Development of allele-specific PCR methodology (AS-PCR) to screening A. lumbricoides and A. suum.

Ascaris lumbricoides and Ascaris suum are described as helminths that infect humans and pigs, respectively. It is estimated that infection by A. lumbricoides affects about 447 million individuals living in tropical regions of developing countries. However, there is an increasing number of cases of human ascariasis in countries with no recent history of autochthonous infection by A. lumbricoides. In these places, pigs have been incriminated as the main source of human infection. Conventional parasitological diagnosis does not allow species-specific identification, and the real epidemiological s

[Microbial diagnosis with PCR will become clinically beneficial with a faster analysis].

PCR was introduced in 1985 by Mullis and was immediately recognized as a valuable tool in biomedical research and was awarded the Nobel Prize in 1993. Two culture-negative meningitis cases are described where Haemophilus influenzae and Neisseria meningitidis were found by 16SRNA-PCR. The modern real time PCR technology using fluorescent probes (hybridization probes, lightup probes, molecular beacons etc) for detection of the PCR-product or on DNA microarray chips, is under development for routine use. Multiplex technology can be used to simultaneously detect multiple microorganisms as well as

Genetic variation of the 8-kDa glycoprotein family from Echinococcus granulosus, Taenia multiceps and Taenia hydatigena.

BACKGROUND: Echinococcosis, coenurosis and cysticercosis are debilitating diseases which prevail in China. Immunological diagnosis of metacestodosis is important in disease control. The 8-kDa glycoproteins from taeniid cestodes have successfully been used for diagnosis of human cysticercosis in immunological assays. The aim of the present study was to investigate genetic variations and phylogenetic relationships of the 8-kDa proteins for evaluating the possibility of utilizing these proteins as diagnostic antigens for other metacestode infections. METHODS: The genes and complementary DNAs (cD

Differentiating Dracunculus medinensis from D. insignis, by the sequence analysis of the 18S rRNA gene.

This study, undertaken as a component of the global Dracunculiasis Eradication Program (DEP), was designed to provide molecular tools to distinguish Dracunculus medinensis, the nematode causing human dracunculiasis, from other tissue-dwelling nematodes, including other Dracunculus species that infect humans and other animals. DNA was extracted from D. medinensis and from a closely related species that infects North American carnivores, D. insignis, so that the genes coding for the small-subunit ribosomal RNA (18S rRNA) of the parasites could be amplified, sequenced and compared. Sequences were

Hydatid echinococcosis (Echinococcus granulosus) in a laboratory colony of pig-tailed macaques (Macaca nemestrina).

Several cases of hydatid echinococcosis were diagnosed in a laboratory colony of 19 pig-tailed macaques (Macaca nemestrina) at the Paul Ehrlich Institute, Germany. Three hydatid cysts were found in the liver of an euthanized animal. The diagnosis of an Echinococcus granulosus infection was confirmed by histopathology and the results of a specific polymerase chain reaction. The serum of five of 14 other monkeys tested for Echinococcus antibodies using a genus-specific enzyme-linked immunosorbent assay (ELISA) was positive or weakly positive; none of the animals, however, showed specific reactio

Ribonucleotide reductase as a therapeutic target for drug repurposing as anthelmintics.

Visceral cestodiases, like echinococcoses and cysticercoses, are zoonoses of worldwide distribution and are responsible for public health problems in many countries, especially in underdeveloped regions. Current treatments have low efficiency and there are few drugs currently in use for chemotherapy, making the development of new anthelmintics an urgent matter. The nucleotide salvage pathways are the only ones available for nucleotide synthesis in cestodes and other parasitic helminths, and, here, we used in silico approaches to assess the potential of the enzymes in these pathways as targets

Hidden population structure and cross-species transmission of whipworms (Trichuris sp.) in humans and non-human primates in Uganda.

BACKGROUND: Whipworms (Trichuris sp.) are a globally distributed genus of parasitic helminths that infect a diversity of mammalian hosts. Molecular methods have successfully resolved porcine whipworm, Trichuris suis, from primate whipworm, T. trichiura. However, it remains unclear whether T. trichiura is a multi-host parasite capable of infecting a wide taxonomic breadth of primate hosts or a complex of host specific parasites that infect one or two closely related hosts. METHODS AND FINDINGS: We examined the phylogenetic structure of whipworms in a multi-species community of non-human primat

A method for detecting microfilaraemia, filarial specific antigens and antibodies and typing of parasites for drug resistance and genotypes using finger prick blood sample.

Monitoring and evaluation of programme to eliminate lymphatic filariasis (LF) depends on epidemiological assessment using appropriate indicators. Minimum efforts using reliable tests are necessary to guide the programme managers in decision-making. Impact of Mass Drug Administration (MDA) towards filariasis elimination can be assessed by the detection of microfilariae (mf) or parasite DNA (infective), filarial antigens (infected) and antibodies (exposure). It is also important to monitor drug resistance and variation in genetic structure of parasite populations using molecular markers. We deve

The mitochondrial genome of the sheep roundworm Ascaris ovis (Ascaridida: Nematoda) from Southwest China.

