Thyroid function in children with growth hormone deficiency, either idiopathic or caused by diseases of the central nervous system.

Thyroid function was assessed in thirty two patients with growth hormone deficiency (GHD) by clinical examination and by measurement of T4-levels, free T4-indices, basal TSH values and TSH responses to TRH (100 mu/m2). Sixteen patients (50%) were hypothyroid. In thirteen patients, the endocrine disorders were considered to be of hypothalamic origin. Ten of them showed prolonged responses to TRH and in the other three the responses were exaggerated. In three patients hypothyroidism was due to a primary pituitary disorder. Sixteen patients were euthyroid although three of them showed impaired TS

The endocrinology of anorexia nervosa and bulimia nervosa.

Considerable evidence exists of hypothalamic dysfunction in patients with anorexia nervosa and bulimia nervosa. This dysfunction is reflected in disturbances of endocrine function including abnormalities of gonadotropin, growth hormone, and corticotropin-releasing hormone secretion. Whereas these disturbances are generally reversed with nutritional rehabilitation and weight restoration, it is not evident to what extent nutritional factors are the primary etiology or whether they unmask an otherwise existing but compensated central disturbance. Similarly, endocrine disturbances may be a final c

Studies of hypothalamic function in Huntington's chorea.

In eight patients with classical Huntington's chorea hypothalamic function was assessed by the insulin tolerance test, the thyrotrophin releasing hormone test, the gonadotrophin releasing hormone test and water deprivation and the results compared with those of 10 control subjects. All patients ceased to have choreiform movements for approximately 60 minutes during the insulin tolerance test. Four of the patients failed to show clinical features of stress in response to hypoglycaemia. The fasting blood glucose level and blood glucose response to insulin were similar for the two groups. However

Thyroid axis activity and dopamine function in depression.

BACKGROUND: Several lines of evidence suggest alterations in both hypothalamic-pituitary-thyroid (HPT) axis and dopamine (DA) function in depressed patients. However, the functional relationships between HPT and DA systems have not been well defined. METHODS: We examined thyrotropin (TSH) response to 0800 h and 2300 h protirelin (TRH) challenges, and adrenocorticotropic hormone (ACTH), cortisol and growth hormone (GH) responses to apomorphine (APO, a DA receptor agonist), in 58 drug-free DSM-IV major depressed inpatients without a suicidal behavior, and 22 healthy hospitalized controls. GH Stimulation testing is unnecessary in patients with hypothalamic-pituitary disease, preserved pituitary function, and IGF-I SDS ≥ 0.

PURPOSE: In adults with hypothalamic-pituitary disease, the presence of at least three additional pituitary hormone deficiencies, with or without markedly reduced IGF-I levels, is considered diagnostic of growth hormone deficiency (GHD) without the need for stimulation testing. In all other cases, dynamic testing is required despite its well-recognized limitations. To date, no formal strategy has been proposed to exclude GHD without performing stimulation tests. This study aims to develop a probabilistic model to estimate the likelihood of GHD in adults with hypothalamic-pituitary disease but

Growth Without Growth Hormone in Sellar Germ Cell Tumors: Clinical Characteristics and Predictive Factors.

OBJECTIVE: Sellar germ cell tumors (GCTs) commonly cause growth hormone deficiency (GHD) and impaired growth in children. A subset of patients maintains near-normal growth despite GHD, a phenomenon termed growth without growth hormone (GWGH), with unclear mechanisms. This study aimed to characterize the clinical features and predictors of GWGH in pediatric sellar GCTs. METHODS: Patients with sellar GCTs and GHD presenting to our hospital between July 2021 and December 2024, with hypogonadotropic hypogonadism or prepubertal status, were included. Patients with annual growth ≥5 cm were classifi

Modulating the RFamide-related peptide-3/G protein-coupled receptor 147 signaling pathway with nourishing Yin-removing fire herbal mixture to alleviate precocious puberty in female rats: An experimental study.

