Chemotherapy-induced peripheral neuropathy (CIPN) lacks disease-modifying therapies, and how epitranscriptomic regulation drives central sensitization remains unclear. In the present study focusing on paclitaxel-induced peripheral neuropathy (PIPN), we identified a METTL3-circRNA axis in the spinal dorsal horn wherein upregulated METTL3 expanded the m6A-modified circMtmr7 pool in paclitaxel-treated mice. Accordingly in paclitaxel-treated mice, upregulated METTL3 paralleled elevated m6A-circMtmr7 expression; METTL3 knockdown prevented the paclitaxel-evoked rise in m6A-circMtmr7, normalized mini

OBJECTIVE: Tuberculosis (TB) remains a global health threat with an incompletely understood pathogenesis. The RNA epigenetic modification N6-methyladenosine (m6A) is a key regulator of immune responses in various diseases, but its role in human TB is largely unexplored. This study aimed to investigate the involvement of m6A modification in TB by profiling its dynamics and the expression of its regulatory enzymes in clinical cohorts. METHOD: In this cross-sectional study, 105 participants were enrolled and classified into four groups based on clinical and laboratory findings: microbiologically-

N6-methyladenosine (m6A) modification plays a crucial role in regulating immune responses against Mycobacterium tuberculosis (Mtb) infection, but the underlying mechanisms remain unclear. Here, we analyzed m6A regulatory gene expression in GEO dataset GSE139825 and validated the finding in THP-1-derived macrophages. Bioinformatics analysis showed that METTL14, ALKBH5, and YTHDC1 were significantly downregulated, while METTL3 was upregulated in Mtb-infected alveolar macrophages compared to controls. In vitro experiments confirmed METTL14 expression was dose-dependently reduced with Mtb infectio

Over the past three decades, the biology of EIF3E (int6) has emerged as far more complex than originally anticipated. Initially identified as a common integration site of Mouse Mammary Tumor Virus (MMTV) and subsequently recognized as a component of the eukaryotic translation initiation factor 3 (eIF3) complex, EIF3E is now understood as a multifunctional regulator involved in selective mRNA translation, proteostasis, genome stability, cellular stress responses, and tissue homeostasis. Recent studies have further extended this framework by demonstrating that conditional Eif3e deficiency promot

Esophageal squamous cell carcinoma (ESCC) is a highly aggressive malignancy with poor prognosis, and the upstream regulatory mechanisms driving oncogenic signaling remain insufficiently defined. In this study, we identify a previously unrecognized nuclear function of the cytoskeletal protein ACTB in promoting ESCC progression. We show that ACTB translocates to the nucleus and directly binds to the promoter of the m6A reader gene YTHDC2, transcriptionally activating its expression. YTHDC2, in turn, binds to m6A-modified sites on AKT mRNA, preventing its degradation and enhancing its stability.

DNA and RNA methylation are key epigenetic and epitranscriptomic modifications involved in gene regulation, genome stability, RNA metabolism, and disease progression. Aberrant methylation patterns in cell-free DNA and RNA have emerged as valuable biomarkers for cancer detection, disease monitoring, and therapeutic stratification. However, conventional methods such as bisulfite sequencing, methylation-specific PCR, MeRIP-seq, SCARLET, and LC-MS/MS often require harsh processing, high sample input, complex instrumentation, or lack site-specific resolution, limiting their clinical and point-of-ca

Mammary gland inflammation, clinically expressed as mastitis in dairy ruminants and lactating women, arises from a self-amplifying loop of pathogen sensing, oxidative stress and innate immune activation, and remains a leading driver of antibiotic use, milk-quality loss and morbidity. Mounting pressure to reduce antimicrobial dependence has refocused attention on flavonoids-structurally diverse plant polyphenols with multi-target bioactivity, derived mainly from in vitro and rodent mastitis models. This review integrates contemporary evidence on the six principal flavonoid subclasses within a u

Circular RNAs (circRNAs) have emerged as a pervasive and functionally diverse class of regulatory molecules in breast cancer. While early studies predominantly described circRNAs as competing endogenous RNAs that modulate gene expression through microRNA sequestration, accumulating evidence indicates that this paradigm captures only a limited aspect of circRNA biology. Recent advances reveal that circRNAs function as central organizers of multilayered regulatory networks, integrating post-transcriptional control, RNA-protein interactions, noncanonical translation, metabolic adaptation, and imm

