Rats given dopamine-depleting brain lesions as neonates are subsensitive to dopaminergic antagonists as adults.

Extensive damage to central dopamine (DA)-containing neurons are known to produce akinesia and sensory neglect when the lesions are made in adult rats. Similar behavioral impairments occur when dopaminergic function is disrupted temporarily by DA receptor blocking agents, and brain-damaged rats are particularly sensitive to the effects of those drugs. The present experiments offer a striking contrast to these well-accepted findings that central DA-containing neurons are critical to the initiation of voluntary movement. After near-total destruction of the dopaminergic neurons in 3-day-old rats,

The roles of brain noradrenaline and dopamine in the anorectic activity of diethylpropion in rats: a comparison with d-amphetamine.

The anorectic activity of diethylpropion and d-amphetamine was studied in rats subjected to various treatments known to affect brain monoamines. The effect of diethylpropion, like that of d-amphetamine, was completely prevented by a lesion of the ventral noradrenergic bundle, which selectively decreases brain noradrenaline, but was not significantly modified in desipramine pretreated rats by an intraventricular injection of 6-hydroxydopamine, a condition decreasing only dopamine. Pretreatment with penfluridol significantly reduced the effect of d-amphetamine but not that of diethylpropion. A n

Ultrastructrual effects of chemical sympathectomy on brown adipose tissue.

Adult rats maintained at 20-22 degrees C, were exposed to 4 degrees C for 30-60 min and injected with 50 or 100 mg/kg 6-hydroxydopamine (6-OHDA) in an attempt to achieve a similar degree of chemical sympathectomy of nerves terminating among the adipocytes and the smooth muscle cells of the blood vessels in the interscapular brown adipose tissue (BAT). After 1, 4 or 10 days the pads of BAT were removed and small sections from each pad prepared for electron microscopy; the remaining tissue was used for noradrenaline (NA) analysis for fluorescence histochemistry. Ultrastructural observations show

Chemical sympathectomy increases the innate immune response and decreases the specific immune response in the spleen to infection with Listeria monocytogenes.

Many investigators have shown that ablation of the sympathetic nervous system (SNS) with 6-hydroxydopamine (6-OHDA) can alter cell-mediated and humoral immune responses to antigenic challenge. Fewer studies have examined 6-OHDA-induced changes in natural immunity. In this study, we have examined the effect of chemical sympathectomy on the nonspecific and specific phases of the response to infection with Listeria monocytogenes. Sympathectomy decreased splenic bacterial loads 3 and 5 days post-infection and increased splenic neutrophils 3 days post-infection. Sympathectomy decreased splenocyte n

Denervation-induced supersensitivity to forskolin and pinacidil is not related to changes in the adenylate cyclase transduction pathway in the rabbit femoral artery.

1. The present study investigates whether chemical sympathectomy compromises the relaxation of the rabbit femoral artery precontracted with serotonin. The vasodilating agents promoted cyclic adenosine monophosphate (cAMP) accumulation or opening of the potassium channels. The effect of denervation on the adenylyl cyclase transduction pathway was also studied. 2. 6-Hydroxydopamine treatment did not impair the relaxation to adenosine, 8-bromoadenosine-cAMP (a membrane-permeable analog of cAMP) and 3-isobutyl-1-methylxanthine (a cAMP phosphodiesterase inhibitor). Moreover, denervation enhanced th

Norepinephrine depletion in the amygdala inhibits CRF-41, ACTH, and corticosterone responses following photic simulation.

The effects of amygdaloid norepinephrine depletion by 6-hydroxydopamine on changes in corticotropin releasing factor-41 (CRF-41) and serum adrenocorticotropic hormone (ACTH) and corticosterone levels, following neural stimuli were investigated. In intact animals, photic or acoustic stimulation caused CRF-41 depletion from the median eminence and a rise in serum ACTH and corticosterone levels. In rats with amygdalar norepinephrine depletion there were no changes in basal CRF-41, ACTH, or corticosterone levels. However, the above responses of the hypothalamo-pituitary adrenocortical axis were bl

Nigrostriatal dopamine system mediates baroreflex sensitivity in rats.

