Population differences in finger-length ratios: ethnicity or latitude?

The relative length of the second and fourth fingers (the 2D:4D ratio) has been taken to be an indicator of prenatal exposure to testosterone, and hence possibly relevant to sexual orientation and other sex-differentiated behaviors. Studies have reported a difference in this ratio between Caucasian males in Britain and in the U.S.: higher average 2D:4D ratios were obtained in Britain. This raises the question of whether differences among different Caucasian gene pools were responsible or whether some environmental variable associated with latitude might be involved (e.g., exposure to sunlight …

A quantitative view of the transcriptome of Schistosoma mansoni adult-worms using SAGE.

BACKGROUND: Five species of the genus Schistosoma, a parasitic trematode flatworm, are causative agents of Schistosomiasis, a disease that is endemic in a large number of developing countries, affecting millions of patients around the world. By using SAGE (Serial Analysis of Gene Expression) we describe here the first large-scale quantitative analysis of the Schistosoma mansoni transcriptome, one of the most epidemiologically relevant species of this genus. RESULTS: After extracting mRNA from pooled male and female adult-worms, a SAGE library was constructed and sequenced, generating 68,238 t

Generation of high-expressing transposon-based stable pools to produce (1 + 1) and (2 + 1) bispecific common light chain antibodies.

Bi- and multispecific antibodies can deliver effective protein-based therapeutics by enabling novel biology. Transient expression of these complex asymmetric antibody formats may become a bottleneck in providing sufficient amounts of high-quality material for preclinical discovery and developability assessment to support candidate selection activities. In contrast, transposase-based stable cell line generation is being recognized as a robust and rapid path to enable the evaluation of product quality from stable pool derived material. Transposase-based systems feature a semi-targeted integratio

First Evidence of Bovine Leukemia Virus in Culicoides Biting Midges and the Stable Fly Stomoxys calcitrans Collected on Cattle Farms in Poland.

Bovine leukemia virus (BLV) is a cell-associated retrovirus of cattle transmitted mainly through the transfer of infected lymphocytes, but the possible role of hematophagous insects in its epidemiology remains incompletely understood. This study investigated BLV DNA in field-collected Culicoides biting midges and Stomoxys calcitrans from cattle farms in Poland. Insects were collected on four farms in northeastern Poland. Culicoides spp. were identified morphologically, classified by gonotrophic status, and pooled by species and status, whereas S. calcitrans were analyzed individually. DNA qual

Characterization of the sex determining region of channel catfish (Ictalurus punctatus) and development of a sex-genotyping test.

Channel catfish is an important species for aquaculture that exhibits a sexually dimorphic growth in favor of males. Genetic sexing and development of sex markers are crucial for the early identification of sex and of particular genotypes (YY males) for the production of all-male population in channel catfish aquaculture. In this study, we sequenced genomic DNA from pools of males and pools of females to better characterize the sex determining region (SDR) of channel catfish and to develop sex-specific markers for genetic sexing. Performing comparative analyses on male and female pooled genomi

Phylogenetic characterization of tick-borne encephalitis virus from Bornholm, Denmark.

The Danish island of Bornholm in the Baltic Sea has been known as a tick-borne encephalitis (TBE) natural focus for more than 60 years. TBE in humans is diagnosed on a regular basis either in inhabitants or tourists of the island. Other areas in Denmark have been suggested as possible risk areas of TBE. Despite the long-known endemicity on Bornholm and the possibility of the virus circulating in other areas, no data on the prevalences of TBE virus (TBEV) in ticks, or adequate molecular characterization and phylogenetic studies are available for the circulating TBEV strains. This study aimed to

Genetic deregulation of REL in germinal center B cells induces generation of a pool of lymphoma precursor cells.

