Rapid formulation alginate dressing with sustained nitric oxide release: a dual-action approach for antimicrobial and angiogenic benefits.

Infected chronic wounds resulting from bacterial, viral, or other microbial invasion disrupt the normal wound-healing process, often prolonging recovery and requiring surgical intervention. A high nitric oxide (NO) concentration damages bacterial DNA and disrupts metabolic functions, indicating its effective antibacterial potential, whereas a low NO concentration exerts detectable provascularization activity. These properties render NO an effective treatment agent for infected chronic wounds. However, challenges remain in generating NO efficiently, controlling its dosage, and selecting an appr

Enhanced cytotoxicity of cisplatin-loaded Brij S100-alginate-taurine nanogels against HeLa cervical cancer cells.

Water-insoluble anti-cancer drug delivery systems play a vital role in enhancing the effectiveness, stability, and selectivity of chemotherapeutic agents. By improving these properties, such systems offer better clinical outcomes, reduced systemic toxicity, and expanded therapeutic options, particularly for cancers that exhibit resistance to conventional treatments. In this context, nanogel-based delivery platforms constructed from Brij S100 grafted with alginate have demonstrated considerable promise. Two formulations were studied: Brij-Alg and Brij-Alg-Tau, the latter featuring surface modif

Retinoic acid-loaded alginate microspheres as a slow release drug delivery carrier for intravitreal treatment.

Pharmacotherapy were the mainstream methods for the treatment of proliferative vitreoretinopathy (PVR), one of the leading causes of blindness. Due to the negligible side-effects, retinoic acid (RA) was considered to be a promising candidate drug. However, it was still a challenge to deliver drugs into vitreous with both long-term efficiency and biosafety. Sodium alginate was considered to be an excellent drug carrier owing to its controlled size, good biocompatibility and degradability. Based on a one-time synthetic strategy of preparing RA-sodium alginate microspheres, the study was aimed to

A Retrospective Study of Pilonidal Sinus Healing by Secondary Intention Using Negative Pressure Wound Therapy Versus Alginate or Gauze Dressings.

Pilonidal sinus (PS) disease is an inflammatory skin and subcutaneous tissue condition that presents with infection, acute abscess, chronic discharging wounds, and/or pain. Surgery with open healing by secondary intention typically is used to achieve the fastest healing time with minimal recurrence rates. A retrospective analysis was conducted of data extracted from the medical records of 73 consecutive patients who had symptomatic natal cleft PS over a 10-year period to compare use of NPWT to alginate-based/gauze daily dressing (DD) changes in terms of healing time and recurrence. Variables e

Removal of endocrine disrupting compounds from wastewater by microalgae co-immobilized in alginate beads.

Microalgae systems have been found to be efficient for removing microcontaminants from wastewater effluents, but the effectiveness of immobilized microalgae for removing endocrine disrupting compounds (EDCs) has not yet been addressed. This paper assesses the effect of free and immobilized microalgae on removal efficiency for 6 EDCs by mixing them in 2.5 L reactors with treated wastewater. The experimental design also included control reactors without microalgae. After 10 days of incubation, 64 and 89% of the NH4-N and 90 and 96% of total phosphorous (TP) had been eliminated in the free microa

Regulation of Complement and Contact System Activation via C1 Inhibitor Potentiation and Factor XIIa Activity Modulation by Sulfated Glycans - Structure-Activity Relationships.

The serpin C1 inhibitor (C1-INH) is the only regulator of classical complement activation as well as the major regulator of the contact system. Its importance is demonstrated by hereditary angioedema (HAE), a severe disease with potentially life-threatening attacks due to deficiency or dysfunction of C1-INH. C1-INH replacement is the therapy of choice in HAE. In addition, C1-INH showed to have beneficial effects in other diseases characterized by inappropriate complement and contact system activation. Due to some limitations of its clinical application, there is a need for improving the effica

An in vitro study of peptide-loaded alginate nanospheres for antagonizing the inhibitory effect of Nogo-A protein on axonal growth.

The adult mammalian central nervous system has limited ability to regenerate after injury. This is due, in part, to the presence of myelin-associated axon growth inhibitory proteins such as Nogo-A that bind and activate the Nogo receptor, leading to profound inhibition of actin-based motility within the growing axon tip. This paper presents an in vitro study of the use of a Nogo receptor-blocking peptide to antagonize the inhibitory effect of Nogo-A on axon growth. Alginate nanospheres were fabricated using an emulsion technique and loaded with Nogo receptor-blocking peptide, or with other mod

Function and viability of human islets encapsulated in alginate sheets: in vitro and in vivo culture.

Islet encapsulation offers an immune system barrier for islet transplantation, and encapsulation within an alginate sheetlike structure offers the ability to be retrievable after transplanted. This study aims to show that human islets encapsulated into islet sheets remain functional and viable after 8 weeks in culture or when transplanted into the subcutaneous space of rats. Human islets were isolated from cadaveric organs. Dissociation and purification were done using enzymatic digestion and a continuous Ficoll-UWD gradient. Purified human islets were encapsulated in alginate sheets. Human Is

Pseudomonas aeruginosa alginate promotes Burkholderia cenocepacia persistence in cystic fibrosis transmembrane conductance regulator knockout mice.

Pseudomonas aeruginosa, a major respiratory pathogen in cystic fibrosis (CF) patients, facilitates infection by other opportunistic pathogens. Burkholderia cenocepacia, which normally infects adolescent patients, encounters alginate elaborated by mucoid P. aeruginosa. To determine whether P. aeruginosa alginate facilitates B. cenocepacia infection in mice, cystic fibrosis transmembrane conductance regulator knockout mice were infected with B. cenocepacia strain BC7 suspended in either phosphate-buffered saline (BC7/PBS) or P. aeruginosa alginate (BC7/alginate), and the pulmonary bacterial load

In vivo experience with natural scaffolds for myocardial infarction: the times they are a-changin'.

Treating a myocardial infarction (MI), the most frequent cause of death worldwide, remains one of the most exciting medical challenges in the 21st century. Cardiac tissue engineering, a novel emerging treatment, involves the use of therapeutic cells supported by a scaffold for regenerating the infarcted area. It is essential to select the appropriate scaffold material; the ideal one should provide a suitable cellular microenvironment, mimic the native myocardium, and allow mechanical and electrical coupling with host tissues. Among available scaffold materials, natural scaffolds are preferable

Plant immunity triggered by engineered in vivo release of oligogalacturonides, damage-associated molecular patterns.

Oligogalacturonides (OGs) are fragments of pectin that activate plant innate immunity by functioning as damage-associated molecular patterns (DAMPs). We set out to test the hypothesis that OGs are generated in planta by partial inhibition of pathogen-encoded polygalacturonases (PGs). A gene encoding a fungal PG was fused with a gene encoding a plant polygalacturonase-inhibiting protein (PGIP) and expressed in transgenic Arabidopsis plants. We show that expression of the PGIP-PG chimera results in the in vivo production of OGs that can be detected by mass spectrometric analysis. Transgenic plan
