Potential Targets for Antifungal Drug Discovery Based on Growth and Virulence in Candida albicans.

Fungal infections, especially infections caused by Candida albicans, remain a challenging problem in clinical settings. Despite the development of more-effective antifungal drugs, their application is limited for various reasons. Thus, alternative treatments with drugs aimed at novel targets in C. albicans are needed. Knowledge of growth and virulence in fungal cells is essential not only to understand their pathogenic mechanisms but also to identify potential antifungal targets. This article reviews the current knowledge of the mechanisms of growth and virulence in C. albicans and examines po

Imbalance of renin-angiotensin system in calcium oxalate crystals-induced renal oxidative stress and autophagy.

BACKGROUND: Calcium oxalate (CaOx) stones are the most prevalent urological calculi with high incidence and recurrence. Effective preventive drugs remain lacking because of unclear pathogenesis. Although the imbalance of the dual renin-angiotensin system (RAS) axes is closely linked to oxidative stress and autophagy, its role in CaOx stone formation remains undefined. METHODS: In vitro, human renal tubular epithelial cells were stimulated with CaOx crystals, with siRNA-mediated AT1R or MasR knockdown or pretreatment with Ang Ⅱ, Ang-(1-7), antioxidant Nacetylcysteine (NAC) or autophagy inhibit

Cardiovascular health across childhood and adolescence and proteomic biomarkers in late adolescence.

AIMS: The American Heart Association's Life's Essential 8 cardiovascular health (CVH) construct strongly predicts cardiovascular disease (CVD), yet biological processes associated with early-life CVH trajectories are unclear. We examined associations of CVH score and trajectory parameters across childhood with cardiovascular-related proteins. METHODS AND RESULTS: We derived CVH scores (0-100 points) in 424 children at median ages 3.2, 7.7, 13, and 17.5y. Segmented mixed-effect models estimated three CVH trajectory parameters: timing of inflection when CVH declines and slope before and after i

Insights into the suppression of filamentous bulking by quorum quenching in the activated sludge process with low dissolved oxygen: Endogenous quorum quenching mediated multi-enzymatic degradation of AHLs and carbon-flux redistribution.

Low dissolved oxygen (DO) operation can substantially reduce aeration energy consumption in activated sludge systems but often induces filamentous sludge bulking dominated by Sphaerotilus natans (S. natans). This study isolated endogenous quorum quenching (QQ) bacteria from activated sludge and evaluated their potential to control low-DO bulking. Three endogenous QQ strains were newly isolated, among which Pseudochrobactrum sp. (P. sp.) showed excellent environmental adaptability and broad-spectrum N-acyl homoserine lactone degradation through multi-enzyme synergy, quenching C6-HSL, C8-HSL, an

Dendrobium officinale polysaccharide-spermidine composite hydrogels: Tunable rheological properties and improved in vitro colonic fermentation.

Dendrobium officinale polysaccharide (DOP) and spermidine (Spd) exhibit synergistic anti-aging effects in Caenorhabditis elegans, suggesting potential functional complementarity. Here, a DOP-Spd composite hydrogel (DS) was constructed to integrate structural tunability with fermentation-related functionality. Three weight ratios (18:2, 18:4, and 18:6, DOP:Spd) successfully formed hydrogels. Increasing Spd content enhanced gel strength and thermal stability, as reflected by higher hardness (149.62-353.75 g), chewiness (41.04-119.07), and DTG peak temperature (331.7-334.0 °C). All DS hydrogels e

Transcriptome Analysis Reveals a Potential Response Mechanism of Morchella to Soybean Meal Addition.

Morchella is an edible and medicinal fungus valued for its distinctive flavor and nutritional properties. However, unstable yield and quality remain major constraints on its commercial cultivation. Although soybean meal is used as an organic nutrient supplement in edible mushroom production, its effects on protein accumulation and transcriptional regulation in Morchella remain unclear. This study combined fruiting-body protein measurements with transcriptome analysis to compare soybean meal supplied through soil application or exogenous nutrient bags and to characterize tissue-specific respons

β-carotene attenuates calcium oxalate crystal-induced renal injury through suppression of NF-κB-mediated inflammation.

Calcium oxalate (CaOx) nephrolithiasis represents the most prevalent form of kidney stone disease and is characterized by persistent renal inflammation and tissue damage. β-Carotene, a naturally occurring carotenoid with well-documented anti-inflammatory properties, has demonstrated beneficial effects in a variety of inflammatory conditions; however, its potential role in CaOx crystal-associated renal injury remains largely unexplored. In this study, the effects of β-carotene on CaOx-induced renal damage were evaluated using both cellular and animal models. In human renal proximal tubular epit

Microbial allies in a cotton pest: A descriptive account of associated microbiota dynamics in Dysdercus cingulatus across development.

