Photobiomodulation and the Wnt/β-catenin pathway in apexogenesis: Rationale and evidence gaps.

Apexogenesis is a vital pulp therapy that preserves vascularized pulp in immature teeth and relies on dental pulp stem cells (DPSCs) and stem cells from the apical papilla (SCAPs) for continued root growth and apical closure. Wnt/β-catenin signaling has an established role in odontoblast differentiation, reparative dentinogenesis, and root development, although normal root development requires regulated rather than simply increased Wnt activity. Photobiomodulation therapy (PBMT) is proposed to act through mitochondrial photoreception, and in vitro studies report increased expression of odontog

Endodontic management of taurodontism in a patient with Morquio syndrome: Case report of a 16-year-old girl.

BACKGROUND AND AIM: Mucopolysaccharidosis (MPS) type IV, referred to as Morquio A syndrome (MAS), is a rare genetic disorder characterized by an insufficient level in the storage of glycosaminoglycans within lysosomes. Diagnosis generally depends on clinical examination, skeletal radiographs, and histochemical tests. The condition is characterized by prominent skeletal deformities, limited joint mobility, significant growth impairment, abnormalities in tooth alignment, and defects in tooth enamel. The present clinical case report aims to provide details of a MAS case with peculiar dental findi

The Effect of Culture Medium Obtained from Dental Pulp Stem Cells (Conditioned Medium) on Esophageal Squamous Cell Carcinoma Cell Line (KYSE-30).

INTRODUCTION: Esophageal squamous cell carcinoma (ESCC) is the eighth most prevalent cancer worldwide. Head and neck squamous cell carcinoma (HNSCC) represents over 90% of all head and neck cancers, and nearly 40% of patients fail treatment. Therefore, discussion of cancer treatment is essential. In this in vitro study, we examined how conditioned medium from dental pulp stem cells (DPSC-CM) influences the ESCC cell line (KYSE-30). MATERIALS AND METHODS: First, the middle pulp tissue of the wisdom tooth was extracted and, following sterilization, transferred to a cell culture flask containing

Guanylate-binding protein 5-mediated autophagy can promote the clearance of intracellular F. nucleatum in dental pulp cells during pulpitis.

BACKGROUND: IFN-γ is crucial in induction of inducible cell-autonomous immunity, and IFN-γ signaling pathway is activated in pulpitis. Guanylate-binding proteins (GBPs) are a family of IFN-inducible GTPases and could utilize autophagy or pyroptosis to mitigate infection. GBP5 is abundantly expressed in inflamed pulp and human dental pulp cells (HDPCs). Therefore, we hypothesize that GBP5 in HDPCs exerts an immune-regulatory role in defending against bacterium infection. METHODS: Fusobacterium nucleatum (F. nucleatum) was used to infect HDPCs, and immunoblotting and qRT-PCR were used to detect

Tissue Biocompatibility and Antimicrobial Properties of Sympathomimetic Nasal Solutions for Potential Use in Dental Pulpal Management.

OBJECTIVES: Sympathomimetic nasal solutions (SNS), containing oxymetazoline or phenylephrine, may be used directly onto dental pulp tissue and initiate hemostasis via activation of alpha-receptors that innervate arteriole smooth muscles. The aim of this study was to assess these SNS's biocompatibility with human dental pulp stem cells (hDPSCs) and ability to kill a deep caries-associated pathogen, Rothia dentocariosa. MATERIALS AND METHODS: A Kirby-Bauer disk diffusion susceptibility test assessed SNS zones of inhibition (ZOI) toward R. dentocariosa growth on agar plates. Over-the-counter (OT

Myricetin Modulates Matrix Metalloproteinases Expression Induced by TEGDMA in Human Odontoblast-Like Cells.

The activity of matrix metalloproteinases (MMPs) plays a crucial role in the aging of the resin-dentin interface. The in situ action of MMP-2 and MMP-9 has been confirmed in the process of dentin-collagen degradation. However, the involvement of dental pulp cells in MMP secretion as a response to oxidative stress induced by contact with resin monomers has not been fully elucidated. Myricetin (MYR), like proanthocyanidin (PAC), has antioxidant properties and may help prevent extracellular matrix degradation. The objective was to evaluate the effect of MYR on the MMP expression and activity in r

Melatonin antagonizes lipopolysaccharide-induced pulpal fibroblast responses.

