Protein-centric omics analysis reveals circulating complements linked to non-viral liver diseases as potential therapeutic targets.

BACKGROUND/AIMS: To evaluate the causal correlation between complement components and non-viral liver diseases and their potential use as druggable targets. METHODS: We conducted Mendelian randomization (MR) to assess the causal role of circulating complements in the risk of non-viral liver diseases. A complement-centric protein interaction network was constructed to explore biological functions and identify potential therapeutic options. RESULTS: In the MR analysis, genetically predicted levels of complement C1q C chain (C1QC) were positively associated with the risk of autoimmune hepatitis

The burden of a brain tumor: guiding patient centric care in neuro-oncology.

PURPOSE: Brain tumor patients report an overwhelming sense of uncertainty when navigating the course of their terminal disease. Historically, organizational experts and/or treating physicians have established neuro-oncology programs. However, given the disease burden and incurable nature of current medical treatments, patient-centric care should be prioritized alongside institutional and academic objectives. Integrating patient perspectives into interdisciplinary programmatic development can improve comprehensive care and empower patients to advocate for their own quality healthcare needs. ME

[Health-related strength and power training in seniors: Purpose and recommendations].

The proportion of older people in western societies rapidly increases. Aging-induced disease conditions accompanied with declines in cardiocirculatory and neuromuscular performance constitute a major individual and economic health burden. Besides decreasing vascular and cardiac function during the process of aging, a loss of skeletal muscle mass, muscle structure and function seem to mainly account for decreasing maximal strength, strength development and strength endurance. These findings adversely interfer with static and dynamic postural control and may lead to an increased risk of falling

Revisiting the supraforaminal horizontal oblique osteotomy of the mandible.

PURPOSE: Today, the most common orthognathic procedure for correction of mandibular deformities is the bilateral sagittal split osteotomy, also called sagittal ramus osteotomy. Permanent injury to the mandibular nerve (V3) is one of the main complications, with a reported incidence between 5% and 30%. Orthognathic surgery using sagittal ramus osteotomy of the mandible as the procedure of choice should be re-evaluated because of the complexity and the relatively high risk of damage to the inferior alveolar nerve. Surgical techniques that allow for accurate condylar positioning with a lower risk

The relationship between malocclusion, benign joint hypermobility syndrome, condylar position and TMD symptoms.

The current study investigated the association between temporomandibular disorders, malocclusion patterns, benign joint hypermobility syndrome and the initial condylar position. One hundred sixty-two subjects were analyzed using the Rocabado Temporomandibular Pain Analysis; Helkimo Index parameters; the Carter-Wilkinson modified test; and a mounting cast with condylar position indicator registration (MPI). The study revealed a significant association between: 1. Delta H, skeletal pattern (p = 0.034); 2. Delta Y, transversal malocclusion (p = 0.04); 3. right and left, Delta Z, right and left po

Protein Language Models Identify Early Folding Residues as Critical Hotspots for Pathogenic Variants and Cancer Prognosis.

Identifying the specific residues that initiate protein folding is critical for understanding protein stability and the mechanisms of misfolding-related diseases. However, the systematic, proteome-wide identification of these early folding residues (EFRs) has been hampered by both experimental and computational limitations. Here, we present LMEFold, a deep learning framework using protein language models (PLMs) to accurately predict EFRs from amino acid sequence alone. LMEFold outperforms existing sequence-based predictors on the Start2Fold benchmark (ROC AUC = 0.831), and its predicted scores

Reprogramming pathological secretory phenotype of chondrocytes inhibits neurovascular invasion for osteoarthritis treatment.

Pain severity in osteoarthritis (OA) often dissociates from the extent of cartilage structural damage, indicating that non-structural mechanisms contribute to symptom persistence. Here, we identify articular chondrocytes as active orchestrators of pathological neurovascular invasion through a pathogenic secretome, rather than merely passive victims of degeneration. Integrated human single-cell transcriptomic analysis reveals that OA chondrocytes undergo phenotypic reprogramming characterized by coordinated activation of angiogenic and neurotrophic programs. Functionally, inflammatory chondrocy

The evolving role of artificial intelligence in ophthalmology: basic science, translation, and clinical integration.

