Atopic dermatitis (AD) is a chronic inflammatory skin disease, driven by barrier and immune dysregulation, which causes significant impairment in quality of life. The introduction of biologics and oral small molecules has substantially improved treatment outcomes. However, achieving complete and durable clinical clearance for most patients remains challenging, and concerns related to long-term safety and healthcare burden persist as key unmet needs. These limitations have catalyzed a new phase of therapeutic innovation in AD. Next-generation biologics targeting type 2 cytokines are being refin

Atopic dermatitis (AD) is driven by complex interactions between keratinocytes and immune cells. In this issue of Cell Chemical Biology, Shao et al.1 identify that NAT10-dependent ac4C modification stabilizes Relb mRNA in keratinocytes, promoting chemokine production, neutrophil recruitment, and dermatitis progression.

BACKGROUND: Atopic dermatitis (AD) is a prevalent, chronic inflammatory skin disease with diverse clinical presentations, often overlapping with other dermatoses. Its diagnosis remains largely dependent on clinical expertise, leading to variability and limited diagnostic accuracy, particularly among general practitioners. This study aimed to develop and evaluate a multimodal artificial intelligence (AI) model that integrates lesion image analysis and structured anamnesis to improve AD diagnosis.
METHODS: This diagnostic study was conducted in two phases: Phase 1 used retrospective data from 2

BACKGROUND: Atopic dermatitis (AD) is a chronic inflammatory skin condition that significantly impairs quality of life. Current therapies, including corticosteroids and calcineurin inhibitors, often provide incomplete relief or have undesirable side effects. Exosome-based therapies derived from adipose-derived mesenchymal stem cells (ADMSCs) offer a novel, multifaceted approach to addressing inflammation, skin barrier dysfunction, and pruritus, presenting a promising alternative for AD management.
OBJECTIVE: This case series evaluates the efficacy and safety of ADMSC-derived exosome-based top

Atopic dermatitis (AD) is a recurrent and chronic inflammatory skin condition characterized by a high lifetime prevalence and significant impairment of patients' quality of life, primarily due to intense itching and discomfort. However, current pharmacological interventions provide only moderate efficacy and are frequently accompanied by adverse side effects. The immune-pathogenesis of AD involves dysregulation of the Th2 immune response and exacerbation of inflammation related to excessive reactive oxygen species (ROS). Therefore, to address these issues, in this study, we targeted the upstre

BACKGROUND/PURPOSE: Among the characteristics that appear in the epidermis of the skin, erythema is primarily evaluated through qualitative scales, such as visual assessment (VA). However, VA is not ideal because it relies on the experience and skill of dermatologists. In this study, we propose a new evaluation method based on hyperspectral imaging (HSI) to improve the accuracy of erythema diagnosis in clinical settings and investigate the applicability of HSI to skin evaluation.
METHODS: For this study, 23 subjects diagnosed with atopic dermatitis were recruited. The inside of the right arm

PURPOSE OF REVIEW: To analyze the efficacy and safety of Janus kinase inhibitors (JAKi) in the treatment of pediatric AD.
RECENT FINDINGS: Adolescents with moderate and severe atopic dermatitis (AD) need systemic therapies, as stated several recent practice guidelines. (JAKi) have shown their efficacy in the treatment of adult AD, however, there is a lack of information concerning efficacy and safety of their use in pediatric AD. We found that the JAKi's abrocitinib (ABRO), baricitinib (BARI), and upadacitinib (UPA), are all an effective treatment option with a very fast onset of action for a

BACKGROUND: Hand eczema (HE) is a common inflammatory skin disease that may have serious consequences. The age of HE onset varies, but is estimated to be early- to mid-20s. However, very little is known about HE in childhood and adolescence.
OBJECTIVE: We aimed to explore the epidemiology, aetiology and severity of HE among a random sample of Danish adolescents drawn from the general population.
METHODS: The study was designed as a self-administered questionnaire study. An electronic questionnaire was sent to a random sample of 13 000 individuals aged 15-19 years.
RESULTS: The point-prevale

