[Effect of Yuxuebi Tablets on mice with inflammatory pain based on GPR37-mediated inflammation resolution].

In order to investigate whether the effect of Yuxuebi Tablets on the peripheral and central inflammation resolution of mice with inflammatory pain is related to their regulation of G protein-coupled receptor 37(GPR37), an inflammatory pain model was established by injecting complete Freund's adjuvant(CFA) into the paws of mice, with a sham-operated group receiving a similar volume of normal saline. The mice were assigned randomly to the sham-operated group, model group, ibuprofen group(91 mg·kg~(-1)), and low-, medium-, and high-dose groups of Yuxuebi Tablets(60, 120, and 240 mg·kg~(-1)). The

Targeted suppression of cytokine-mediated inflammation in rheumatoid arthritis by Melia azedarach L. extract.

Inflammation is a critical contributor to the pathogenesis of rheumatoid arthritis (RA), primarily mediated by NF-κB signalling. Medicinal plants have long been utilised for the treatment of inflammatory disorders, including RA, due to their minimal side effects and cost effectiveness. The current study was designed to explore the anti-inflammatory potential of Melia azedarach L. and to validate its traditional use in rheumatic disorders. Ethanolic extract of Melia azedarach (Ma.EE) was prepared and characterized by GCMS and FTIR techniques. Phytochemicals presents in the Ma.EE were quantified

Glycoprotein Ib-CD11b + monocyte-derived macrophages mediate atherosclerosis-exacerbated psoriatic inflammation.

BACKGROUND: Psoriasis is a systemic inflammatory skin disease affecting 2-3 % of the global population, with significant cardiovascular comorbidities. A complex inflammatory interaction exists between psoriasis and atherosclerosis, but the exact mechanism remains unclear. METHODS: To elucidate this relationship, we collected skin biopsies from treatment-naïve patients with comorbid psoriasis and atherosclerosis underwent single-cell RNA sequencing, integrated with public atherosclerosis datasets. Bioinformatics analysis identified cell populations and interactions. Validation included immunoh

Inflammation-mediated regional brain alterations associated with mild cognitive impairment in knee osteoarthritis.

OBJECTIVES: Knee osteoarthritis (KOA), a degenerative joint disease marked by chronic pain, is associated with systemic inflammation that may extend to neurocognitive dysfunction. While chronic low-grade inflammation in KOA has been implicated in mild cognitive impairment (MCI), a prodromal stage of dementia, the mediating role of inflammation in brain functional reorganization remains unclear. METHODS: This study integrated neuroimaging, inflammatory biomarkers, and machine learning to investigate inflammation-mediated brain functional alterations in 63 KOA patients with/without MCI. Serum l

Tocilizumab Treatment for Microvascular Inflammation and Chronic Active Antibody-mediated Rejection in Kidney Transplantation.

BACKGROUND: Interleukin-6 (IL-6) is an important driver of humoral immunity and a target for treatment of antibody-mediated rejection (AMR) in kidney transplantation. Data on IL-6 inhibition for treatment of AMR are mixed and their efficacy remains inconclusive. In this retrospective observational study, we investigated the association of monthly tocilizumab infusions with the trajectory of estimated glomerular filtration rate (eGFR) among kidney transplant patients with histologic features of chronic active AMR. METHODS: Through institutional database review, 85 adult kidney transplant patie

MiR-124 и MIR-155 в качестве терапевтических целей в Microglia-Mediated Inflammation in Multi-Sclerosis.

Scar matrix drives Piezo1 mediated stromal inflammation leading to placenta accreta spectrum.

Scar tissue formation is a hallmark of wound repair in adults and can chronically affect tissue architecture and function. To understand the general phenomena, we sought to explore scar-driven imbalance in tissue homeostasis caused by a common, and standardized surgical procedure, the uterine scar due to cesarean surgery. Deep uterine scar is associated with a rapidly increasing condition in pregnant women, placenta accreta spectrum (PAS), characterized by aggressive trophoblast invasion into the uterus, frequently necessitating hysterectomy at parturition. We created a model of uterine scar,

Association between tooth loss and depression mediated by lifestyle and Inflammation- a cross-sectional investigation.