Both sham-operated rats and rats with lesions of the nigrostriatal dopamine (DA) pathway were subjected to increasing or decreasing carotid sinus pressure (respectively produced by phenylephrine injection or bilateral carotid occlusion) and their striatal DA release and baroreflex responses were compared. In sham-operated rats, phenylephrine produced both increased striatal DA release and decreased heart rate, while bilateral carotid occlusion produced the opposite effects. Both the striatal DA release and baroreflex responses produced by phenylephrine or carotid occlusion were attenuated by l

Accumbens microinjection of LY 171555 and sulpiride: effects on circling behaviour, in the rat.

In rats with unilateral 6-hydroxydopamine lesions of the substantia nigra (pars compacta) and amphetamine i.p. some parameters of the rotational activity, following intra-accumbens microinjection of D2 agonist (LY 171555) and antagonist (sulpiride), have been analyzed. An increase in the number of turns was observed with LY 171555, whereas sulpiride determined a decrease. The type of circling was also analyzed: D2 agonist induced a very close turn around the central axis of the animal and D2 antagonist a close turn in the centre of the cage. Data were compared with control condition, which exh

Adenosine release may mediate spinal analgesia by morphine.

Spinal analgesia produced by morphine is blocked by methylxanthine adenosine receptor antagonists. In biochemical studies, morphine releases adenosine from spinal cord synaptosomes prepared from the dorsal spinal cord, as well as from the intact spinal cord in vivo. Adenosine release is reduced by intrathecal and neonatal pretreatment with capsaicin but not by intrathecal pretreatment with 6-hydroxydopamine or 5,7-dihydroxytryptamine, indicating that adenosine originates from small-diameter primary afferent neurons but not descending monoaminergic pathways. Immunomodulatory drugs alleviate l-dopa-induced dyskinesia in a rat model of Parkinson's disease.

BACKGROUND: Thalidomide and closely related analogues are used clinically for their immunomodulatory and antiangiogenic properties mediated by the inhibition of the proinflammatory cytokine tumor necrosis factor α. Neuroinflammation and angiogenesis contribute to classical neuronal mechanisms underpinning the pathophysiology of l-dopa-induced dyskinesia, a motor complication associated with l-dopa therapy in Parkinson's disease. The efficacy of thalidomide and the more potent derivative 3,6'-dithiothalidomide on dyskinesia was tested in the 6-hydroxydopamine Parkinson's disease model. METHODS

Reversible neurochemical changes mediated by delayed intrastriatal glial cell line-derived neurotrophic factor gene delivery in a partial Parkinson's disease rat model.

BACKGROUND: Efficient protection of dopaminergic neurons against a subsequent 6-hydroxydopamine lesion by glial cell line-derived neurotrophic factor (GDNF) gene delivery has been demonstrated. By contrast, the neurorestorative effects of GDNF administered several weeks after the toxin have been less characterized. In particular, whether these were permanent or dependent on the continuous presence of GDNF remains elusive. METHODS: A tetracycline-inducible adeno-associated virus (AAV)-1 vector expressing human GDNF cDNA was administered unilaterally in the rat striatum 5 weeks after 6-hydroxyd

Time-dependent changes in striatal xCT protein expression in hemi-Parkinson rats.

Altered glutamate signaling is associated with Parkinson's disease. To study the involvement of the cystine/glutamate antiporter in the pathogenesis of Parkinson's disease, we developed new polyclonal antibodies recognizing xCT, the specific subunit of this antiporter. The striatal xCT protein expression level was investigated in a hemi-Parkinson rat model, using semiquantitative western blotting. We observed time-dependent changes after a unilateral 6-hydroxydopamine lesion of the nigrostriatal pathway with increased expression levels in the deafferented striatum after 3 weeks. Twelve weeks p

Neural repair strategies for Parkinson's disease: insights from primate models.

