Non-immunologic methods of diagnosis of babesiosis.

The diagnosis of tick-borne diseases such as babesiosis still depends on observing the parasite in the infected erythrocyte. Microscopic observation is tedious and often problematic in both early and carrier infections. Better diagnostic methods are needed to prevent clinical disease, especially when susceptible cattle are being moved into disease enzootic areas. This study evaluates two techniques for early diagnosis of Babesia bovis infections in cattle, DNA probes specific for the organism and fluorescent probes specific for nucleic acid. The radioisotopically labeled DNA probes are used in

Lepidopteran defence droplets - a composite physical and chemical weapon against potential predators.

Insects often release noxious substances for their defence. Larvae of Zygaena filipendulae (Lepidoptera) secrete viscous and cyanogenic glucoside-containing droplets, whose effectiveness was associated with their physical and chemical properties. The droplets glued mandibles and legs of potential predators together and immobilised them. Droplets were characterised by a matrix of an aqueous solution of glycine-rich peptides (H-WG11-NH2) with significant amounts of proteins and glucose. Among the proteins, defensive proteins such as protease inhibitors, proteases and oxidases were abundant. The

The occurrence of schistosomin, an antagonist of female gonadotropic hormones, is a general phenomenon in haemolymph of schistosome-infected freshwater snails.

In haemolymph of Lymnaea stagnalis, parasitized with the digenetic trematode parasite Trichobilharzia ocellata, a neuropeptide (schistosomin) occurs which antagonizes female gonadotropic hormones, e.g. calfluxin (CaFl). By means of an ultracytochemical hormone-assay, the CaFl assay, it was demonstrated that the occurrence of schistosomin is a general phenomenon in schistosome-infected freshwater snails. Haemolymph of the schistosomiasis-transmitting snail species Biomphalaria glabrata and B. pfeifferi, parasitized with Schistosoma mansoni, also appeared to contain an antagonizing factor, i.e.

Recognition and function analysis of MKK6 in immune reaction against Vibrio Parahaemolyticus in Crassostrea hongkongensis.

MKK6 is a key protein kinase that regulates inflammatory response under pathogen infection by activating MAPK signal pathway. However, the function of MKK6 remained unclear within oysters. Our research recognized a novel KK6 gene from C. hongkongensis, which is named ChMKK6. The cDNA of ChMKK6 gene was 2,760 bp in length, containing an ORF of 1,032 bp encoding 344 amino acids, with a conserved S-TKc region and a DVD region. As illustrated by amino acid alignment and phylogenetic tree, ChMKK6 shared the maximal (99.0%) identity showing MKK6 of Crassostrea gigas. As indicated by tissue expressio

C3-CD18-VATPase-ATG16L1 and C3-ATG16L1 axes restrict the escape of microbes through the autophagy-lysosome pathway.

As the key component of the complement system, C3 plays important roles in complement activation to regulate phagocytosis, lyse cells, mediate inflammation, and clear immune complexes. In the present study, CgC3 in the cell-free hemolymph of the Pacific oyster Crassostrea gigas was found to be able to bind various polysaccharides and microbes and then interacted with membrane receptor CgCD18 to mediate the entry of the CgC3-coated Vibrio into hemocytes. CgATPV1D in hemocytes sensed the CgC3-coated Vibrio vacuole and recruited CgATG16L1. The free CgC3 in hemocytes could also recognize and bind

Oral RNAi in Euschistus heros: Evaluation of Gene Silencing Using Cationic Polymer-Stabilized dsRNA.

Euschistus heros, a key pest of soybean in the Neotropical region, represents a growing challenge to sustainable agricultural production. With the urgent need for more targeted and environmentally safe control strategies, RNA interference (RNAi) emerges as an innovative tool. This study investigated the gene-silencing potential of RNAi targeting V-ATPase and act, two genes involved in essential cellular processes. The sensitivity of E. heros to RNAi in adult and immature stages was evaluated by microinjection of dsRNA, as well as the ex-vivo degradation of double-stranded RNA (dsRNA) by nuclea

Shrimp Shapes a Nitrite Tolerance Trait via Regulating Autophagy and Apoptosis.