BACKGROUND: Depression and tooth loss are associated with the occurrence of systemic disease or the progression of multi-factorial disease, and both are considered important public health issues by World Health Organization (WHO). Previous research just suggested that tooth loss can generate psychological stress, low self-esteem, anxiety and other emotional disturbances. However, the precise correlation and underlying mechanisms between depression and tooth loss remains poorly understood. Consequently, we aim to explore the association between depression and tooth loss through a cross-section

Inflammation-mediated drug interactions of olokizumab and cytochrome P450 activities in patients with rheumatoid arthritis.

AIMS: In patients with rheumatoid arthritis (RA), interleukin (IL)-6 affects the activity of cytochrome P450 (CYP) enzymes. Treatment with anti-IL-6 therapy can reverse the IL-6-mediated downregulation of CYP enzymes, resulting in changes in plasma levels of CYP substrates. The primary objective of this study was to evaluate the impact of the IL-6 inhibitor olokizumab on the pharmacokinetics of CYP probe substrates in subjects with active RA. METHODS: Seventeen patients with active RA were orally administered a phenotyping cocktail of midazolam (CYP3A4 substrate), omeprazole (CYP2C19 substrat

Gasdermin E-mediated keratinocyte pyroptosis participates in the pathogenesis of psoriasis by promoting skin inflammation.

BACKGROUND: Psoriasis is a common chronic inflammatory disease with an unclear aetiology. Keratinocytes in psoriasis are susceptible to exogenous triggers that induce inflammatory cell death. OBJECTIVES: To investigate whether gasdermin E (GSDME)-mediated pyroptosis in keratinocytes contributes to the pathogenesis of psoriasis. METHODS: Skin samples from patients with psoriasis and from healthy controls were collected to evaluate the expression of GSDME, cleaved caspase-3 and inflammatory factors. We then analysed the data series GSE41662 to further compare the expression of GSDME between le

IFITM3 mediates inflammation induced myocardial injury through JAK2/STAT3 signaling pathway.

Myocarditis is an inflammation of the heart muscle often associated with viral infections and can lead to dilated cardiomyopathy. Interferon-induced transmembrane protein 3 (IFITM3) is a small endosomal membrane protein with anti-viral activity against multiple viruses and is also implicated in non-infectious diseases such as cancer and Alzheimer's Disease. Since the IFITM3 proteins are expressed both in T cells and in cardiomyocytes, it is reasonable to hypothesize that these molecules could affect myocarditis either through their effect on the autoimmune response or through direct modulation

Immune-mediated impairment of tonic immobility defensive behavior in an experimental model of colonic inflammation.

Ulcerative colitis has been associated with psychological distress and an aberrant immune response. The immunomodulatory role of systemic cytokines produced during experimental intestinal inflammation in tonic immobility (TI) defensive behavior remains unknown. The present study characterized the TI defensive behavior of guinea pigs subjected to colitis induction at the acute stage and after recovery from intestinal mucosa injury. Moreover, we investigated whether inflammatory mediators (tumor necrosis factor (TNF)-α, interleukin (IL)-1β, IL-8, IL-10, and prostaglandins) act on the mesencephal

Deficient chaperone-mediated autophagy in macrophage aggravates inflammation of nonalcoholic steatohepatitis by targeting Nup85.

BACKGROUND & AIMS: Nonalcoholic steatohepatitis (NASH), a more severe subtype of nonalcoholic fatty liver disease, can cause cirrhosis and hepatocellular carcinoma. Macrophages play critical roles in initiating and maintaining NASH-induced liver inflammation and fibrosis. However, the underlying molecular mechanism of macrophage chaperone-mediated autophagy (CMA) in NASH remains unclear. We aimed to investigate the effects of macrophage-specific CMA on liver inflammation and identify a potential therapeutic target for NASH treatment. METHODS: The CMA function of liver macrophages was detected

Nocardia seriolae mediates liver granulomatous chronic inflammation in Micropterus salmoides through pyroptosis.

Granulomatous diseases caused by Nocardia seriously endanger the health of cultured fish. These bacteria are widely distributed, but prevention and treatment methods are very limited. Chronic granulomatous inflammation is an important pathological feature of Nocardia infection. However, the molecular mechanisms of granuloma formation and chronic inflammation are still unclear. Constructing a granuloma infection model of Nocardia is the key to exploring the pathogenesis of the disease. In this study, we established a granuloma model in the liver of largemouth bass (Micropterus salmoides) and as