BACKGROUND: Precocious puberty (PP) involves early activation of the hypothalamic gonadotropin-releasing hormone (GnRH) generator. The RFamide-related peptide/G protein-coupled receptor 147 (RFRP3/GPR147) signaling pathway is vital in inhibiting GnRH and delaying puberty onset. The nourishing Yin-removing fire (NYRF) herbal mixture has shown promising results in treating PP. OBJECTIVE: This study aimed to assess the impact of the NYRF herbal mixture on the RFRP3/GPR147 signaling pathway in the hypothalamus and its potential in alleviating PP in female rats. MATERIALS AND METHODS: In a contro

Using endocrine profiles to explore subgroups among transdiagnostic neurodiverse children and adolescents.

There is considerable heterogeneity in neurodevelopmental disorders (NDDs) that complicates research and clinical support. Although the biological mechanisms of NDDs remain uncertain, emerging studies suggest potential roles of hormone pathways as gonadal, thyroid, glucocorticoid and growth hormones all contribute to brain development. Alterations to these pathways and imbalances of hormones may modulate neural and behavioural development and contribute to the biology of NDDs. Importantly, the field lacks transdiagnostic investigations of endocrine-related phenotypic characteristics. The prese

Hypothalamic-pituitary-gonadal axis response to photoperiod changes in female guanacos (Lama guanicoe).

The guanaco, a wild South American camelid, is renowned for its remarkable resilience to extreme conditions. Despite this, little is known about how reproductive hormones in female camelids are influenced during their seasonal breeding period, which occurs during long photoperiod. To explore this, the study investigated the response of the hypothalamic-pituitary-gonadal axis in female guanacos during short days (10L:14D; July) and long days (16L:8D; December) in the Mediterranean ecosystem (33°38'28″S, 70°34'27″W). Blood samples from 14 adult animals were collected, and measurements of melaton

Hypothalamus as an endocrine organ.

The endocrine hypothalamus constitutes those cells which project to the median eminence and secrete neurohormones into the hypophysial portal blood to act on cells of the anterior pituitary gland. The entire endocrine system is controlled by these peptides. In turn, the hypothalamic neuroendocrine cells are regulated by feedback signals from the endocrine glands and other circulating factors. The neuroendocrine cells are found in specific regions of the hypothalamus and are regulated by afferents from higher brain centers. Integrated function is clearly complex and the networks between and amo

Clinical investigation of a unique type of hypothalamic adrenal insufficiency.

Hypothalamic adrenal insufficiency (AI) is a rare but distinct type of AI. The leading cause of hypothalamic AI is a secondary side-effect of exogenous steroid intake, particularly in large amounts and/or long-term periods. The next cause would be the effect of the tumor in the hypothalamic lesions. We show here 9 cases of hypothalamic AI without any disorder on imagings and a history of steroid administration. All patients had general fatigue; 7 patients (77.8%) had a history of hypoglycemia; 5 patients (55.6%) had a history of hypotension. None of the patients had hyponatremia, hyperkalemia,

Preserved circadian variation in cortisol and androgens during a ski traverse of Antarctica in summer.

Antarctic expeditions present extreme physiological challenges due to cold temperatures, high physical exertion, and 24-hour daylight. This observational study evaluated endocrine adaptation in nine participants (six men, three women) during a 47-day, 1,000 km unassisted ski traverse. Detailed salivary sampling was conducted before, during and after the expedition, corroborated by blood and hair sampling before and after the expedition. Cortisol, testosterone, and androstenedione were measured using mass spectrometry, and thyroid hormones via immunoassay. Diurnal cortisol, androstenedione and

Annual rhythms of thyroid hormone signaling: Environmental influences on thyroid function and disease implications.

Hormones within the hypothalamic-pituitary-thyroid (HPT) axis play a central role in acclimatization, dynamically responding to nutritional, thermal, and photoperiodic cues to coordinate metabolic, thermoregulatory, and reproductive functions. Abundant food elevates thyroid hormone (TH), driving energy storage and foraging behaviors, while scarcity reduces TH levels, inducing energy-saving states like hypometabolism or hibernation, in which TH-leptin crosstalk is important. Cold exposure upregulates TH to enhance mitochondrial thermogenesis, with TH acting as a pivotal mediator in the coordina

Selenium protects against nesfatin-1 modulation of the hypothalamic-pituitary-testicular axis during hypothyroidism in male rats.