Global warming poses a considerable threat to crop production, making heat stress a challenge in agriculture, yet how epitranscriptomic modifications contribute to plant heat-stress responses remains to be explored. This study reveals the role of phase separation in plant heat-stress tolerance and shows that N-acetyltransferase 10 (NAT10), which encodes a cytosine N4 acetyltransferase, contributes to heat resistance. We found that NAT10 interacts with polyadenylate-binding protein (PABP), which contains intrinsically disordered regions, thereby facilitating the selective recruitment of ac4C-mo

Sepsis-induced cardiomyopathy (SCM) is a severe complication of sepsis, characterized by profound cardiac dysfunction and high mortality, but its molecular mechanisms remain poorly defined. RNA epigenetic regulation, particularly by the m6A demethylase FTO, has been implicated in cardiovascular disease, yet its role in SCM is unclear. This study aimed to elucidate the function of FTO in SCM by integrating transcriptomic and proteomic analyses. A cecal ligation and puncture (CLP) mouse model was used for in vivo experiments, while lipopolysaccharide (LPS)-stimulated H9c2 cardiomyocytes were emp

Japanese encephalitis virus (JEV) is a neurotropic flavivirus that causes a substantial threat to human health and livestock; however, the epitranscriptomic mechanisms that support its replication remain poorly defined. Here, we identify a proviral host factor C2H2 zinc-finger protein ZNF33B that promotes JEV infection through coupling N6-methyladenosine (m6A) RNA modification to autophagy regulation. Mechanistically, ZNF33B recruits METTL14 to stabilize the METTL3-METTL14 methyltransferase complex, thereby increasing global m6A deposition. Multi-omics analyses reveal that ZNF33B selectively b

Oral squamous cell carcinoma (OSCC) is a common malignant tumor with high metastasis rates and poor prognosis. This study investigated the role of NOP2/Sun RNA methyltransferase family member 2 (NSUN2), a key 5-methylcytosine (m5C) methyltransferase, and m5C methylation in the progression of OSCC, particularly in relation to ferroptosis resistance. NSUN2 is significantly overexpressed in OSCC tissues and cell lines and its high expression correlates with poor prognosis and aggressive tumor characteristics. Knockdown of NSUN2 in ferroptosis-resistant OSCC cells resulted in increased sensitivity

N6-methyladenosine (m6A) is the most prevalent internal modification of cellular and viral RNA and is critical to the regulation of its localization, stability, and translation. Previous studies on the role of m6A during HIV-1 replication have produced conflicting results. Since m6A function can vary dramatically by cell type and state, here we aimed to clarify the role of the m6A machinery during HIV-1 replication in primary CD4+ T cells. Using CRISPR-Cas9 we targeted 46 cellular genes implicated in m6A or 5-methylcytosine (m5C) regulation and measured subsequent HIV-1 replication in primary

An increasing number of individuals are at high risk of type 2 diabetes (T2DM) and its cardiovascular (CV) complications, which challenges healthcare systems with an increased risk of developing CV diseases. Patients with T2DM exhibit a unique cardiac phenotype termed diabetic cardiomyopathy (DCM). DCM usually involves complex and multifactorial pathogenic drivers, including myocardial inflammation, fibrosis, hypertrophy, and early diastolic dysfunction, which potentially evolve into systolic dysfunction and heart failure. There is a lack of effective treatments for DCM on the basis of the com

N6-Methyladenosine (m6A) modification is a common epitranscriptomic mark of eukaryotic RNAs. This modification is installed by a methyltransferase like 3 (METTL3)-METTL14 complex and is eliminated by fat mass and obesity-associated protein (FTO) and AlkB homolog 5 (ALKBH5). Aberrant m6A modification is associated with the development and progression of cancer. Cigarette smoking is a major lifestyle habit and risk factor for lung cancer. This study aimed to clarify the effects of cigarette smoking on the expression of m6A modification-regulating enzymes and the significance of m6A modification