In diffuse large B-cell lymphomas (DLBCLs), gains and amplifications of the 2p15-16 region, which always encompass the REL gene, are mostly restricted to the germinal center (GC) B-cell DLBCL subtype (GCB-DLBCL) for which c-Rel is the pivotal Rel/NF-κΒ subunit. Although REL plays a key role in the GC reaction, its contribution to GCB-DLBCLs remains unclear. To understand the role of REL in the very first steps of GCB transformation, that is, when B cells with deregulated REL are competing with other B cells during chronic antigenic stimulation, we have created a dual-color mouse model that allo

Rescue and sprouting of motoneurons following ventral root avulsion and reimplantation combined with intraspinal adeno-associated viral vector-mediated expression of glial cell line-derived neurotrophic factor or brain-derived neurotrophic factor.

Following avulsion of a spinal ventral root, motoneurons that project through the avulsed root are axotomized. Avulsion between, for example, L2 and L6 leads to denervation of hind limb muscles. Reimplantation of an avulsed root directed to the motoneuron pool resulted in re-ingrowth of some motor axons. However, most motoneurons display retrograde atrophy and subsequently die. Two neurotrophic factors, glial cell line-derived neurotrophic factor (GDNF) and brain-derived neurotrophic factor (BDNF), promote the survival of motoneurons after injury. The long-term delivery of these neurotrophic f

Inappropriately high iron regulatory protein activity in monocytes of patients with genetic hemochromatosis.

In genetic hemochromatosis (GH), excess iron is deposited in parenchymal cells, whereas little iron is found in reticuloendothelial (RE) cells until the later stages of the disease. As iron absorption is inversely related to RE cells stores, a failure of RE to retain iron has been proposed as the basic defect in GH. In RE cells of GH subjects, we examined the activity of iron regulatory protein (IRP), a reliable indicator of the elusive regulatory labile iron pool, which modulates cellular iron homeostasis through control of ferritin (Ft) and transferrin receptor gene expression. RNA-bandshift

Molecular detection of antibiotic-resistant Campylobacter spp. among broiler and layer chickens in Egypt: An outlook from a One Health concept.

Campylobacter is a zoonotic pathogen that poses a public health concern, with poultry acting as the main reservoir of human infection. This study aimed to look into the prevalence and risk factors associated with antibiotic-resistant Campylobacter spp. found in chicken cloacal swabs using direct polymerase chain reaction (PCR) detection, highlighting their importance for public health. In this study, 108 pooled chicken cloacal swabs from 270 broilers and 270 layers were included. Following DNA extraction, the extracted DNA was examined for the presence of Campylobacter 16S rRNA, along with the

Finding novel molecular connections between developmental processes and disease.

Identifying molecular connections between developmental processes and disease can lead to new hypotheses about health risks at all stages of life. Here we introduce a new approach to identifying significant connections between gene sets and disease genes, and apply it to several gene sets related to human development. To overcome the limits of incomplete and imperfect information linking genes to disease, we pool genes within disease subtrees in the MeSH taxonomy, and we demonstrate that such pooling improves the power and accuracy of our approach. Significance is assessed through permutation.

Integrated analysis of Mendelian Randomization and Bayesian colocalization reveals bidirectional causal association between inflammatory bowel disease and psoriasis.

BACKGROUND: Observational studies have suggested an association between inflammatory bowel disease [IBD] and psoriasis. However, the detailed genetic basis, causality, and direction of this association remain unclear. METHODS: Bidirectional two-sample Mendelian Randomization [MR] analysis was conducted using summary statistics from published genome-wide association studies. Bayesian Colocalization and multivariable MR [MVMR] analyses were performed to identify candidate variants and risk genes involved in the shared genetic basis between IBD, psoriasis, and their subtypes. RESULTS: Genetical

Liraglutide Reduces Postprandial Hyperlipidemia by Increasing ApoB48 (Apolipoprotein B48) Catabolism and by Reducing ApoB48 Production in Patients With Type 2 Diabetes Mellitus.

