Systematic review on the anxiolytic and hypnotic effects of flower extracts in in vivo pre-clinical studies published from 2010 to 2020.

Anxiety disorders are prevalent conditions in the world population, whose standard approaches include pharmacotherapy, psychotherapy, and combinations of these interventions. Different classes of psychopharmaceuticals are recommended as the first line of drugs to treat these disorders, which can have several adverse effects, treatment resistance, dependence, and drug-drug interactions making it necessary to search for new therapeutic agents. In particular, diazepam (DZP), a prototype drug from the group of benzodiazepines, has been commonly used and evaluated for its efficacy and safety in dif

Interactions of a standardized flavonoid fraction from Tilia americana with Serotoninergic drugs in elevated plus maze.

ETHNOPHARMACOLOGICAL RELEVANCE: Tilia americana var. mexicana (Schltdl) Hardin (Tiliaceae) aerial parts (bracts and flowers) are used in the traditional Mexican medicine to treat nervous disorders, as sedative and to treat insomnia. A fraction of this species called FC1 (organic fraction from this plant) was proposed, described as anxiolytic and characterized by the presence of flavonoids. In the present work, this fraction was standardized, and its interaction with different serotonergic drugs was tested. We used the elevated plus maze model as anxiety test and the open field test so as to ob

Effect of Electroconvulsive Therapy on Anxiety in Linear and Non-Linear Rats with Depressive-Like Disorders Induced by Ultrasound Stimulation.

We performed a comparative study of the effect of electroconvulsive therapy on anxiety in outbred rats and Sprague-Dawley rats with depressive-like disorder induced by exposure to ultrasound of variable frequency (20-45 Hz). The anxiogenic effect of therapy was revealed in elevated plus maze and open-field tests in both linear and non-lineal rats, but was more pronounced in linear animals. Differences between outbred rats and Sprague-Dawley rats by the anxiety level were demonstrated. These results suggest that electroconvulsive therapy should be used with cautious in patients with depression

Sedative and anticonvulsant effects of hydroalcoholic extract of Equisetum arvense.

The hydroalcoholic extract of Equisetum arvense (HAE) tested at the doses of 200 and 400 mg/kg showed a significant activity on the open-field, enhanced the number of falls in the rota-rod reducing the time of permanence in the bar and increased the sleeping time (46% and 74%) in the barbiturate-induced sleeping time. In the pentylenetetrazole-seizure, it increased the first convulsion latency, diminished the severity of convulsions, reduced the percentage of animals which developed convulsion (50% and 25%) and protected animals from death. On the contrary, in the elevated plus maze, the doses

Influence of differential housing on emotional behaviour and neurotrophin levels in mice.

Environmental enrichment condition (EC) induces profound behavioural, neurochemical and neuroanatomical changes. Increasing evidence has shown that the hippocampus, which is implicated in a range of cognitive functions, including learning and memory, is one of the most susceptible brain areas to the effects of enriched rearing. Recent work also suggests that the hippocampus is functionally segregated; lesion studies have shown that the dorsal hippocampus is important for spatial learning, whereas the ventral part is critical in emotional behaviour in rats. We investigated the effects of differ

Combined therapy with cyclodextrin/allopregnanolone and miglustat improves motor but not cognitive functions in Niemann-Pick Type C1 mice.

Niemann-Pick Type C1 (NPC1) is an autosomal recessive disorder characterized by the accumulation of cholesterol and glycosphingolipids. Combination-treatment utilizing cyclodextrin, allopregnanolone and miglustat (CYCLO/ALLO/miglustat) can ameliorate NPC1 disease in a mutant mouse model. The present study was designed to add behavioral analysis in NPC1 mutant mice upon CYCLO/ALLO/miglustat therapy. NPC1 mutant (BALB/cJ NPC1NIH) and control mice were used. For the combination treatment mice were injected with CYCLO/ALLO weekly, starting at P7. The miglustat injection was performed daily from P1

The impact of age on emotional and cognitive behaviours triggered by experimental neuropathy in rats.

Chronic pain syndromes encompass several clinical entities that frequently affect the individuals' emotional and cognitive behaviours which, in turn, can also alter pain perception. Additionally, both pain perception and motivational-affective behaviours change with increasing age. In order to evaluate the influence of age upon the interaction between chronic pain and affective/cognitive behaviours, 3-, 10- and 22-month-old rats with 1 month neuropathy (spared nerve injury, SNI model) were compared with age-matched sham-operated controls in the open field (OF; locomotor and exploratory behavio

Memory and mood-enhancing neuroprotective effects of visible light photoactivated gold nanoparticles and PEG3350 coating in a rat animal model.

