Distance-dependent quenching and enhancing of electrochemiluminescence from a CdS:Mn nanocrystal film by Au nanoparticles for highly sensitive detection of DNA.

The quenching of electrochemiluminescence (ECL) from a CdS:Mn nanocrystal film by proximal Au nanoparticles was observed as a result of Förster energy transfer, while an enhancement of ECL takes place after hybridization with target DNA due to the energy transfer of ECL excited surface plasmon resonances in AuNPs to the CdS:Mn NCs at large separation, based on which an ultrasensitive and specific DNA biosensor was constructed.

Vibronic Coherence in a Minimal Pigment-Protein Complex Probed by Two-Dimensional Electronic-Vibrational Spectroscopy.

Observations of coherence in light-harvesting pigment-protein complexes have sparked significant interest in their role in energy transfer processes. However, resolving and understanding the origin of coherence in photosynthetic complexes has been challenging due to their structural complexity and overlapping spectral signals. Water-soluble chlorophyll-binding protein (WSCP), which binds only four chlorophyll molecules, offers a minimal model to study coherence and exciton dynamics. Using two-dimensional electronic-vibrational (2DEV) spectroscopy, we identify distinct excited-state features th…

A Competition among Three Ultrafast Photoinduced Events in Thiotropolone: Internal Conversion vs Intersystem Crossing vs ESIPT.

Gas phase excited-state quantum wavepacket dynamics simulations of the thiotropolone demonstrate the ultrafast triplet formation upon photoexcitation to the dipole-allowed S2 state. The dominant relaxation pathway of the S2-T4 intersystem crossing, facilitated by the strong spin-orbit coupling and narrow energy gap, competes with the S2 to S1/S3 internal conversion. The wavepacket populated in T4 via the former pathway decays to lower triplet states. Computed potential energy profiles suggest proton transfer via S2, which might compete with internal conversion and intersystem crossing. The non

Fluorescence Microscopy Imaging of Light-Harvesting in Self-Assembled Nanofibers of Naphthalenediimides toward Perylenediimide Guests.

The self-assembly of a bichromophoric naphthalenediimide (NDI) into nanofibers showed efficient energy transfer (light-harvesting) to perylenediimide (PDI) molecules through a host-guest interaction, which can be visualized by microscopy using samples deposited on glass surfaces. In combination with atomic force microscopy, spectral, and polarization analyses, fluorescence imaging unveiled the inhomogeneity, packing defects, and relative spatial arrangement of PDI and NDI molecular units, which were found to affect the exciton mobility along the NDI nanofibers and the energy transfer efficienc…

Quantum chain amplification in nanocrystalline Dewar benzenes by intramolecular sensitization.

Quantum chain reactions are characterized by the formation of several photoproducts per photon absorbed (ΦQC > 1) and constitute a promising signal amplification mechanism. The triplet-sensitized isomerization of Dewar benzene is known to undergo quantum chain reactions characterized by an adiabatic valence-bond isomerization to the excited state of Hückel benzene, which is able to transfer its triplet energy to a new ground state Dewar benzene that reacts to continue the chain. Given that diffusion-mediated energy transfer is the chain-limiting event in solution, we demonstrate here that reac…

When Dielectric Constants Deceive: Interrogating Solvation in Ionic Liquids with Cyclic Voltammetry.

Ionic liquids are increasingly discussed as alternatives to conventional organic solvents for applications based on photoinduced electron transfer. For the rational design of such applications, reliable estimates of electron-transfer driving forces are essential. Based on the Born model of solvation, the moderate dielectric constants of ionic liquids (εr ≈ 8 - 15) suggest that they should resemble medium-polarity solvents such as dichloromethane or pyridine in photoinduced electron transfer and exhibit comparable solvation energies. Here, we test this assumption by experimentally comparing the

High-k water intermediate layer-mediated charge transfer modulation toward stable and augmented output generation of droplet-based electricity generators.