Alopecia areata (AA) is regarded as a tissue-specific and cell-mediated autoimmune disorder. Regarding the cytokine balance, AA has been considered a type 1 inflammatory disease. On the other hand, AA often complicates atopic dermatitis (AD) and AD is regarded as type 2 inflammatory disease. However, the immunological aspects of AA in relation to AD are still poorly understood. Therefore, we aim to clarify the immunological properties of AD-associated AA. In this study, we performed comparative analysis of the expression of intracytoplasmic cytokines (IFN-γ, IL-4, and IL-13), chemokine recepto

Ciclosporin (CsA) is a systemic immunosuppressive agent indicated for use in the treatment of patients with recalcitrant atopic dermatitis (AD). Studies demonstrate reasonable evidence of benefit compared with placebo. However as biologic agents are under development for use in AD, an assessment of the use, attitude to and safety of CsA is essential in understanding unmet needs in severe AD. We present the results of a survey asking questions relating to initial dose, duration of prescription, precautions and monitoring undertaken during treatment, adverse events seen, and hierarchy of use for

The consequences of atopic dermatitis reach beyond the skin and past childhood. Patients with atopic dermatitis are at risk of developing allergic comorbidities, but less is known about the associations between atopic dermatitis and non-allergic conditions. Understanding these non-allergic comorbidities has the potential to improve patient outcomes and to help mitigate the cost and burdens associated with these conditions. Atopic dermatitis is associated with cutaneous bacterial infections, more severe forms/courses of cutaneous viral infections, and extra-cutaneous infections. Atopic dermatit

Atopic dermatitis (AD) is a common chronic, pruritic skin disease in children. As the incidence of AD increases, especially in high-income countries, paediatricians may see an increasing number of recalcitrant AD cases in their practice. Although these cases are principally managed by paediatric dermatologists, it is important for paediatricians to be aware of the use and side effects of non-topical treatment like phototherapy and systemic agents as well as the evidence for alternative treatment, which caregivers may ask about. This review presents paediatric evidence for the practical use of

Coxsackievirus A6 (CV-A6) is an emerging pathogen that has in recent years been associated with atypical hand, foot and mouth disease. This manifests as a generalized papular or vesicular eruption, which may be associated with fever and systemic disturbance. We report a series of six children presenting to a single centre in the UK with disseminated CV-A6 infection on a background of atopic dermatitis (AD). Our patients exhibited a widespread papular or vesicular eruption in association with exacerbation of AD. Several of our cases mimicked eczema herpeticum, but the extent was more generalize

A 14-year-old patient with severe widespread atopic dermatitis (AD) and concurrent molluscum contagiosum virus (MCV) presented with MCV papules that were surrounded by dermatitis-free zones. It appeared that components inhibited the expression of AD, with inhibitory effects that persisted throughout 6 years of follow-up, even in the presence of a Staphylococcus aureus infection, which was proven by skin culture.

BACKGROUND: Primary localized cutaneous amyloidosis (PLCA) is a chronic pruritic dermatological disorder of unknown aetiology. Genetic mutations in cases of familial PLCA have been mapped to the oncostatin-M receptor (OSMR) β, a subunit of interleukin (IL)-31 receptor. IL-31 has been implicated in the pathogenesis of atopic dermatitis (AD).
OBJECTIVES: To assess if AD is more prevalent in patients with PLCA compared to patients with other conditions attending the same dermatology clinic. Secondarily, to investigate if the prevalence of AD, severity of itch, morphology and locations of PLCA di

Subjects afflicted with atopic dermatitis are particularly predisposed to such viral superinfections as verruca, herpes (eczema herpeticum) and vaccinia (eczema vaccinatum). Knowledge of superinfection by the molluscum contagiosum virus is more recent and the authors describe one case. The orf virus (responsible for ovine ecthyma contagiosum), however, has never been described as responsible for the profuse superinfection of an atopic eczema. The authors observed one case of this in a child, atypical as result of his age (16 months), the cervical localization and the aspect, which was a telang
