A method has been developed for the separation of leucine, 2-ketoisocaproic acid, isovaleryl CoA, 3-methylcrotonyl CoA, 3-hydroxy-3-methylglutaryl CoA, 3-methylglutaconyl CoA, acetyl CoA, and acetoacetic acid by ion-exchange high-performance liquid chromatography. The analysis requires 180 min. A rapid and simple method for the separation of trimethylselenonium ion and other cationic forms of selenium in urine by HPLC on a strong cation exchanger is described. Most of the inorganic salts in urine are removed prior to chromatography by means of ethanolic precipitation, thus minimizing interferences. This simplified determination of serotonin in human plasma involves "high-performance" liquid chromatography with electrochemical detection. Deproteinated samples of platelet-poor plasma are injected directly into the chromatograph without preliminary chromatographic purification. A high-performance liquid chromatographic (HPLC) assay for acetazolamide is presented. A 100-microliter sample is mixed with an aliquot of the internal standard solution and the mixture, buffered at pH 4.5, is extracted with ethyl acetate. The extract is evaporated to dryness and the residue is analyzed by HPLC, using a reverse-phase octadecylsilane column. The wavelength of the detection is 254 nm. We describe a "high-performance" liquid-chromatographic procedure for measuring amino acids in 0.5 mL of urine. The procedure includes direct derivatization of amino acids in urine samples with "dabsyl chloride" (4-dimethylaminoazobenzene-4'-sulfonyl chloride). An aliquot of this dabsylated amino acid solution is analyzed on a muBondapak C18 column with ethanol/sodium acetate (20 mmol/L, pH 4.0) 4/6 (by vol.), as mobile phase. We report a specific and sensitive quantitative assay for salicylate in serum or plasma. As little as 3 microliter of sample can be analyzed by an external standard procedure and larger volumes by diluting with the internal standard (o-methoxybenzoic acid). No extraction or derivatization is necessary and salicylic acid and the internal standard elute in 6 and 3.75 min, respectively. Peak-height ratios are linearly related to concentrations between 10 and 500 mg/liter. By comparison with an existing fluorometric method, our procedure is faster, more specific, and more sensitive.

We describe a gas-chromatographic micromethod, with use of a solid injection system and an electron capture detector, for determination of underivatized clonazepam in serum. Vigorous mixing of 100 microliter of serum with 20 microliter of borate buffer (pH 9.0) and 100 microliter of a cyclohexane/dichloromethane mixture (4/6 by vol) containing methyl nitrazepam as internal standard suffices to extract 86% of the clonazepam into the organic layer. We describe a method for measuring therapeutic and overdose levels of barbiturate, sedative, and anticonvulsant drugs in serum by gas-liquid chromatography (GLC). This method is a unique combination of techniques including a simple extraction, cochromatography of the specimen with authentic carrier drugs, and the use of multiple dissimilar gas chromatographic columns.
