The filum terminale of the frog spinal cord, a non transformed preparation: I. Morphology and uptake of gamma-aminobutyric acid.

The filum terminale of the frog spinal cord is a rather pure glial cell preparation, largely devoid of neuronal elements. gamma-Aminobutyric … evel radioautography clearly shows that GABA uptake occurs primarily in the glial cells and also in ependymal cells present in the FT. In that the FT contains few ependymal cells and a large number of glia, it is fair to state that most of the GABA accumulated by the FT reflects the glial transport of this amino acid. Unlike the adult frog, the spinal cord …

[Ventricular enlargement in experimental occlusion of superior sagittal sinus--in reference to histopathological findings (author's transl)].

An experimental occlusion of superior sagittal sinus was made in dogs according to ligation with thread at the posterior third portion of the sinus. Ventricular enlargement and histological changes in the cerebrum were studied in various stage after occulusion of the sinus. In the acute stage, no ventricular enlargement occured. However, in the chronic stage, nine dogs out of 12 dogs showed enlargement of the ventricle. Hitological examination revealed peeled ependymal layer from subependymal tissue, edematous findings in the periventricular tissue, perivascular gliofibrosis, perivascular hemo…

Cerebrospinal fluid mechanics across CNS barriers: from production, circulation, and clearance to mechanomedicine.

BACKGROUND: Cerebrospinal fluid (CSF) is often framed as a transport medium, yet its motion and pressure dynamics impose continuous mechanic … and is sensed by specialized mechanotransduction modules in choroid plexus epithelium, ventricular ependyma, perivascular astrocytic endfeet/neurovascular unit, and meningeal outflow/lymphatic pathways. We discuss how shear, pulsatile forcing, and pressure-compliance relationships interact with mechanosensitive ion/transport channels, ciliary polarity, gl …

ADAM10-dependent disruption of N-cadherin and ventricular zone in post-hemorrhagic hydrocephalus.

Intraventricular hemorrhage (IVH) and subsequent post-hemorrhagic hydrocephalus (PHH) are common, severe complications of preterm birth associated with substantial morbidity and mortality. The pathophysiology of IVH/PHH involves the disruption of N-cadherin-mediated cell junctions in the ventricular zone (VZ), impaired differentiation of radial glial cells into ependymal cells, and reactive astrogliosis. A disintegrin and metalloproteinase domain-containing protein 10 (ADAM10) is a widely distributed metalloproteinase known to cleave cell junction proteins under inflammatory conditions similar…

Multicontrast single-slab 3D MRI to detect cerebral metastasis.

OBJECTIVE: The purpose of this study was to evaluate the sensitivities and specificities of four single-slab 3D MRI sequences in the detecti … 01). The double IR sequence was superior to the T2 FLAIR sequence for imaging metastases located in ependyma (81.8% vs 36.4%) and GM (66.7% vs 48.1%). Delayed enhancement did not affect the sensitivities of the contrast-enhanced T2 FLAIR and contrast-enhanced IR-prepared FSPGR sequences. CONCLUSION: Contrast-enhanced T2 FLAIR is the most sensitive sequenc …

[Intracranial metastasis of thymoma: report of three cases].

Thymoma with metastasis to the central nervous system is extremely rare. Herein, three rare cases of thymoma with intracranial metastasis are reported. Case 1 (thymoma, B3 in the WHO classification); cranial magnetic resonance imaging (MRI) revealed a mass lesion with a ring enhancement effect in the right temporal lobe. The cystic and hemorrhagic lesion was subtotally removed and radiation therapy of 40 Gy was administered to the tumor cavity containing the remains of the tumor around the ependyma. The patient received combination chemotherapy with carboplatin and gemcitabine hydrochloride, a…

Plasmalemmal vesicle associated protein-1 is a novel marker implicated in brain tumor angiogenesis.

PURPOSE: Plasmalemmal vesicle associated protein-1 (PV-1) is up-regulated in the endothelium of human glioblastoma. We sought to further cha … schemia. Expression levels were reduced in low-grade neoplasia; however, tumors associated with the ependyma and choroid plexus, known sites of PV-1 expression, also exhibited robust expression. Cerebral expression of PV-1 mRNA was confined to endothelial cells in all cases. PV-1 expression was induced in HMVEC cells in vitro by exposure to medium conditio …

Hypersensitivity to type I interferon as a cause of hydrocephalus development.

Ubiquitin specific protease 18 (USP18) serves as a potent inhibitor of Type I interferon (IFN) signaling. Previous studies have shown that Usp18 deficient (homozygous Usp18 gene knockout) mice exhibit hydrocephalus; however, the precise molecular mechanism underlying hydrocephalus development remains elusive. In this study, we demonstrate that mice lacking both type I IFN receptor subunit 1 (Ifnar1) and Usp18 (Ifnar1/Usp18 double knockout mice) are viable and do not display a hydrocephalus phenotype. Moreover, we observed that suppression of USP18 in ependymal cells treated with IFN significan…

Diversity of Adult Neural Stem and Progenitor Cells in Physiology and Disease.

Adult neural stem and progenitor cells (NSPCs) contribute to learning, memory, maintenance of homeostasis, energy metabolism and many other essential processes. They are highly heterogeneous populations that require input from a regionally distinct microenvironment including a mix of neurons, oligodendrocytes, astrocytes, ependymal cells, NG2+ glia, vasculature, cerebrospinal fluid (CSF), and others. The diversity of NSPCs is present in all three major parts of the CNS, i.e., the brain, spinal cord, and retina. Intrinsic and extrinsic signals, e.g., neurotrophic and growth factors, master tran…

Uncovering a New Cause of Obstructive Hydrocephalus Following Subarachnoid Hemorrhage: Choroidal Artery Vasospasm-Related Ependymal Cell Degeneration and Aqueductal Stenosis-First Experimental Study.