Gold nanoparticles (AuNPs) emerge as promising neuromodulatory biomaterials due to their tunable optical properties, biocompatibility, and ability to cross the blood-brain barrier. Here, we investigated the behavioral and neuroprotective effects of citrate-stabilized AuNPs (~ 19 nm) coated with PEG3350 and irradiated under white (AuWL+PEG) or green light (AuGL+PEG), compared with non-irradiated PEGylated AuNPs (AuNP+PEG). Comprehensive physicochemical analyses demonstrated that green-light irradiation enhanced surface plasmon resonance (SPR) activity, improved PEG adsorption, and yielded super

Impaired neural circuitry of hippocampus in Pax2 nervous system-specific knockout mice leads to restricted repetitive behaviors.

INTRODUCTION: Restricted repetitive behaviors (RRBs), which are associated with many different neurological and mental disorders, such as obsessive-compulsive disorder (OCD) and autism, are patterns of behavior with little variation and little obvious function. Paired Box 2 (Pax2) is a transcription factor that is expressed in many systems, including the kidney and the central nervous system. The protein that is encoded by Pax2 has been implicated in the development of the nervous system and neurodevelopmental disorders. In our previous study, Pax2 heterozygous gene knockout mice (Pax2+/- mice

Memantine ameliorates learning and memory disturbance and the behavioral and psychological symptoms of dementia in thiamine-deficient mice.

Several studies have reported on the beneficial effects of memantine on behavioral and psychological symptoms of dementia (BPSD) in patients with Alzheimer's disease. However, the effects of memantine on BPSD-like behaviors in animals have not been well addressed. Here, the effects of memantine on memory disturbance and BPSD-like behaviors were evaluated in thiamine-deficient (TD) mice. Memantine (3 and 10 mg/kg, b.i.d.) was orally administered to ddY mice fed a TD diet for 22 days. During the treatment period, the forced swimming test, elevated plus-maze test, passive avoidance test, and loco

Anxiolytic-like effect of α-asarone in mice.

The effects of α-asarone in four assays predictive of anxiolytic activity in male mice were studied, with diazepam as a positive anxiolytic control. The use of the elevated plus-maze test revealed that diazepam (2 mg/kg) or α-asarone (3.5 mg/kg) increased the percentage of entries into open arms and of the time spent on open arms. In the light/dark transition test, as with 2 mg/kg diazepam, 7 mg/kg α-asarone increased the time spent in the light area and the number of transitions between the two compartments. In the novel food consumption test, α-asarone (3.5, 7 and 14 mg/kg) caused significan

Astrocyte Activation, but not Microglia, Is Associated with the Experimental Mouse Model of Schizophrenia Induced by Chronic Ketamine.

Ketamine is a noncompetitive antagonist of N-methyl-D-aspartate (NMDA) receptors. Many experimental studies have shown that ketamine can induce cognitive impairments and schizophrenia-like symptoms. While much data have demonstrated that glial cells are associated with the pathophysiology of psychiatric disorders, including schizophrenia, the response of glial cells to ketamine and its significance to schizophrenia are not clear. The present study was intended to explore whether chronic ketamine treatment would induce behavioral and glial changes in mice. First, ketamine was used to stimulate

Immunoglobulin-mediated neuro-cognitive impairment: new data and a comprehensive review.

Excessive influx of immunoglobulin (IgG) into the brain has been reported to induce central nervous system (CNS) dysfunction. Depressed patients may exhibit immune activation manifested by elevated inflammatory markers and pro-inflammatory cytokines. The brain and especially the limbic system contain high concentrations of high affinity Fc receptors. We reviewed the literature on this phenomena and present data on the behavioral effects of pooled normal IgG on the brain. Many disease states are associated with depression and we examined whether this may be linked to high IgG influx. Female Bal

Thiamine Deficiency Causes Long-Lasting Neurobehavioral Deficits in Mice.

Thiamine deficiency (TD) has detrimental effects on brain health and neurobehavioral development, and it is associated with many aging-related neurological disorders. To facilitate TD-related neuropsychological studies, we generated a TD mouse model by feeding a thiamine-deficient diet for 30 days, followed by re-feeding the control diet for either one week or 16 weeks as recovery treatment. We then performed neurobehavioral tests in these two cohorts: cohort of one week post TD treatment (1 wk-PTDT) and 16 weeks post TD treatment (16 wks-PTDT). The TD mice showed no significant difference fro
