Pathophysiological role of dopamine on the development of hypertension in rats.

The aim of the study is to investigate the pathophysiological role of dopamine (DA) in the development of hypertension in DOCA-salt hypertensive rats and spontaneously hypertensive rats (SHRs). The augmentation of dopaminergic activity by chronic administration of bromocriptine, a DA agonist, suppressed the increase of blood pressure in DOCA-salt hypertensive rats. In contrast, suppression of dopaminergic activity by chronic administration of carbidopa, an inhibitor of dopa decarboxylase, accelerated the development of hypertension in SHRs, and this acceleration was also increased by salt load…

Chronic pharmacological manipulation of dopamine receptors in brain.

Dopamine receptors in the brain play an important role in the treatment of schizophrenia and in the development of tardive dyskinesia. In Parkinson's disease the loss of dopamine innervation and the use of chronic administration of L-DOPA or therapy with dopamine agonists also affects the function of dopamine receptors in brain. Subacute administration of neuroleptic drugs to rodents for a few weeks followed by the withdrawal of the drug induces supersensitivity of dopamine receptors in the striatum. However, the long-term administration of neuroleptic drugs to rodents shows that typical neuro…

[Plasma dopamine concentrations in various types of hypertension].

The concentrations of unconjugated plasma dopamine (PDA) were studied in patients with various types of hypertension. Catecholamines were extracted from plasma specimens (1.0-3.0 ml) through an Amberlite CG50 (Li+-form) microcolumn and eluted by a magnesium sulfate - ethanol solution. The elute was then desalinated and deproteinized by the ethanol-treated precipitation procedure and dried in a vacuum oven at 25 degrees C. A fraction of catecholamines was assayed with the modified procedures of the COMT-mediated radio-enzymatic method. This assay system was sensitive enough to permit an accurat…

Pharmacology and therapeutic use of low-dose dopamine.

The renal pharmacologic effects of intravenous dopamine in doses of 0.5-3.0 micrograms/kg/min include increases in renal blood flow, glomerular filtration rate, solute excretion, and urine flow. Clinical studies revealed that low-dose dopamine can reverse oliguria, but these studies were poorly controlled, were confounded by the use of other diuretics, had small patient populations, and often did not evaluate mortality or long-term renal function. When used in low doses, side effects are rarely seen. Because of dopamine's effect on hepatic and renal function, changes in drug clearance may occu…

[Role of dopamine receptors in the mechanism of action of neuroleptics].

The authors analyze the literature and their own data on the influence of neuroleptics on the cerebral dopamine and adrenoreceptors. It is shown that antipsychotic activity of neuroleptics correlates with their ability to block D2 dopamine receptor whereas their sedative properties are in a better agreement with the blockade of alpha 1-adrenoreceptor. Effects at the molecular level, other than the blockade of the above receptors, may be involved in forming the activity spectrum of each drug.

Ventilatory stimulation by dopamine-receptor antagonists in the mouse.

Ventilation was measured by a plethysmographic method in awake mice before and after intraperitoneal injection of neuroleptic drugs to test the hypothesis that dopaminergic mechanisms modulate control of breathing in this species. Dose-dependent augmentation of ventilation at rest and during hypoxia, and reduced ventilation during hypercapnia was demonstrated for haloperidol, droperidol, prochlorperazine and chlorpromazine (P less than 0.05 or less for each drug). Doses of drugs causing maximal increase of the ventilatory response to hypoxia were linearly related (r = 0.98, P less than 0.001) …

Assay of dopamine receptors with [alpha-3H]flupenthixol.

Two major classes of dopamine receptors, called D-1 receptors and D-2 receptors, have been identified. [alpha-3H]Flupenthixol has been used as a radioligand for the study of D-1 receptors, which are thought to act through stimulation of adenylate cyclase activity. Previous studies in our laboratory have shown that the D-2 receptors in rat caudate labeled by [3H]spiroperidol also have a high affinity for alpha-flupenthixol. The present experiments show that although Scatchard analysis of the binding of [alpha-3H]flupenthixol is consistent with the presence of a homogeneous population of recepto…

Increased dopamine production in patients with carcinoid tumors.

Although both carcinoid tumors and pheochromocytomas arise from neural crest origin, they are thought to each secrete distinctive monoamines; carcinoid tumors, which contain tryptophan hydroxylase, secrete serotonin, and pheochromocytomas, which contain tyrosine hydroxylase, may secrete dopamine (DA), norepinephrine (NE), or epinephrine (E). The purpose of this study was to determine if patients with carcinoid tumors have evidence of increased DA production. Of patients with serotonin-producing carcinoid tumors, 18%, 27%, and 35%, respectively, had increased urinary homovanillic acid (HVA) exc…

[Vagal influence in experimental arrhythmias caused by dopamine].

Dopamine induced cardiac arrhythmias in control and reserpine-treated rats which was successfully++ prevented by functional inhibition of the parasympathetic system. Both EKG and intracarotid mean blood pressure were recorded through a polygraph. Animals were anesthetized with urethane, intravenously heparinized and enhanced its spontaneous pulmonary ventilation with a tracheal cannula. Dopamine (50, 100, 200 and 400 micrograms/100 g,) produced cardiac arrhythmias characterized by atrioventricular block, ventricular premature depolarization and ventricular tachycardia. The parasympathetic inhi…

The effect of dopamine on glomus cell membranes in the rabbit.

