Role of CD47 in Hematological Malignancies.

CD47, or integrin-associated protein, is a cell surface ligand expressed in low levels by nearly all cells of the body. It plays an integral role in various immune responses as well as autoimmunity, by sending a potent "don't eat me" signal to prevent phagocytosis. A growing body of evidence demonstrates that CD47 is overexpressed in various hematological malignancies and its interaction with SIRPα on the phagocytic cells prevents phagocytosis of cancer cells. Additionally, it is expressed by different cell types in the tumor microenvironment and is required for establishing tumor metastasis.

The consensus on indications, conditioning regimen, and donor selection of allogeneic hematopoietic cell transplantation for hematological diseases in China-recommendations from the Chinese Society of Hematology.

Allogeneic hematopoietic stem cell transplantation (allo-HSCT) is widely used to treat malignant hematological neoplasms and non-malignant hematological disorders. Approximately, 5000 allo-HSCT procedures are performed in China annually. Substantial progress has been made in haploidentical HSCT (HID-HSCT), pre-transplantation risk stratification, and donor selection in allo-HSCT, especially after the establishment of the "Beijing Protocol" HID-HSCT system. Transplant indications for selected subgroups in low-risk leukemia or severe aplastic anemia (SAA) differ from those in the Western world.

Immunological heterogeneity in human ferritinemia.

Hyperferritinemia in various diseases, mainly hematological, was confirmed by immunological methods. For ferritin detection, anti-human placental ferritin antiserum, anti-human hepatic ferritin antiserum, and anti-human leukemia cell ferritin antiserum were used and the result was compared with each other. Leukemia, malignant lymphoma, multiple myeloma, and aplastic anemia are hematological diseases which showed a positive reaction in this test, among which leukemia showed the highest positivity. Cases of hepatic diseases and non-hematological malignant neoplasms also showed a positive reactio

Beyond the In-Practice CBC: The Research CBC Parameters-Driven Machine Learning Predictive Modeling for Early Differentiation among Leukemias.

A targeted and timely treatment can be a beneficial tool for patients with hematological emergencies (particularly acute leukemias). The key challenges in the early diagnosis of leukemias and related hematological disorders are their symptom-sharing nature and prolonged turnaround time as well as the expertise needed in reporting confirmatory tests. The present study made use of the potential morphological and immature fraction-related parameters (research items or cell population data) generated during complete blood cell count (CBC), through artificial intelligence (AI)/machine learning (ML)

Hematological and Other Cancers in People Using Clozapine: Analysis of Australian Spontaneous Reports Between 1995 and 2020.

BACKGROUND: Recent observational study evidence suggests that clozapine, unlike other antipsychotics, may be associated with a small increased risk of hematological malignancy. This study described characteristics of hematological and other cancers in those taking clozapine reported to the Australian Therapeutic Goods Administration. METHODS: We analyzed public case reports for "clozapine," "Clozaril," or "Clopine" from January 1995 to December 2020 classified as "neoplasm benign, malignant and unspecified" by the Australian Therapeutic Goods Administration. Data on age, sex, dose, clozapine

Rare Primary ALK-Positive Anaplastic Large Cell Lymphoma of the Central Nervous System.

Anaplastic large cell lymphoma (ALCL) is a rapidly growing and aggressive hematological malignancy. We present the case of a 12-year-old adolescent boy with a 2-week history of left iliac fossa and presacral pain radiating to the lower limbs associated with emesis and constipation. Subsequently, the patient developed poorly controlled hypertension and progressive lower limb weakness. Imaging revealed an intradural extramedullary mass at the L1 level, and pathology reported large, atypical cells consistent with ALK -positive ALCL. This case highlights the rarity of isolated intradural extramedu

Liver histopathology in scope of hematological disorders.