PURPOSE OF REVIEW: Ophthalmology has emerged as the proving ground for medical artificial intelligence (AI) because of its imaging-centric workflows, standardized data acquisition, and well defined clinical endpoints. Recently, the field has started transitioning from narrowly trained, task-specific algorithms toward multimodal models that are trained on large datasets and can be adapted to many tasks. These models may integrate information from images, text, and clinical data and are increasingly trialed in real-world settings. This review examines recent advances and ongoing controversies in

The gut-liver axis in animal diseases: roles, mechanisms, and veterinary clinical applications.

The liver serves as the core organ for metabolism, detoxification, and immune regulation in animals. Its functional homeostasis directly determines the animal's health status and production performance. The gut-liver axis, as a critical inter-organ regulatory network connecting the intestine and the liver, plays a vital role in regulating immune responses, maintaining nutritional metabolism balance, and preventing pathogen invasion. The review systematically elucidates the central role of the gut-liver axis in regulating metabolic homeostasis and immune defense in animals. It comprehensively i

Beyond VEGF: AEG-1/MTDH as a Systems-Level Orchestrator of Angiogenesis in Hepatocellular Carcinoma.

Hepatocellular carcinoma (HCC) remains one of the leading causes of cancer-related mortality worldwide and is characterized by extensive vascularization, aggressive progression, and limited therapeutic responsiveness. Angiogenesis plays a central role in HCC development by supporting tumor growth, metabolic adaptation, invasion, and metastatic dissemination. Although anti-angiogenic therapies targeting the vascular endothelial growth factor (VEGF) pathway have improved clinical management, their overall survival benefit remains modest because of compensatory signaling, adaptive resistance, and

The Oncogenic EGFR-SHC1 Fusion Confers Insensitivity to EGFR-TKIs via Dual Activation of N-EGFR Kinase Domain and C-SHC1 Phosphorylation Sites in Lung Cancer.

UNLABELLED: Although epidermal growth factor receptor (EGFR) fusions in non-small cell lung cancer (NSCLC) typically show sensitivity to tyrosine kinase inhibitors (TKI), we identified an EGFR-SHC1 fusion subtype that exhibits intrinsic resistance to EGFR-TKI monotherapy through a dual-activation mechanism in the preclinical and clinical settings. EGFR-SHC1 fusion protein comprises of N-terminal EGFR and C-terminal SHC1. We demonstrated that EGFR-SHC1 simultaneously activates the EGFR kinase domain (KD) and SRC-mediated phosphorylation of the SHC1 fusion partner, thereby driving ERK/AKT pathwa

Flower-centric temperature control: Optimizing osmanthus black tea aroma via environmental temperature regulation of floral vitality and enzymatic responses.

The temperature parameter of scenting process, which governs the vitality of fresh flowers and the release of floral volatiles, is still empirically determined. Here, we investigate how environmental temperature regulates osmanthus vitality and enzymatic activity to shape the aroma quality of osmanthus black tea (OBT). Results showed that the moderate-temperature (25 °C and 30 °C) exhibited superior sensory quality, accompanied by slower water loss and delayed wilting of osmanthus, which favored aroma retention during scenting. Carotenoid cleavage dioxygenase (CCD) content peaked (1268 U/g), a

Chronic Disease Patients' Engagement in Interprofessional Telehealth Collaboration in Primary Care: A Scoping Review.

With the rise of people being affected with chronic illness, now the leading cause of mortality worldwide, primary care is overwhelmed with the demand for healthcare services. Primary healthcare is the first resource for patients living with chronic illness, but in 2019, COVID-19 brought healthcare professionals to increase services through virtual care for patients living with chronic illness. In the workplace, such professionals often need to be sufficiently resourced to collaborate, to address collaborative care barriers in telehealth and to keep patients engaged in their health. We perform

Introduction and methodology: Standards of Care in Overweight and Obesity-2025.

Obesity is a chronic, relapsing, and progressive disease requiring long-term, interprofessional treatment strategies to improve health outcomes. With over 40% of US adults and nearly 20% of children affected, obesity remains a significant public health concern. Despite the American Medical Association's recognition of obesity as a chronic disease, gaps persist in education, training, and access to effective treatments. These gaps contribute to inadequate obesity management and reinforce stigma and weight bias in healthcare settings. The Standards of Care in Overweight and Obesity-2025, develope
