In vitro spectrum of pexiganan activity when tested against pathogens from diabetic foot infections and with selected resistance mechanisms.

Pexiganan, a 22-amino-acid synthetic cationic peptide, is currently in phase 3 clinical trials as a topical antimicrobial agent for the treatment of mild infections associated with diabetic foot ulcers. Bacterial isolates from the 2013 SENTRY Antimicrobial Surveillance Program designated as pathogens from diabetic foot infections (DFI) and Gram-negative and -positive pathogens from various infection types that harbored selected resistance mechanisms/phenotypes were tested against pexiganan in reference cation-adjusted Mueller-Hinton broth. The MIC50 and MIC90 against all organisms tested from

Paediatric bacteraemias in tropical Australia.

AIM: Bacteraemias in children are an important cause of morbidity and mortality. Knowledge of local epidemiology and trends is important to inform practitioners of likely pathogens in the sick child. This study aimed to determine trends over time in pathogenic organisms causing paediatric bacteraemia in North Queensland and to audit a hospital's blood culture results with respect to contamination rate. METHODS: This was a retrospective review of 8385 blood cultures collected from children attending a tertiary centre in North Queensland over a 10-year period (2001-2010). RESULTS: There were 6

Delayed insertion of blood culture bottles into automated continuously monitoring blood culture systems increases the time from blood sample collection to the detection of microorganisms in bacteremic patients.

This study examined the effects on patients with bacteremia of delaying the insertion of a blood culture bottle into an automated, continuously monitoring blood culture system. We investigated the time taken from the collection of blood samples (collection) to the insertion of blood culture bottles inoculated with blood samples into the instrument (insertion), and compared the mean detection time from collection to a positive signal from the instrument with the time between collection and insertion. The study was conducted from January 2003 to December 2004 at Kyoto University Hospital. Insert

Optical Characterization of Gram-Positive and Gram-Negative Biofilms Using Swept-Source Optical Coherence Tomography and Deep Learning.

Biofilms are microbial communities enclosed in a matrix that increases antibiotic tolerance and underlies persistent device-associated infections, making label-free, non-destructive characterization important. Established differences in cell-wall structure between Gram-positive and Gram-negative bacteria are expected to alter light scattering, motivating the hypothesis that the two groups differ in optical attenuation. This proof-of-concept study tested that optical prediction using optical coherence tomography. The optical attenuation coefficient was extracted from cross-sectional images. Gra

In vitro evaluation of the antimicrobial efficacy of Eupatorium odoratum ethanol extract against Gram-positive and Gram-negative bacterial strains.

BACKGROUND: Eupatorium odoratum (EO) L (Siam weed) is a highly invasive species that contains various beneficial active compounds. This stud … ract showed moderate to strong effectiveness against S. aureus, it was less effective against other Gram-positive bacteria and showed no activity against the tested Gram-negative bacteria. Therefore, the ethanolic extract of EO was considered to have potential as an antimicrobial agent, particularly against S. aureus.

Photo-Controlled Reversible Uptake and Release of a Modified Sulfamethoxazole Antibiotic Drug from a Pillar[5]arene Cross-Linked Gelatin Hydrogel.

Pillararene cross-linked gelatin hydrogels were designed and synthesized to control the uptake and release of antibiotics using light. A suite of characterization techniques ranging from spectroscopy (FT-IR, 1H and 13C NMR, and MAS NMR), X-ray crystallographic analysis, scanning electron microscopy (SEM), and thermogravimetric analysis (TGA) was employed to investigate the physicochemical properties of hydrogels. The azobenzene-modified sulfamethoxazole (Azo-SMX) antibiotic was noncovalently incorporated into the hydrogel via supramolecular host-guest interactions to afford the A-hydrogel. Whi

Synthesis, Crystal Structures, Lipophilic Properties and Antimicrobial Activity of 5-Pyridylmethylidene-3-rhodanine-carboxyalkyl Acids Derivatives.

The constant increase in the resistance of pathogenic bacteria to the commonly used drugs so far makes it necessary to search for new substances with antibacterial activity. Taking up this challenge, we obtained a series of rhodanine-3-carboxyalkyl acid derivatives containing 2- or 3- or 4-pyridinyl moiety at the C-5 position. These compounds were tested for their antibacterial and antifungal activities. They showed activity against Gram-positive bacteria while they were inactive against Gram-negative bacteria and yeast. In order to explain the relationship between the activity of the compound…

Neuroprotective Effects of a Novel Demeclocycline Derivative Lacking Antibiotic Activity: From a Hit to a Promising Lead Compound.

The antibiotic tetracycline demeclocycline (DMC) was recently reported to rescue α-synuclein (α-Syn) fibril-induced pathology. However, the antimicrobial activity of DMC precludes its potential use in long-term neuroprotective treatments. Here, we synthesized a doubly reduced DMC (DDMC) derivative with residual antibiotic activity and improved neuroprotective effects. The molecule was obtained by removal the dimethylamino substituent at position 4 and the reduction of the hydroxyl group at position 12a on ring A of DMC. The modifications strongly diminished its antibiotic activity against Gram

Mechanism of the lethal effect of Riparin E against bacterial and yeast strains.

Riparins are alkamides naturally found in the fruits of Aniba riparia (Nees) Mez, but currently synthetic molecules as Riparin E (Rip-E) can be obtained. Potential biological of Rip-E as schistosomicidal agent against Schistosoma mansoni worms, as well as against Staphylococcus aureus strains has already been described. However, the mechanism of action related to antimicrobial activity of Rip-E against bacterial or fungi species has not yet been reported. This study had as objective to evaluate the Rip-E antimicrobial activity against Gram-positive and Gram-negative bacteria, as well as agains

Microbiological patterns in sepsis: what happened in the last 20 years?

Changes in the microbiological characteristics of sepsis have been observed over the past few decades. Infections caused by Gram-positive bacteria and fungi have become more common in some areas, possibly as a consequence of the increased use of intravascular and other medical devices, an increase in invasive procedures and the use of immunosuppressant agents. As observed with other infections there has been an emergence of multiresistant Gram-positive and Gram-negative pathogens. Most of the published data come from wealthy countries in temperate climates, and although many of the same change…

A rapid bacterial labeling method for phenotypic antimicrobial susceptibility testing.

Bacterial infections pose significant diagnostic and treatment challenges due to the increasing prevalence of multidrug-resistant pathogens. Current Antimicrobial Susceptibility Test (AST) workflows, including automated systems, require bacterial isolation which is a time-consuming process. Providing timely AST results can lead to better patient outcomes, effective antibiotic stewardship, and significant cost savings. This study evaluates a novel diagnostic method, the Bacteria Labeling Antibiotic Susceptibility Test (BLAST), using a selection of strains that typically cause urinary tract infe