BACKGROUND: Hydrocephalus is a serious complication of subarachnoid hemorrhage (SAH). Obstruction of the cerebral aqueduct may cause hydrocephalus after SAH. Although various etiologic theories have been put forward, choroidal artery vasospasm-related ependymal desquamation and subependymal basal membrane rupture as mechanisms of aqueductal stenosis have not been suggested in the literature. METHODS: This study was conducted on 26 hybrid rabbits. Five rabbits were placed in a control group, 5 were placed in a sham group, and the remaining rabbits (n = 16) were placed in the SAH group. In the …

Forebrain ependymal cells are Notch-dependent and generate neuroblasts and astrocytes after stroke.

Neurons are continuously generated from stem cells in discrete regions in the adult mammalian brain. We found that ependymal cells lining the lateral ventricles were quiescent and did not contribute to adult neurogenesis under normal conditions in mice but instead gave rise to neuroblasts and astrocytes in response to stroke. Ependymal cell quiescence was actively maintained by canonical Notch signaling. Inhibition of this pathway in uninjured animals allowed ependymal cells to enter the cell cycle and produce olfactory bulb neurons, whereas forced Notch signaling was sufficient to block the e…

[Histochemical study of tanycytes in connection with the adrenocorticotropic function of the hypophysis].

Chronic experiments were conducted on sexually mature rats; histochemical study of the activity of some redox enzymes (glutamic dehydrogenase, lactic dehydrogenase, glucoso-6-phosphoric dehydrogenase, glycerophosphoric dehydrogenase, and succinic dehydrogenase) was carried out in the ependymal cells of the floor of the third cerebral ventricle, the so called tanycytes, in case of an increased adrenocorticotrophic function of the hypophysis attained by bilaterial adrenalectomy, and in depression of this function as a result of chronic dexametasone administration. The activity of the enzymes und…

Metabolic reprogramming in amyotrophic lateral sclerosis ependymal stem cells by FM19G11 nanotherapy.

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease affecting motor neurons in the motor cortex, brainstem, and the spinal cord. In response to neurodegeneration, spinal cord exhibits ineffective regenerative attempt, thus suggesting that therapeutic strategies aimed at enhancing regenerative capacity of ependymal stem/progenitor cells (epSPCs), residing in the spinal cord, could promote neurogenesis. Dysregulated levels of metabolites might disturb epSPC differentiation, and their restoration might favour neurogenesis. This study aimed to investigate the metabolomic profi…

PHACE syndrome with intracerebral hemangiomas, heterotopia, and endocrine dysfunction.

PHACE is an acronym to describe the association of posterior fossa brain malformations, hemangiomas, arterial anomalies, coarctation of the aorta, cardiac defects, and eye abnormalities. More than 200 cases have been reported. The present report presents the cases of two female infants with PHACE syndrome, both of whom had additional congenital defects of subependymal gray matter heterotopia, craniofacial arterial anomalies, and pituitary dysfunction. One had an extensive segmental facial hemangioma with ipsilateral intracranial hemangiomas. The other had multiple cutaneous hemangiomas, but no…

Organ culture of the bovine subcommissural organ: evidence for synthesis and release of the secretory material.

The subcommissural organ (SCO) is a brain circumventricular organ formed by ependymal and hypendymal secretory cells. It secretes glycoproteins into the cerebrospinal fluid of the third ventricle where they condense into a thread-like structure known as Reissner's fiber (RF). The present study was designed to investigate whether or not the bovine SCO continues to synthesize and release glycoproteins after a long-term culture. Cultured explants of SCO survive for several months. The content of the secretory granules present in the cultured ependymocytes displayed immunoreactive and lectin-bindi…

Magnetization transfer in the investigation of patients with tuberous sclerosis.

We examined 21 patients aged 5 months to 19 years, on a 1.5 T magnet. T1-weighted spin-echo images, proton density and T2-weighted images with spin-echo and turbo spin-echo sequences, and contrast-enhanced magnetization transfer (MT) T1-weighted images were obtained in all cases. MT T1-weighted images were performed before injection in 9 patients. Subependymal nodules were found in 14, and cortical and subcortical tubers in 20 of the 21 patients. MT T1-weighted images showed tubers and subependymal nodules as higher signal than normal gray matter and revealed more tubers than conventional sequ…

Axon-glial relationships in the anterior medullary velum of the adult rat.

The anterior medullary velum is a thin sheet of CNS tissue which roofs the rostral part of the IVth ventricle and contains fascicles of myel … nd where the basal lamina tubes of the Schwann cell units were discontinued. The basal processes of ependymal cells lining the ventricular wall of the velum, passed between axon bundles before abutting on the basal lamina of the pia. Many of these processes branched and ran along the axonal bundles. A monolayer of microglia occupied a subependymal stratum …

Roles of a subependymal nodule of tuberous sclerosis on pathophysiology of epilepsy.

Polysomnographies (PSG) were performed on two cases with tuberous sclerosis (TS), both having subependymal nodules on the medial wall of the caudate nucleus adjacent to the thalamostriatal sulci. Clinically one had suffered from infantile spasm and which later turned out to be complex partial seizure with a rotation toward the right. The other had developed tonic seizure on the right with a rotation toward the left in early childhood. Clinico-pharmacological studies revealed the existence of synaptic supersensitivity of the dopamine (DA) receptor in the left caudate and PSG confirmed the synap…