Nonhuman primate models of Parkinson's disease (PD) have been invaluable to our understanding of the human disease and in the advancement of novel therapies for its treatment. In this review, we attempt to give a brief overview of the animal models of PD currently used, with a more comprehensive focus on the advantages and disadvantages presented by their use in the nonhuman primate. In particular, discussion addresses the 6-hydroxydopamine (6-OHDA), 1-methyl-1,2,3,6-tetrahydopyridine (MPTP), rotenone, paraquat, and maneb parkinsonian models. Additionally, the role of primate PD models in the

Chemical sympathectomy and clorgyline-induced stimulation of rat pineal melatonin synthesis.

The response to administration of the specific monoamine oxidase A (MAO-A) blocker clorgyline was investigated in adult male Sprague-Dawley rats which were sympathectomized by injection of the false neurotransmitter 6-hydroxydopamine as newborns. In intact animals which served as controls, the contents of pineal indoles melatonin, serotonin, 5-hydroxytryptophan were augmented, and the content of 5-hydroxyindoleacetic acid decreased 90 min following clorgyline injections when compared to saline receiving rats. Sympathectomized animals exhibited similar responses but these were less pronounced.

Noradrenergic modulation of serotonin synthesis and metabolism. II. Stimulation by 3-isobutyl-1-methylxanthine.

The hypothesis that norepinephrine neurons facilitate serotonin metabolism was tested by employing the adenosine antagonist 3-isobutyl-1-methylxanthine (IBMX) as a pharmacological probe to enhance central noradrenergic metabolism. IBMX elevated brain concentrations of 3-methoxy-4-hydroxyphenylglycol and 5-hydroxyindoleacetic acid and increased the accumulation rates of 3,4-dihydroxyphenylalanine and 5-hydroxytryptophan. Maximal effects were observed 3 hr after drug administration, with 14 mg/kg of IBMX. The effects of IBMX on serotonin metabolism were observed in cortex, striatum and hippocamp

Cardiovascular effects of the novel histamine H2 receptor agonist amthamine: interaction with the adrenergic system.

The cardiovascular effects of the new histamine H2 receptor agonist amthamine were studied in the anaesthetized rat, with particular reference to a possible interaction with the adrenergic system. Amthamine (0.03-3 mumol/kg i.v.) caused vasodepressor responses which were antagonized by famotidine (3 mumol/kg i.v.). At higher doses (30-100 mumol/kg i.v.), amthamine induced a modest increase in the mean arterial pressure, which was significantly enhanced by the blockade of H2 receptors and significantly reduced by the alpha 2 adrenoceptor antagonist yohimbine (1 mumol/kg i.v.). The vasopressor r

Are large, fluorescent spots in aged mouse brain due to lesioning of catecholamine neurons?

Age-related, large intensely fluorescent (LIF) catecholamine-containing spots were discovered in mouse brain. The similarity of their appearance to "piled-up" fluorescent material observed in catecholamine (CA) nerve tracts after 6-hydroxydopamine (6-OHDA) lesions suggests that LIF spots may be due to accumulation of CA in neurons whose distal portions are undergoing degeneration. When senescent mouse brain were examined longitudinal to monoaminergic tracts by the Falck-Hillarp histofluorescence method, some elongated, large fluorescent structures were observed. This suggests an axonal swellin

Studies on muscarinic receptors in mouse and rat hypothalamus: a comparison of sex and cyclical differences.

The ligand binding properties of muscarinic receptors present in whole hypothalamus as well as in its three regions (preoptic area, median and posterior regions) were studied in male rats and mice, as well as in female rats during various stages of the estrous cycle, using the tritiated antagonist N-methyl-4-piperidyl benzilate. Kinetic and equilibrium analysis of antagonist binding as well as competition experiments with agonists, were used to probe the nature of the binding sites and possible differences between specific muscarinic sites in the various preparations. We could detect differenc