Normal gonadal function can be disrupted by hypothyroidism. Hypothyroidism disturbs testicular function directly and centrally by affecting the hypothalamic-pituitary-testicular axis with unclear mechanism. As nesfatin-1 neurons co-localized with TRH and GnRH neurons in the hypothalamus, it could play a role in centrally hypothyroidism induced testicular dysfunction. Selenium (Se), by affecting thyroid iodide supply, could relieve these disturbances. So, we aim to identify the role of nesfatin-1 as a link between testicular dysfunction and hypothyroidism through modulating the MAPK/ERK pathway

Thyroid and Pseudothyroid Dysfunction as a Cause That is Promoting the Relapse of Benign Focal Thyroid Pathology.

Several studies deal with learning causes stipulating nodular formations in the thyroid tissue, including those occurring against the ground of metabolic disorders of thyroid hormones. Our study's objective was to determine the peculiarities of thyroid homeostasis disorders in patients suffering from benign nodular thyroid pathology with relapses of the disease and its relapse-free course. For this purpose, 96 female patients suffering from nodular thyroid pathology and 20 without thyroid pathology were examined. In the course of the study, the following were found in patients with benign foca

Functional sex differences ('sexual diergism') of central nervous system cholinergic systems, vasopressin, and hypothalamic-pituitary-adrenal axis activity in mammals: a selective review.

Sexual dimorphism of the mammalian central nervous system (CNS) has been widely documented. Morphological sex differences in brain areas underlie sex differences in function. To distinguish sex differences in physiological function from underlying sexual dimorphisms, we use the term, sexual diergism, to encompass differences in function between males and females. Whereas the influence of sex hormones on CNS morphological characteristics and function of the hypothalamic-pituitary-gonadal axis has been well-documented, little is known about sexual diergism of CNS control of the hypothalamic-pitu

[Hypothalamic dysfunction. 2 cases: the contribution of nuclear magnetic resonance, therapeutic trial of naltrexone].

Two cases of idiopathic hypothalamic dysfunction (one boy and one girl) are reported. Symptoms of hypothalamic dysfunction were noted by the age of 2 years: initial polyphagia and obesity with subsequent anorexia and emaciation were observed in one patient. Thermoregulation and thirst disorders, recurrent accesses of hypernatremia, acrocyanosis and profuse sweating were present. Impaired growth and delayed puberty in one case, and in the other hypogonadism, absence of growth hormone and gonadotrophins release in response to provocative stimuli were observed as well as abnormal thyroid stimulat

Congenital hypothyroidism (cretinism) in neuroD2-deficient mice.

Mice lacking neuroD2, a basic helix-loop-helix transcription factor involved in brain development, show growth retardation and other abnormalities consistent with hypothalamic-pituitary-thyroid (HPT) axis dysfunction. neuroD2 is expressed in the paraventricular hypothalamic nuclei, the anterior lobe of pituitary, and the thyroid gland. In neuroD2-deficient mice, thyrotropin-releasing hormone, thyroid-stimulating hormone, and thyroid hormone are decreased in these three regions, respectively. neuroD2-null mice typically die 2 to 3 weeks after birth, but those treated with replacement doses of t

GnIH-induced mitochondrial dysfunction lead to oxidative stress and apoptosis in thyroid follicular cells, causing hypothyroidism.

Thyroid disorders profoundly disrupt metabolism, development, growth, pubertal timing, and fertility in domestic animals. Gonadotropin-inhibitory hormone (GnIH), a key inhibitory neuropeptide regulating reproductive function, has been implicated in metabolic dysfunction-associated infertility as well as thyroid dysfunction-related pubertal abnormalities. These observations suggest potential crosstalk between GnIH and thyroid hormones (THs), positioning GnIH as a possible integrative regulator linking the hypothalamic-pituitary-thyroid (HPT) and hypothalamic-pituitary-gonadal (HPG) axes.