Nitrite is a significant toxic substance that causes acute toxicity in aquatic animals. However, the molecular mechanisms underlying nitrite tolerance remain largely unidentified. Here, we investigated the physiological and transcriptomic profiles of nitrite-sensitive and nitrite-tolerant families of the white shrimp Penaeus vannamei under nitrite stress. Exposure to a lethal concentration of nitrite for 72 h caused higher mortality, higher nitrite accumulation, and more severe physiological damage in both the hemolymph and hepatopancreas of nitrite-sensitive families than in nitrite-tolerant

Interactions between Lipopolysaccharide and Peptide Bacteriocin BacSp222 Influence Their Biological Activities.

This study describes the interactions between two different pro-inflammatory factors produced by bacteria, lipopolysaccharide (LPS) from Gram-negative bacteria and the peptide BacSp222 produced by a Gram-positive zoonotic strain, Staphylococcus pseudintermedius 222. We demonstrate that the mentioned molecules interact, forming a complex, and this phenomenon selectively reduces their biological activities in vitro and in vivo. Specifically, the levels of tumor necrosis factor (TNF) and nitric oxide (NO) produced by monocyte-macrophage cells were lower in samples treated with both LPS and BacSp2

Killing of Plasmodium Sporozoites by Basic Amphipathic α-Helical Fusion Peptides.

Membranolytic molecules constitute the first line of innate immune defense against pathogenic microorganisms. Plasmodium sporozoites are potentially exposed to these cytotoxic molecules in the hemolymph and salivary glands of mosquitoes, as well as in the skin, blood, and liver of the mammalian host. Here, we show that sporozoites are resistant to bacteriolytic concentration of cecropin B, a cationic amphipathic antimicrobial insect peptide. Intriguingly, anti-tumoral cell-penetrating peptides derived from the anti-apoptotic protein AAC11 killed P. berghei and P. falciparum sporozoites. Using

Whole genome enrichment approach for genomic surveillance of Toxoplasma gondii.

Pathogenic bacteria, viruses, fungi, and protozoa can cause food and waterborne diseases. Surveillance methods must therefore screen for these pathogens at various stages of water distribution and of food from production to consumption. Detection using nucleic acid amplification methods offer rapid identification, but such methods have limited utility for characterizing populations, variant types or virulence traits of pathogens. Whole genome sequencing (WGS) can be used to determine this information. However, pathogens must be isolated and cultured to yield sufficient DNA for WGS, which is la

Exorista sorbillans (Diptera: Tachinidae) parasitism shortens host larvae growth duration by regulating ecdysone and juvenile hormone titers in Bombyx mori (Lepidoptera: Bombycidae).

The tachinid fly, Exorista sorbillans, is a notorious ovolarviparous endoparasitoid of the silkworm, Bombyx mori, causing severe damage to silkworm cocoon industry. Silkworm larvae show typically precocious wandering behavior after being parasitized by E. sorbillans; however, the underlying molecular mechanism remains unexplored. Herein, we investigated the changes in the levels of 20-hydroxyecdysone (20E) and juvenile hormone (JH) titer, and they both increased in the hemolymph of parasitized silkworms. Furthermore, we verified the expression patterns of related genes, which showed an upregul

Correlation between populations of Rhodnius and presence of palm trees as risk factors for the emergence of Chagas disease in Amazon region, Brazil.

Among the states that comprise the legally defined Amazon region of Brazil, Pará has presented the highest occurrences of acute cases of Chagas disease over the last two decades. These cases have been attributed to consumption of fruits from native palm trees. In surveys in rural and wild areas of the municipality of Oriximiná, Pará, triatomine fauna, their main ecotopes and the infection rate due to Trypanosoma cruzi were identified using active and passive search methods: manual capture and Noireau traps, respectively. A total of 582 ecotopes were surveyed using 1496 Noireau traps. Out of 44

Evaluation of parenteral drugs for anesthesia in the blue crab (Callinectes sapidus).