Liver involvement is commonly seen in various haematological disorders. They present clinically with elevation of liver enzymes and organomegaly, with or without mass lesions. However, liver involvement may be silent in many hematological disorders or there may be specific findings in liver biopsy that can lead to the diagnosis of clinically inapparent hematological disorders. Present review highlights features of hepatic manifestations in various hematological diseases with special emphasis on histopathological findings. Among RBC disorders, secondary hemochromatosis is the commonest among pa

Merkel cell carcinoma.

Merkel cell carcinoma is a rare, highly aggressive neuroendocrine cutaneous neoplasm with a variable clinical presentation. Histologically, it is a predominantly dermal-based lesion composed of monotonous small round cells with scanty cytoplasm, often difficult to differentiate from small round cell tumors, metastatic small cell carcinoma, blastic hematologic malignancies, and melanoma. The malignant cells express both epithelial and neuroendocrine immunohistochemical markers, a unique feature that helps differentiate this neoplasm from other entities. The pathogenesis of Merkel cell carcinoma

The Emerging Role of Oncodermatology in a Hemato-Oncology Unit in a Tertiary Hospital.

BACKGROUND: Our dermatology department consistently receives the largest proportion of internal on-call referrals from the Hematology-Oncology Department. Individuals with hematological malignancies are particularly susceptible to dermatologic conditions secondary to immunosuppression and multi-agent exposure, which can impact cancer therapy, leading to morbidity and mortality. METHODS: Our primary objective was to analyze the range of dermatologic conditions observed in patients with hematologic malignancies and to identify potential associations with anticancer therapies. We conducted a ret

The murine Irs1 knockout model, generated by the spontaneous Irs1S57X mutation in homozygosity, exhibits high embryonic lethality, limiting studies of adult hematopoiesis.

INTRODUCTION: Insulin Receptor Substrate 1 (IRS1) is differentially expressed in hematological neoplasms suggesting a role in hematopoiesis and neoplastic transformation. Irs1 knockout mice represent a tool to investigate IRS1 function. This study compared hematological parameters of wild-type and heterozygous Irs1S57X mice and assessed fetal lethality in homozygous mice. METHODS: Hematological parameters were analyzed monthly in wild-type and heterozygous mice from 8 to 22 weeks of age. Successive intercrosses failed to yield homozygous knockouts. Fetal lethality was evaluated through timed

Prevalence of hematological malignancies in Africa: A systematic review and meta-analysis.

Hematological malignancies are a class of neoplasms that include a variety of diverse diseases that all develop from and change into lymphatic and bone marrow cells. Hematological malignancies significantly contribute to illness and mortality in African nations. The prevalence of these malignancies has not been evaluated in this continent. The purpose of this systematic review and meta-analysis was to assess the pooled prevalence of hematological malignancies in Africa. From October to November 2023, the electronic databases PubMed, Google Scholar, Web of Science, Research Gate, Embase, and Sc

Risk factors associated with high probability of pulmonary hypertension in patients with myeloproliferative neoplasms.

Myeloproliferative neoplasms (MPNs) are associated with pulmonary hypertension (PH). Recent studies have demonstrated associations between PH and adverse cardiovascular outcomes and MPN disease progression. However, risk factors for development of PH and the pathophysiology of PH in MPNs remain unclear. We conducted an analysis of a multicenter retrospective cohort of patients with MPN with ≥ 2 transthoracic echocardiograms (TTEs) after MPN diagnosis. Patients with low probability of PH on the first TTE after MPN diagnosis were included. Backward stepwise Cox proportional hazards regression mo

How we manage multiple myeloma with clonal hematopoiesis of indeterminate potential (CHIP): a case report.

BACKGROUND: Clonal hematopoiesis of indeterminate potential (CHIP) is characterized by genetic alterations associated with hematologic neoplasms, posing clinical challenges in managing concurrent hematological malignancies. CHIP may complicate the treatment landscape due to its potential to influence disease progression and treatment response. We report a 73-year-old male with multiple myeloma (MM) harboring a CHIP PPM1D mutation, elucidating the complexities and therapeutic considerations in such cases. CASE DESCRIPTION: After four cycles of cyclophosphamide, bortezomib, and dexamethasone th…