Droplet-based electricity generators (DEGs), which share structural analogies with field-effect transistors (FETs), have garnered increasing attention for droplet energy harvesting. DEGs typically exhibit sharp output peaks with short charge transfer durations, which need to be extended to enhance the root mean square (RMS) output for practical use in energy harvesting. To address this limitation, this study introduces a methodology for spontaneously forming a high-k water intermediate (WIn) layer by capturing working droplets, inspired by strategies for FET. Based on the operation mechanism o

Probing excited-state dynamics and intramolecular proton transfer in 1-acylaminoanthraquinones via the intermolecular solvent response.

The dynamics of a series of 1-acylaminoanthraquinones with varying degrees of excited-state intramolecular proton transfer are studied in ac … n subpicosecond time scales. In addition, the solvent response probes the effects of intermolecular energy transfer as energy released during the reactive event is rapidly transferred to the local solvent environment and then dissipates to the bulk solvent on about a 10 ps time scale. A brief initial account of a portion of this work has appeared previously, J. A …

Mitochondrial micro-nano reactors via enhancing mitochondrial transfer and mitophagy for alleviating metabolic crisis.

Energy metabolic dysfunction is a major cause of impaired chronic wounds healing, in which disrupted mitochondrial transfer and autophagy imbalance further aggravate the cellular energy crisis. In this study, single-cell RNA sequencing (scRNA-seq) of clinical diabetic wound samples first identified a pivotal role for macrophage-to-fibroblast mitochondrial transfer in wound healing. This finding was further validated using diabetic wound models and histological analyses, highlighting these processes as potential therapeutic targets for alleviating energy metabolic stress. Based on these finding

Ab initio study of carbon-chlorine bond cleavage in carbon tetrachloride.

Chlorinated solvents in groundwater are known to undergo reductive dechlorination reactions with Fe(ll)-containing minerals and with corroding metals in permeable-barrier treatment systems. This research investigated the effect of the reaction energy on the reaction pathway for C-Cl bond cleavage in carbon tetrachloride (CCl4). Hartree-Fock, density functional theory, and modified complete basis set ab initio methods were used to study adiabatic electron transfer to aqueous-phase CCl4. The potential energies associated with fragmentation of the carbon tetrachloride anion radical (CCl4-) into a

A competitive fluorescence immunoassay based on intermolecular quenching using an N-terminally fluorescent-labeled IgG antibody.

The development of antibody-based fluorescent sensors relying on tryptophan-mediated quenching, such as Quenchbody (Q-body), often exhibits limited fluorescence responses because dye quenching depends on the location of tryptophan residues within the antibody. Here, we developed a competitive fluorescence immunoassay, termed an intermolecular Quenchbody (iQ-body), that utilizes intermolecular Förster resonance energy transfer (FRET) between an N-terminally fluorescent-labeled IgG antibody and an antigen-quencher conjugate. Anti-thyroxine IgG antibody (clone 6901 SPTN-5) showed minimal fluoresc…

Convergently evolved linear actuators in ballistic tongues.

Rapid movements in animals depend on efficient energy transfer to overcome inertia. Among vertebrates, dense tissue and limited elastic storage are thought to impose profound constraints on power output, making extreme ballistic performance noteworthy. Here, we show that chameleons (Chamaeleonidae) and some lungless salamanders (Plethodontidae) have independently converged on a shared biomechanical solution: a sliding-based linear actuator that launches the tongue via muscular squeezing of a tapered skeletal rod. This design decouples muscle action from skeletal movement, enabling acceleration…

Multi intercalators FRET enhanced detection of minute amounts of DNA.

A novel approach is presented that increases sensitivity and specificity for detecting minimal traces of DNA in liquid and on solid samples. Förster Resonance Energy Transfer (FRET) from YOYO to Ethidium Bromide (EtBr) substantially increases the signal from DNA-bound EtBr highly enhancing sensitivity and specificity for DNA detection. The long fluorescence lifetime of the EtBr acceptor, when bound to DNA, allows for multi-pulse pumping with time gated (MPPTG) detection, which highly increases the detectable signal of DNA-bound EtBr. A straightforward spectra/image subtraction eliminates sampl…

Closing-upon-repair DNA tetrahedron nanoswitch for FRET imaging the repair activity of 8-oxoguanine DNA glycosylase in living cells.