The objective of this study was to evaluate the efficacy and safety of several parenteral anesthetics in blue crabs (Callinectes sapidus). Thirty-one animals were administered one or more of the following drugs by injection into the hemolymph (i.v.) through an arthrodial membrane: etomidate, ketamine, lidocaine, pentobarbital, propofol, tiletamine-zolazepam, xylazine, and ketamine-xylazine. A subset of crabs received intracardiac ketamine. Etomidate had no effect. Lidocaine effects were ultrashort (<3 min). Pentobarbital had prolonged inductions (2 min) and often caused violent excitement and

Identification and characterization of homologues of vertebrate beta-thymosin in the marine mollusk Aplysia californica.

The beta-thymosins have been known as actin-sequestering proteins, but now are recognized as molecules with multiple and diverse intracellular and extracellular functions. Two closely related proteins, beta-thymosin(His) and beta-thymosin(Gln), have been de novo sequenced by top-down mass spectrometry in the common neurobiology model, Aplysia californica. As determined by nanoelectrospray quadrupole-enhanced Fourier-Transform mass spectrometry with collisionally activated and electron-capture dissociations, both of these Aplysia beta-thymosins are acetylated and differ by a single residue in t

Lipophorin as a yolk protein precursor in the mosquito, Aedes aegypti.

We examined expression of the lipophorin (Lp) gene, lipophorin (Lp) synthesis and secretion in the mosquito fat body, as well as dynamic changes in levels of this lipoprotein in the hemolymph and ovaries, during the first vitellogenic cycle of females of the yellow fever mosquito, Aedes aegypti. Lipophorin was purified by potassium bromide (KBr) density gradient ultracentrifugation and sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Polyclonal antibodies were produced against individual Lp apoproteins, apolipoprotein-I (apoLp-I) and apolipoprotein-II (apoLp-II), with mole

Mosquito cathepsin B-like protease involved in embryonic degradation of vitellin is produced as a latent extraovarian precursor.

Here we report identification of a novel member of the thiol protease superfamily in the yellow fever mosquito, Aedes aegypti. It is synthesized and secreted as a latent proenzyme in a sex-, stage-, and tissue-specific manner by the fat body, an insect metabolic tissue, of female mosquitoes during vitellogenesis in response to blood feeding. The secreted, hemolymph form of the enzyme is a large molecule, likely a hexamer, consisting of 44-kDa subunits. The deduced amino acid sequence of this 44-kDa precursor shares high similarity with cathepsin B but not with other mammalian cathepsins. We ha

Synergistic role of CSPBP and SGS1 in sporozoite entry into Aedes aegypti salivary glands.

Vector-borne diseases, particularly malaria, continue to pose a major global health challenge, underscoring the urgent need for innovative and sustainable vector control strategies. Among emerging solutions, genetic control holds great promise for interrupting disease transmission at its source. Malaria transmission relies on the parasite's ability to overcome multiple barriers within the mosquito and ultimately invade the salivary glands during the sporozoite stage. Using Plasmodium gallinaceum as a model parasite, this study investigates two Aedes aegypti proteins, the salivary gland surface

Cellular Uptake and Tissue Retention of Microplastics in Black Soldier Fly Larvae.

Microplastic pollution is a pressing global concern, yet its immunotoxicological impacts on invertebrates remain poorly understood. The Black Soldier Fly (Hermetia illucens) larva has gained attention for its role in waste management and potential bioremediation, making it essential to evaluate its interactions with microplastics. In this study, fluorescent carboxylate-modified polystyrene microbeads were directly injected into the hemocoel of larvae to bypass gut-associated variables and investigate systemic immune responses. Experimental groups were analyzed at multiple time points (1 h, 6 h

Vangl2 regulates intercellular junctions by remodeling actin-based cytoskeleton through the Rock signaling pathway during spermatogenesis in Eriocheir sinensis.

As a key planar cell polarity protein, Van Gogh-like 2 (Vangl2) is essential for mammalian spermatogenesis. As a decapod crustacean, Eriocheir sinensis exhibits distinct spermatogenic processes due to its unique seminiferous tubule morphology and hemolymph-testis barrier (HTB). To determine whether Vangl2 performs analogous functions in E. sinensis, we identified the Es-Vangl2. Es-Vangl2 exhibited high expression and wide distribution in the testes, indicating its crucial involvement in spermatogenesis. Following targeted knockdown of Es-Vangl2in vivo, the structure of seminiferous tubules was
