polymerase that extends the poly(A) tails of mRNAs. Tent5a mRNA expression increases w

Survival outcomes and treatment patterns in primary vertebral diffuse large B-cell lymphoma.

Primary vertebral diffuse large B-cell lymphoma (PV-DLBCL) is a rare manifestation of extranodal non-Hodgkin lymphoma that presents with nonspecific symptoms and poses significant diagnostic and treatment challenges. While chemotherapy, radiation, and surgery are frequently employed, their relative impacts on long-term survival remain unclear. Using the SEER database (2000–2021), we identified 584 adult patients with histologically confirmed PV-DLBCL. Demographic, tumor, and treatment characteristics were analyzed. Cox proportional hazards models were used to evaluate predictors of mortality,

Survival Trends in Metastatic Renal Cell Carcinoma Across Therapeutic Eras and Histologic Subtypes.

Introduction Immune checkpoint inhibitors have transformed the treatment of metastatic renal cell carcinoma (RCC). Whether these advances translated into population-level survival improvements across histologic subtypes remains uncertain. Materials and methods We performed a retrospective cohort study using the Surveillance, Epidemiology, and End Results (SEER) database (2006-2022). Patients older than 20 years with distant-stage RCC were categorized into a targeted therapy era (2006-2015) and an immunotherapy era (2016-2022). Overall survival (OS) and cancer-specific survival (CSS) were evalu

Circulating Sex Hormone Levels and Survival in Men With Esophageal Squamous Cell Carcinoma.

BACKGROUND: Men with esophageal squamous cell carcinoma (ESCC) have worse survival than women, a disparity in which sex hormones may play a role. Yet, whether circulating sex hormone levels are associated with survival in ESCC remains largely unexplored. METHODS: This prospective cohort study included 402 men with ESCC who underwent curative surgery in 2013-2020, with follow-up until May 1, 2023. Associations between preoperative levels of 11 sex hormone measures and all-cause mortality were assessed using multivariable Cox regression, reporting hazard ratios (HRs) and 95% confidence interval

T cell phenotype and clonality changes in myeloma patients with short overall survival.

Overall survival (OS) in multiple myeloma (MM) varies between a couple of months to more than 20 years, influenced by tumor characteristics, the tumor microenvironment (TME), and patient factors such as age and frailty. We analyzed sequential BM samples from 45 MM patients with OS less than 3 years versus more than 8 years using mass cytometry and bulk TCRβ sequencing. Patients with long OS demonstrated stability in the TME and T cell environments, while those with short OS had significant changes at relapse, including fewer T cells, increased Tregs, and more activated and exhausted CD8+ T cel

c-Jun regulates postpartum β-cell apoptosis and survival downstream of prolactin signaling.

OBJECTIVE: Pregnancy and postpartum states drive dynamic expansion and regression of maternal β-cell mass. Little is known about what regulates postpartum regression. We recently profiled murine islets at different time points from late gestation to early postpartum to identify regulators of β-cell apoptosis or survival. One hit was c-Jun, a transcription factor which regulates proliferation, apoptosis, and survival in various tissues and cells. Here, we examine c-Jun regulation and function during gestation and postpartum and in murine and human islets. METHODS: To examine regulation of c-Ju

Nanomedicine-enhanced SCAP hybrid spheroids: A novel approach for improved stem cell survival.

In regenerative medicine, the therapeutic potential of mesenchymal stem cells (MSC), such as stem cells from human apical papilla (SCAP), is well-documented and largely attributed to their secretome. However, their poor survival post-transplantation limits their efficacy. This study hypothesized that combining SCAP spheroids with nanomedicines loaded with NecroX-5 (an anti-necrotic drug) and rapamycin (an immunosuppressive agent) would enhance SCAP survival in vivo. The approach aimed to reduce oxidative stress-related cell death and suppress immune reactions towards xeno-/allogenic cells. Two