BACKGROUND: Pulpal inflammation is known to be mediated by multiple signaling pathways. However, whether melatonin plays regulatory roles in pulpal inflammation remains unclear. This study aimed at elucidating an in situ expression of melatonin and its receptors in human pulpal tissues, and the contribution of melatonin on the antagonism of lipopolysaccharide (LPS)-infected pulpal fibroblasts. METHODS: Melatonin expression in pulpal tissues harvested from healthy teeth was investigated by immunohistochemical staining. Its receptors, melatonin receptor 1 (MT1) and melatonin receptor 2 (MT2), w

Resin Infiltration for the Esthetic Improvement of Dental Fluorosis and White Spots: A Case Report.

White spot lesions, characterized by opaque enamel discolorations from early carious lesions, fluorosis, or developmental issues, can be effectively managed through various treatments. Resin infiltration is a unique therapy that bridges the gap between non-invasive and invasive alternatives, potentially delaying the need for restorations by applying a low-viscosity resin that penetrates and seals the lesion, enhancing both appearance and function. This study evaluates the clinical and esthetic outcomes of resin infiltration in a 33-year-old female patient with mild dental fluorosis, who presen

The endodontic diagnostic puzzle.

Endodontic diagnosis is the cornerstone of endodontic treatment. Endodontic diagnosis can be likened to a puzzle, where the pieces must be gathered and pieced together before a clinician can see the complete picture. This article discusses how to collect the pieces and fit them together to see the pulpal and periapical diagnosis emerge.

A lesion of endodontic origin misdiagnosed as a globulomaxillary cyst.

This report presents a case of a lesion originating from a pulpless central incisor misdiagnosed as a globulomaxillary cyst. The initial diagnosis of globulomaxillary cyst was made solely from radiographs by observing an almond-shaped radiolucency between the lateral incisor and cuspid. No pulp testing was performed before surgical removal of the lesion. The final diagnosis of an endodontic lesion caused solely by a pulpless central incisor was complicated by the initial lack of a pulpal response from the lateral incisor next to the surgical site. By delaying the start of endodontic treatment,

Upregulation of TACAN in the trigeminal ganglion affects pain transduction in acute pulpitis.

OBJECTIVE: Acute pulpitis is one of the common causes of tooth pain. TACAN (Tmem120a) is a newly identified ion channel that senses mechanical pain. In this experiment, we studied the expression of the TACAN ion channel in the trigeminal ganglia in a rat model of pulpitis to explore the correlation between the expression of this ion channel and inflammatory pain. DESIGN: Lipopolysaccharide was used to induce acute pulpitis in rats, and pulpitis was assessed histologically. The facial pain threshold of the rats was measured by the von Frey test. TACAN mRNA expression in rat dental pulp and the

Comparative evaluation of 1.8 mL and 3.6 mL of 2% lidocaine with 1:200,000 epinephrine for inferior alveolar nerve block in patients with irreversible pulpitis: a prospective, randomized single-blind study.

INTRODUCTION: There is a decrease in the anesthetic efficacy of inferior alveolar nerve blocks in patients with irreversible pulpitis. It was hypothesized that the increasing the volume of anesthetic solution may improve the success rates of dental pulp anesthesia in patients with pulpal pain. METHODS: Fifty-five adult volunteers, actively experiencing pain, participated in this prospective, randomized, single-blind study. The patients were divided into 2 groups on a random basis and received an inferior alveolar nerve block with either 1.8 mL or 3.6 mL of 2% lidocaine with 1:200,000 epinephr

Reliability in estimating taurodontism of permanent molars from orthopantomograms.

Taurodontism is a morphologic dental trait showing continuous expressivity, and criteria of the degree of pulp chamber elongation vary in different investigations. The aim of this investigation was to test a simple method of assessing taurodontism in the developing dentition from orthopantomograms in order to determine its reliability for later use in epidemiologic investigations. The method was also compared with other methods. Forty-three children 10-16.9 yr of age with one or more taurodontic permanent first or second molars were selected for the study. A subgroup of 16 children with two lo

Effects of lipopolysaccharides on human dental pulp cells.

Human dental pulp cells were treated with 1, 10, and 100 micrograms/ml of lipopolysaccharide (LPS). The effects of treatment were examined by measurement of the DNA content, protein content, and alkaline phosphatase activity of the cells. LPS samples were purified from Porphyromonas gingivalis, Porphyromonas endodontalis, and Fusobacterium nucleatum isolated from root canals, and Escherichia coli 0111:B4 LPS was used as a positive control. At a concentration of 1 microgram/ml, none of the LPSs caused any change in the production of DNA or protein, whereas the amount of DNA was increased at 10