BACKGROUND: Hemipteran insects harbour several symbiotic partners, mainly bacteria, which play pivotal roles for hosts like dietary provision, support overall physiology, xenobiotic degradation and manipulate/regulate behaviour. Most of these symbionts usually reside and operate from the digestive tracts of the animals. Cotton is one of the major cash crops in India and Dysdercus cingulatus (D. cingulatus) though a secondary pest, is causing significant destruction of cotton bolls, poor lint quality and reduce oil content of seeds. Premature opening of cotton bolls often leads to bacterial and

Unveiling the tissue-specific landscape of nuclear-encoded mitochondrial genes involved in amino acid  metabolism in buffalo.

Mitochondria play a pivotal role in energy production, metabolism, and cellular signaling, serving as key regulators of cellular functions, including differentiation and tissue-specific adaptation. The interplay between mitochondria and the nucleus is crucial for coordinating these processes, particularly through the supply of metabolites for epigenetic modifications that facilitate nuclear-mitochondrial interactions. To investigate tissue-specific mitochondrial adaptations at the molecular level, we conducted RNA sequencing data analyses of kidney, heart, brain, and ovary tissues of female bu

A pilot study on hemodynamically stable isolated chest trauma patients reveals dysregulation of oxidative metabolism.

BACKGROUND: Metabolomic dysregulation precedes clinical deterioration following injury. However, despite receiving comparable treatment, patients with similar injury severity often follow different clinical trajectories and outcomes. METHODS: This prospective cohort study at a level 1 trauma centre screened 4541 acutely injured patients with chest trauma between September 2019 and February 2023. Fifty hemodynamically stable patients with isolated chest trauma were recruited for the final analysis. Urine samples were collected on the injury days 1, 3, and 7. For healthy subjects, the urine sam

Metabolomic disorders caused by an imbalance in the gut microbiota are associated with central precocious puberty.

BACKGROUND: Central precocious puberty (CPP) is characterized by the premature activation of the hypothalamic-pituitary-gonadal axis, resulting in early onset of sexual development. The incidence of CPP has been rising in recent years, with approximately 90% of cases lacking a clearly identifiable etiology. While an association between precocious puberty and gut microbiota has been observed, the precise causal pathways and underlying mechanisms remain poorly understood. The study aims to investigate the potential mechanisms through which gut microbiota imbalances may contribute to CPP. METHOD

Analysis of the metabolic profile of humans naturally exposed to RF-EM radiation.

INTRODUCTION: The world is experiencing exponential growth in communication, especially wireless communication. Wireless connectivity has recently become a part of everyone's daily life. Recent developments in low-cost, low-power, and miniature devices contribute to a significant rise in radiofrequency-electromagnetic field (RF-EM) radiation exposure in our environment, raising concern over its effect on biological systems. The inconsistent and conflicting research results make it difficult to draw definite conclusions about how RF-EM radiation affects living things. OBJECTIVES: This study id

Gut Microbial Perturbation and Host Response Induce Redox Pathway Upregulation along the Gut-Liver Axis during Giardiasis in C57BL/6J Mouse Model.

Apicomplexan infections, such as giardiasis and cryptosporidiosis, negatively impact a considerable proportion of human and commercial livestock populations. Despite this, the molecular mechanisms of disease, particularly the effect on the body beyond the gastrointestinal tract, are still poorly understood. To highlight host-parasite-microbiome biochemical interactions, we utilised integrated metabolomics-16S rRNA genomics and metabolomics-proteomics approaches in a C57BL/6J mouse model of giardiasis and compared these to Cryptosporidium and uropathogenic Escherichia coli (UPEC) infections. Shotgun proteomic profiling of dormant, 'non-culturable' Mycobacterium tuberculosis.

Dormant cells of Mycobacterium tuberculosis, in addition to low metabolic activity and a high level of drug resistance, are characterized by 'non-culturability'-a specific reversible state of the inability of the cells to grow on solid media. The biochemical characterization of this physiological state of the pathogen is only superficial, pending clarification of the metabolic processes that may exist in such cells. In this study, applying LC-MS proteomic profiling, we report the analysis of proteins accumulated in dormant, 'non-culturable' M. tuberculosis cells in an in vitro model of self-ac

Melatonin pretreatment does not modify extrasystolic burden in the rat ischemia-reperfusion model.

The mechanism of reentrant ventricular tachyarrhythmias complicating acute myocardial ischemia is largely based on the interaction between an arrhythmogenic substrate and triggers. Melatonin was proposed as an antiarrhythmic medication and was shown to ameliorate the arrhythmogenic substrate. Also, melatonin provides a sympatholytic effect in different settings and might attenuate ectopic activity, which provides reentry triggers. In the present study, we aimed at evaluating the melatonin effects on cardiac sympathetic activity and the incidence of premature ventricular beats during the episod

Umbilical cord blood metabolome differs in relation to delivery mode, birth order and sex, maternal diet and possibly future allergy development in rural children.

Allergy is one of the most common diseases among young children yet all factors that affect development of allergy remain unclear. In a small cohort of 65 children living in the same rural area of south-west Sweden, we have previously found that maternal factors, including prenatal diet, affect childhood allergy risk, suggesting that in utero conditions may be important for allergy development. Here, we studied if metabolites in the umbilical cord blood of newborns may be related to development of childhood allergy, accounting for key perinatal factors such as mode of delivery, birth order and