Objective- Treatment with liraglutide, a GLP-1 (glucagon-like peptide-1) agonist, has been shown to reduce postprandial lipidemia, an important feature of diabetic dyslipidemia. However, the underlying mechanisms for this effect remain unknown. This prompted us to study the effect of liraglutide on the metabolism of ApoB48 (apolipoprotein B48). Approach and Results- We performed an in vivo kinetic study with stable isotopes (D8-valine) in the fed state in 10 patients with type 2 diabetes mellitus before treatment and 6 months after the initiation of treatment with liraglutide (1.2 mg/d). Efficient linkage mapping using exome capture and extreme QTL in schistosome parasites.

BACKGROUND: Identification of parasite genes that underlie traits such as drug resistance and host specificity is challenging using classical linkage mapping approaches. Extreme QTL (X-QTL) methods, originally developed by rodent malaria and yeast researchers, promise to increase the power and simplify logistics of linkage mapping in experimental crosses of schistosomes (or other helminth parasites), because many 1000s of progeny can be analysed, phenotyping is not required, and progeny pools rather than individuals are genotyped. We explored the utility of this method for mapping a drug resis

Potential strategies in porcine liver xenotransplantation: A bridge to allotransplantation and beyond.

Advances in both gene editing of organ-source pigs and novel immunosuppressive therapies have substantially propelled the field of heart and kidney xenotransplantation forward, and have helped in overcoming challenges associated with porcine liver transplantation. In this review, the recent experience with porcine liver xenotransplantation in China for both decedents and living patients is discussed. We suggest that porcine liver xenografts play a much greater role in the management of patients with hepatic failure awaiting or following liver allotransplantation. Additionally, we propose that

Epigenetic changes and neurogenesis associated with socio-sexual behaviors.

Epigenetic mechanisms are essential in neurogenesis during development and adulthood. DNA methylation, histone post-translational modifications, and non-coding RNAs regulate gene expression to maintain the neural stem cell pool and direct the fate of newborn neurons by modulating cell proliferation, migration, differentiation, maturation, and survival. Adult neurogenesis exhibits bidirectional interactions with non-social and socio-sexual factors such as sexual behavior, mate recognition, pair bonding, parental behavior, and offspring recognition. Epigenetic mechanism disruption has been impli

Streptococcus pneumoniae Genome Database (SPGDB): a database for strain specific comparative analysis of Streptococcus pneumoniae genes and proteins.

Streptococcus pneumoniae causes pneumonia, septicemia and meningitis. S. pneumoniae is responsible for significant mortality both in children and in the elderly. In recent years, the whole genome sequencing of various S. pneumoniae strains have increased manifold and there is an urgent need to provide organism specific annotations to the scientific community. This prompted us to develop the Streptococcus pneumoniae Genome Database (SPGDB) to integrate and analyze the completely sequenced and available S. pneumoniae genome sequences. Further, links to several tools are provided to compare the p

An adenovirus-vectored nasal vaccine confers rapid and sustained protection against anthrax in a single-dose regimen.

Bacillus anthracis is the causative agent of anthrax, and its spores have been developed into lethal bioweapons. To mitigate an onslaught from airborne anthrax spores that are maliciously disseminated, it is of paramount importance to develop a rapid-response anthrax vaccine that can be mass administered by nonmedical personnel during a crisis. We report here that intranasal instillation of a nonreplicating adenovirus vector encoding B. anthracis protective antigen could confer rapid and sustained protection against inhalation anthrax in mice in a single-dose regimen in the presence of preexis

Linkage disequilibrium mapping of the gene for Hermansky-Pudlak syndrome to chromosome 10q23.1-q23.3.

Hermansky-Pudlak syndrome (HPS) is an autosomal recessive disorder characterized by the triad of tyrosinase-positive oculocutaneous albinism, bleeding diathesis due to storage-pool deficiency of platelets, and a lysosomal ceroid storage disease. The disorder is particularly frequent in Puerto Rico and in an isolated village in the Swiss Alps. We have used a linkage disequilibrium mapping approach to localize the HPS gene in both of these groups to a 0.6 centiMorgan interval in chromosome segment 10q23.1-q23.3. These results indicate that the Puerto Rican and Swiss forms of HPS are either allel