The Ascaris roundworms (Ascaridida: Nematoda), one of the commonest soil-transmitted helminths (STHs), can cause ascariasis with significant socioeconomic and public health impact. In this study, the mitochondrial genome of Ascaris ovis, a representative of this genus from the sheep in Southwest China was determined using Illumina sequencing technology. The assembled genome was 14,205 bp in size and encoded 36 genes, including 12 protein-coding genes, 22 transfer RNA genes, and two ribosomal RNA genes. Phylogenetic analysis showed that A. ovis grouped with the congeneric Ascaris lumbricoides o

Morphological and molecular characterisation of two new species of Rhipidocotyle (Digenea: Bucephalidae Poche, 1907) from Sphyraena putnamae Jordan & Seale in Mozambique.

Species-level delineation of digenetic trematodes is complex and can be best achieved by integrative taxonomy using both genetic characterisation and morphological analysis. Two new Bucephalidae species of the genus Rhipidocotyle Diesing, 1858 are described here based on specimens collected from the intestine of Sphyraena putnamae Jordan & Seale following this approach. Adults of R. siphonyaka n. sp. and R. nolwe n. sp. possess tentacles and a tegument with scales. They are distinguished from their congeners by the arrangement of the digestive structures, the extent of the uterus relative to v

Parasitological and molecular study of the furcocercariae from Melanoides tuberculata as a probable agent of cercarial dermatitis.

Cercarial dermatitis is caused by animal schistosomes in many parts of the world including Iran. Various stages of the parasites have been studied in intermediate and definitive hosts in northern and southwestern Iran; however, no molecular investigation for species identification and classification of these agents has been carried out, so far. In the present study, more than 3,800 aquatic snails were collected from water sources of Khuzestan, southwest Iran. The snails were identified as Lymnaea gedrosiana, Radix auricularia, Melanoides tuberculata, Melanopsis sp. and Physa acuta. They were e

Molecular epidemiology of Brazilian Biomphalaria: a review of the identification of species and the detection of infected snails.

The three vector species of Schistosoma mansoni in Brazil, Biomphalaria glabrata, B. tenagophila and B. straminea show different susceptibility levels to the trematode besides a wide geographical distribution. The identification of such molluscs is important to further understand the disease epidemiology. Considering the fact that morphological identification may become difficult or even impossible under particular circumstances, the use of molecular-based methods have permitted the generation of more consistent information concerning the population structure of Biomphalaria furthering knowled

CHCHD2 as a prognostic biomarker and therapeutic target in lung adenocarcinoma: Insights from pan‑cancer multi‑omics and functional studies.

Helminths include both parasitic nematodes (roundworms) and platyhelminths (trematode and cestode flatworms) that are abundant, and are of clinical importance. The genetic characterization of parasitic flatworms using advanced molecular tools is central to the diagnosis and control of infections. Although the nuclear genome houses suitable genetic markers (e.g., in ribosomal (r) DNA) for species identification and molecular characterization, the mitochondrial (mt) genome consistently provides a rich source of novel markers for informative systematics and epidemiological studies. Diversity of trematode larvae in Indoplanorbis exustus from Phayao Lake, northern Thailand: Genetic evidence of Schistosoma spindale and a potential novel Schistosomatid associated with swimmer's itch.

Indoplanorbis exustus, a pulmonate snail, is an important component of freshwater ecosystems and serves as the primary intermediate host for the larval stages of several medically and veterinary significant trematodes. However, data on its infection status and associated trematode species in the Phayao Lake area of northern Thailand are currently lacking. This study aimed to investigate the prevalence of infection and identify the trematode species present in snails from this region. A total of 1290 snails were collected from Phayao Lake between November 2024 and February 2025 and examined usi

Heparan sulphate binding controls in vivo half-life of the HpARI protein family.

The parasitic nematode Heligmosomoides polygyrus bakeri secretes the HpARI family, which bind to IL-33, either suppressing (HpARI1 and HpARI2) or enhancing (HpARI3) responses to the cytokine. We previously showed that HpARI2 also bound to DNA via its first complement control protein (CCP1) domain. Here, we find that HpARI1 can also bind DNA, while HpARI3 cannot. Through the production of HpARI2/HpARI3 CCP1 domain-swapped chimeras, DNA-binding ability can be transferred, and correlates with in vivo half-life of administered proteins. We found that HpARI1 and HpARI2 (but not HpARI3) also binds t

Cryptic speciation among Tylodelphys spp.: the major helminth pathogens of fish and amphibians.

Larvae of Tylodelphys Diesing, 1950 are major digenean pathogens of fish and amphibians. Tylodelphys spp. may induce mass mortality of fish and increase their susceptibility to predation. Even though Tylodelphys spp. cause substantial damage to aquaculture systems, surprisingly little is known regarding the taxonomy of this commercially important genus with a limited number of visible autapomorphic identification features. The authors obtained the DNA sequences and analyzed the molecular phylogenetics of Tylodelphys spp. adults isolated from bird hosts of Czech origin and provide comparative m