Rabbit carotid bodies were excised and the membrane potentials (MPs) of the glomus cells were recorded in vitro. Upstream injections of dopamine (DA; 25-250 micrograms) elicited a slow, long lasting depolarization in 76% of the cells studied. The remaining 24% were hyperpolarized by DA. Regardless of polarity, the DA-induced potential was associated with a significant increase in input resistance (Ro). On the other hand, changes in MP which were produced artificially by passing direct current through the recording electrode had little effect on Ro. It is concluded that DA alters the MP and Ro …

Dopamine and frusemide in oliguric acute renal failure.

Into 24 oliguric patients with acute renal failure (ARF) for whom mannitol and high-dose frusemide had failed to promote a diuresis, dopamine (3 micrograms/kg/min) plus frusemide (10-15 mg/kg/h) were infused for 6-24 h. In 19 of the 24 patients this treatment produced significant increases in diuresis (from 11 +/- 7 to 85 +/- 51 ml/h; p less than 0.001) and natriuresis (from 45 +/- 13 to 88 +/- 22 mEq/1; p less than 0.001), without any significant modification of blood pressure, pulse rate or central venous pressure. 10 of the 24 patients required dialysis: 5 because therapy failed to promote …

Dopamine and carotid body function in the newborn lamb.

The effect of dopamine on the acute ventilatory response to hypoxia was investigated in four newborn lambs studied on the 1st day of postnatal life and eight lambs studied between 7 and 12 days of age. Studies were accomplished during nonrapid-eye-movement sleep in unanesthetized tracheotomized animals. Changes in minute ventilation (VE/kg), tidal volume, and frequency induced by intravenous bolus injection of dopamine (10 micrograms/kg) or saline control were assessed while animals were breathing room air or N2, before and after carotid body denervation (CBD). Dopamine depressed resting venti…

Effects of naloxone on dopamine release in the nigrostriatal system.

Simultaneously with a systemic injection of naloxone (NAL), the effects of the nigral application of the d(+)-amphetamine (AMPH) or of right forepaw stimulation on the release of [3H]dopamine [( 3H]DA) in the two caudate nuclei (CN) and the two substantia nigrae (SN) were examined in halothane-anesthetized cats. These experiments were carried out using four push-pull cannulae implanted bilaterally in the CN and SN and continuously supplied with [3H]tyrosine [( 3H]Tyr), the metabolic precursor of dopamine. Nigral AMPH application (1 muM) produced a local increase of the [3H]DA in spite of the N…

Antipodal central effects of apomorphine and dopamine in chickens.

Dopamine, infused into the hypothalamus or injected into the 3rd cerebral ventricle of chickens pretreated with mebanazine, induced behavioural and electrocortical sleep, suppressed motor activity and lowered body temperature and CO2 elimination, effects prevented by phenoxybenzamine but not spiperone. The hypothermic effects of dopamine were intensified by testing chicks below thermoneutrality. Ampomorphine, infused into the hypothalamus, injected into the 3rd cerebral ventricle or given intravenously, evoked behavioural and electrocortical arousal in the intact chicken and in the encéphale i…

Inhibitory and excitatory effects of dopamine on carotid chemoreceptors.

The effect of intravenous dopamine on carotid body chemoreceptor activity was investigated in 6 anesthetized cats which were paralyzed and artificially ventilated. Studies were performed at 4 steady-state PaO2 levels at a constant PaCO2 and at 4 levels of PaCO2 during hyperoxia. Dopamine inhibited carotid chemoreceptors before and excited them after haloperidol. Moderate stimulation of the receptors by hypoxia and hypercapnia augmented dopamine's effects. These results indicate that both inhibitory and excitatory dopamine receptors are present in the carotid body.

Effects of dopamine agonists and antagonists in Tourette's disease.

The actions of haloperidol, dextroamphetamine sulfate, levamfetamine succinate, apomorphine, and piribedil were studied in two patients with Giles de la Tourette's disease in an attempt to clarify the catecholamine mechanisms involved in this condition. Both dextroamphetamine and levamfetamine increased the severity of the symptoms; dextroamphetamine was more potent. Haloperidol controlled the symptoms and also antagonized the effect of dextroamphetamine. Apomorphine injections reduced the severity of symptoms, even in the presence of dextroamphetamine. We conclude that dopamine rather than no…

Inhibitory action of dopamine on cat carotid chemoreceptors.

1. The influence of some drugs which affect the dopaminergic system was studied on chemosensory responses to dopamine (DA), acetylcholine (ACh), sodium cyanide NaCN) and hypoxia during experiments on pentobarbitone anaesthetized cats in which chemoreceptor activity was recorded from the peripheral end of a sectioned sinus nerve. 2. Spontaneous chemosensory activity was inhibited in a dose-dependent manner by DA (0.5-5 microgram, I.A.). Higher doses (10-50 microgram) caused a delayed increase in discharge and were associated with inconsistent inhibitory responses. 3. The DA antagonist alpha-flu…

Inhibition of thyrotropin and prolactin secretion by dopamine in man.

The effect of dopamine on thyrotropin (TSH) and prolactin (PRL) levels was studied in 5 normal subjects, 7 women with galactorrhea, 9 acromegalics and 4 patients with primary hypothyroidism. Dopamine infused at the rate of 280 micrograms/min produced significant decrease in plasma TSH and PRL levels in all four groups, though a lower fall in TSH was noted in acromegalics. A similar reduction in PRL was also noted after 28 micrograms/min dopamine. Phentolamine infusion (0.5 mg/min) had no effect on PRL response to dopamine. These results indicate that a dopaminergic stimulation led to an inhibi…
