Genetic susceptibility to breast cancer in women is conferred by a large number of genes, of which six have so far been identified. In the context of multiple-case families, BRCA1 and BRCA2 are the most important. Mutations in these genes confer high lifetime risks of breast cancer and ovarian cancer, and more moderate risks of prostate cancer and some other cancer types. Mutations in the CHEK2 and ATM genes, by contrast, cause much more modest (2-4 fold) risks of breast cancer. Genes so far identified explain approximately 20% of the familial aggregation of breast cancer. The remaining suscep

WEE1 is a key tyrosine kinase involved in the cell cycle regulation with potent anticancer effects in various cancer types including colorectal cancer. Recent studies have focused on the potential of combinational inhibition of Ataxia Telangiectasia and Rad-3-related protein (ATR) and WEE1 in increasing apoptosis in cancer cells. Therefore, this study investigates the effects of inhibiting WEE1, by employing AZD1775, on colorectal cancer cells' susceptibility to VE-822-induced DNA damage and apoptosis.SW-480 and HT-29 cells were treated with AZD1775 and VE-822, alone and in combination. BACKGROUND/AIM: While normal cells are highly regulated, cancer cells take a dysregulated path which bolsters their survival. Currently, a limited number of uniform treatments are available for cancer cure. Our goal was to deprive cancer cells of the key nutrient methionine and determine what effect it would have on cell death and alterations in DNA methylation of prostate cancer cells.
MATERIALS AND METHODS: PC3 and other cell lines were transfected with plasmid gene constructs for methionine gamma lyase deaminase (MEGL), a methionine-degrading enzyme, targeted for expression in the cytoplas

BACKGROUND & AIMS: Immune checkpoint inhibitors have shed light on the importance of antitumor immunity as a therapeutic strategy for hepatocellular carcinoma (HCC). The altered glucose metabolism known as the Warburg effect recently has gained attention as a cancer immune-resistance mechanism. Considering glycolysis inhibitors as therapeutic agents, their specific delivery to cancer cells is critical not to induce adverse effects. Thus, we investigated antitumor effects of a glycolysis inhibitor, consisting of 2-deoxy-D-glucose (2DG)-encapsulated poly(lactic-co-glycolic acid) (PLGA) nanoparti

Insulin-like growth factor binding protein 2 (IGFBP2) has emerged as a multifarious and context-dependent oncoprotein that links several mechanisms in the tumor microenvironment (TME) including oncogenic signaling, extracellular matrix (ECM) remodeling, immune evasion, and therapy resistance. Beyond its established role in modulating IGF signaling, IGFBP2 exerts IGF-independent effects via its RGD integrin-binding motif and nuclear localization signal (NLS). By binding integrins (αvβ3, α5β1), IGFBP2 activates focal adhesion kinase (FAK), which then triggers PI3K/AKT signaling and MAPK/ERK sign

BACKGROUND: The use of FMS-like tyrosine kinase 3 (FLT3) as a crucial target for kinase inhibitors is well established, but its association with immune infiltration remains unclear. This study aimed to explore the relationship between FLT3 mutations and immune checkpoint molecules (ICMs) in patients with acute myeloid leukemia (AML).
METHODS: The Cancer Genome Atlas (TCGA) and Genotype-Tissue Expression (GTEx) databases were used to identify the ICMs associated with FLT3 mutations. A Gene Ontology (GO) analysis, Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis, and gene set enrichment

INTRODUCTION: Immune checkpoint inhibitor (ICI) therapy has been found to increase the risk/severity of immune-mediated adverse events with subsequent kinase inhibitor treatment in oncogenically driven cancers. We explored the risk for hypersensitivity with selpercatinib, a first-in-class highly selective and potent, central nervous system-active RET inhibitor, in prior ICI-treated patients with RET fusion-positive NSCLC compared with their ICI-naive counterparts.
METHODS: Data from patients enrolled by December 16, 2019, in the ongoing phase 1/2 LIBRETTO-001 (NCT03157128) trial were analyzed

BACKGROUND: UCN-01 (7-hydroxy-staurosporine) is a novel antineoplastic agent targeting cyclin-dependent kinases, which shows potent in vitro and in vivo activity against a broad range of tumor types. Our group has previously shown that UCN-01 potentiates the apoptotic response of agents such as cisplatin in vitro by preventing sequence-specific abrogation of G2 arrest caused by DNA-damaging chemotherapies.
PATIENTS AND METHODS: This National Cancer Institute-sponsored phase I trial was designed to determine the safety, maximum tolerated dose, and pharmacokinetics of escalating doses of cispla

CONTEXT: Genetic testing for inherited mutations in BRCA1 and BRCA2 has become integral to the care of women with a severe family history of breast or ovarian cancer, but an unknown number of patients receive negative (ie, wild-type) results when they actually carry a pathogenic BRCA1 or BRCA2 mutation. Furthermore, other breast cancer genes generally are not evaluated.
OBJECTIVE: To determine the frequency and types of undetected cancer-predisposing mutations in BRCA1, BRCA2, CHEK2, TP53, and PTEN among patients with breast cancer from high-risk families with negative (wild-type) genetic tes

BACKGROUND: The need for new therapies to improve survival and outcomes in pediatric oncology along with the lack of approval and accessible clinical trials has led to "out-of-trial" use of innovative therapies. We conducted a retrospective analysis of requests for innovative anticancer therapy in Canadian pediatric oncology tertiary centers for patients less than 30 years old between 2013 and 2020.
METHODS: Innovative therapies were defined as cancer-directed drugs used (a) off-label, (b) unlicensed drugs being used outside the context of a clinical trial, or (c) approved drugs with limited

Programmed death-ligand 1 (PD-L1) has emerged as an important player in intrinsic tumor cell biology besides its canonical immune checkpoint function. Cumulative experimental data points to a role for PD-L1 in promoting and maintaining an invasive cell phenotype within different cancer cells through its interaction with mechanistic target of rapamycin (mTOR) signaling. In this review, we summarize the main experimental data pointing to the PD-L1/mTOR interaction in tumor cell invasion capacity both in vitro and in vivo. In this crosstalk, PD-L1 expression is induced by IFN-γ/STAT1 and IL-6/STA

Mycoplasma pneumoniae is a primary cause of community-acquired pneumonia, characterized by an excessive host inflammatory response that often drives lung pathology. While Toll-like receptor 2 (TLR2) is the principal sensor for this pathogen, the regulatory mechanisms that calibrate its signaling intensity remain incompletely understood. In this study, we delineate a novel disinhibition-amplification network controlling the macrophage response to M. pneumoniae. We demonstrate that TLR2 engagement triggers the secretion of Matrix metalloproteinase-9 (MMP-9), which subsequently activates the host

Mesothelioma is characterized by the inactivation of tumor suppressor genes, with frequent mutations in neurofibromin 2 (NF2), BRCA1-associated protein 1 (BAP1), and cyclin-dependent kinase inhibitor 2A (CDKN2A). These mutations lead to disruptions in the Hippo signaling pathway and histone methylation, thereby promoting tumor growth. NF2 mutations result in Merlin deficiency, leading to uncontrolled cell proliferation, whereas BAP1 mutations impair chromatin remodeling and hinder DNA damage repair. Emerging molecular targets in mesothelioma include mesothelin (MSLN), oxytocin receptor (OXTR),