In situ imaging the repair activity of 8-oxoguanine (8-OG) DNA glycosylase in living cells is important as it is associated with genetic mutation. However, the existing imaging methods confront the interference of intracellular nuclease and resulting in false positive signal. Here, a closing-upon-repair DNA tetrahedron nanoswitch (CRTN) was designed for FRET imaging the repair activity of 8-OG DNA glycosylase in living cells with high specificity and accuracy. CRTN comprised a DNA tetrahedron, a recognition strand modified with 8-OG bases, and a reporting strand designed as hairpin structure a

High throughput screen of small molecules as potential countermeasures to galactic cosmic radiation induced cellular dysfunction.

Space travel increases galactic cosmic ray exposure to flight crews and this is significantly elevated once travel moves beyond low Earth orbit. This includes combinations of high energy protons and heavy ions such as 56Fe or 16O. There are distinct differences in the biological response to low-energy transfer (x-rays) or high-energy transfer (High-LET). However, given the relatively low fluence rate of exposure during flight operations, it might be possible to manage these deleterious effects using small molecules currently available. Virtually all reports to date examining small molecule man…

An imidazo[1,5-α]pyridines-based ratiometric fluorescent probe for sensing sulfur dioxide derivatives in real samples based on a FRET mechanism.

A novel fluorescence resonance energy transfer (FRET)-based ratiometric emission fluorescent probe AT was designed and developed in which the imidazo[1,5-α]pyridine was served as a FRET donor and tricyanofuran (TCF) as the FRET acceptor to detect SO32-/HSO3- based on the Michael addition reaction. Probe AT had a high energy transfer efficiency (95%) and a large pseudo-Stokes shift (259 nm) in EtOH/PBS buffer (5/5, v/v). It also possessed good selectivity and quick response to SO32-/HSO3-. There was good linearity between the ratio of fluorescence intensity (F499/F645) and the concentrations of…

High-Resolution Single-Molecule FRET via DNA eXchange (FRET X).

Single-molecule FRET is a versatile tool to study nucleic acids and proteins at the nanometer scale. However, currently, only a couple of FRET pairs can be reliably measured on a single object, which makes it difficult to apply single-molecule FRET for structural analysis of biomolecules. Here, we present an approach that allows for the determination of multiple distances between FRET pairs in a single object. We use programmable, transient binding between short DNA strands to resolve the FRET efficiency of multiple fluorophore pairs. By allowing only a single FRET pair to be formed at a time,

A FRET-ICT Dual-Modulated Ratiometric Fluorescence Sensor for Monitoring and Bio-Imaging of Cellular Selenocysteine.

Since the fluctuation of cellular selenocysteine (Sec) concentration plays an all-important role in the development of numerous human disorders, the real-time fluorescence detection of Sec in living systems has attracted plenty of interest during the past decade. In order to obtain a faster and more sensitive small organic molecule fluorescence sensor for the Sec detection, a new ratiometric fluorescence sensor Q7 was designed based on the fluorescence resonance energy transfer (FRET) strategy with coumarin fluorophore as energy donor and 4-hydroxy naphthalimide fluorophore (with 2,4-dinitrobe…

A new control method depending on primary phase angle of transcutaneous energy transmission system for artificial heart.

A new control method for stabilizing output voltage of the transcutaneous energy transmission system for artificial heart is proposed. This method is primary side, is outside of the body, which is not depending on a signal transmission system from the implanted device. The impedance observed from primary side changes from inductive to capacitive and the output voltage decreases drastically when the output current is large and the coupling factor is higher than that of the optimal condition. In this case, the driving frequency should be changed to higher so that the phase angle of the primary i
