Selective labeling by propidium iodide injected into the lateral cerebral ventricle of the rat.

Injections of propidium iodide (PI) into the lateral cerebral ventricle of the rat resulted in a bilateral labeling in the septohippocampal nuclei, substantia nigra (SN), ventral tegmental area (VTA), retrorubral nuclei (rr), dorsal and median raphe nuclei, regions within and dorsal to the medial lemniscus of the caudal midbrain, and Purkinje cells in the cerebellum. No labeled neurons were seen in other areas of the brain. The data suggest that PI appears to exhibit selective labeling, and the mechanism underlying the selective labeling is discussed. Combined with Faglu histofluorescence, it

Effect of cholecystokinin octapeptide analogues on food intake in the dog.

Cholecystokinin octapeptide, administered into the third cerebral ventricle (icv), suppresses feeding in sheep, pigs, chicken, rats, and dogs. Because of the species differences in the feeding response to cholecystokinin (CCK), we studied the pharmacological characterization of this peptide on feeding in 16-h-fasted dogs. We examined the effects of CCK-(26-33)-NH2 (CCK-8) and a variety of its analogues, nonsulfated CCK-(26-33)-NH2 (desulfated CCK-8), CCK-(26-33)-OH (deamidated CCK-8), (Nle28,31)-CCK-(26-33)-NH2 [(Nle28,31)-CCK-8], succinyl-CCK-(27-33)-NH2 (Suc-CCK-7) succinyl-Thr28, Leu29, MeP

The distribution of total mercury in the brain after the lateral ventricular single injection of methylmercury and glutathione.

We determined the time course of the distribution of total mercury in the brain after the lateral ventricular single injection of methylmercury (Me-Hg) and glutathione (GSH) to confirm our assumption that glutathione have a very important role in methylmercury transport into the brain. The following results were obtained: (1) The transport of methylmercury into the brain was accelerated by glutathione, but was retarded by the surplus glutathione. (2) The increase of glutathione to methylmercury ratio in the dose was tend to uniformize the distribution of total mercury in the brain. These resul

Central atrial natriuretic factor reduces vasopressin secretion in the rat.

Atrial natriuretic factor (ANF, Arg 101-Tyr 126; 0.5 and 2.5 micrograms) administered into the lateral cerebral ventricle of conscious euhydrated and dehydrated rats resulted in a significant reduction in the plasma vasopressin concentration. The effect of ANF on vasopressin secretion was greater in the water-deprived animal. Central ANF was without effect on mean arterial blood pressure in both euhydrated and dehydrated rats. The data suggest that ANF occurring in the central nervous system may be important in the control of vasopressin secretion.

CNS toxicity and CSF pharmacokinetics of intraventricular DFMO and MGBG in beagle dogs.

We have developed a beagle dog model to study the pharmacology and toxicology of anticancer drugs administered through the 3rd or lateral ventricles. A Foltz-type reservoir was implanted SC and connected by tube into a cerebral ventricle. Drugs were administered directly into the reservoir; CSF sampling of drugs administered into the ventricle was achieved directly by tapping the reservoir or by percutaneous puncture of the cisterna magna. In the current study, we evaluated the CSF pharmacokinetics and CNS toxicity of two inhibitors of polyamine metabolism, alpha-difluoromethylornitine (DFMO)

Beta-endorphin: effect on thermoregulation in aged monkeys.

In previous experiments small doses of the opiate morphine produced greater hyperthermia in aged than in younger sub-human primates. To test whether this augmented response is due to enhanced sensitivity of CNS opioid receptors with age, beta-endorphin (0.625-5 micrograms), an endogenous opioid peptide, was injected into the lateral cerebral ventricle (ICV) of young (less than 9 years) and aged (greater than 9 years) squirrel monkeys. Significantly greater hyperthermias developed in the older primates after each dose. In the aged monkeys, all but the smallest dose increased core temperature ab

Effect of naloxone on baroreflex, sympathetic tone and blood pressure in the cat.

Studies were designed to determine the effect of naloxone on sympathetic nerve activity and blood pressure and, also to investigate the central effect of naloxone in relation to the baroreflex system in alpha-chloralose-anesthetized cats. Following intravenous injection of naloxone (2 mg/kg), preganglionic splanchnic nerve (PSN) activity significantly increased in parallel to the increase in blood pressure. Set gain (mmHg-1) of carotid sinus baroreflex together with the operational range (mmHg) was increased after naloxone. Intraventricular (4th) injection of naloxone (0.2 mg/kg) produced iden

beta-Endorphin-induced hyperthermia in the cat.

beta-Endorphin was injected into the third cerebral ventricle (ICV) of conscious, unrestrained cats. Hyperthermic response to 50 microgram of this peptide were reduced by 20-100 microgram naloxone given ICV 1 hr later. A dose of 40 microgram beta-endorphin increased body temperature at ambient temperature of 4, 22 and 34 degrees C, with the response being greater the warmer the environment. These results indicate that beta-endorphin acts on a central naloxone-sensitive receptor which is probably the v2 receptor that is activated by low doses of D-Ala2-Met-enkephalinamide to evoke a similar pat

Cholinergic and catecholaminergic interaction and fluid intake in the rat.

The effect of simultaneous topical application of adrenergic agonists was investigated on the polydipsia caused by direct carbachol (CCh) stimulation of the rat brain. In addition, fluid selection (4-choice arrangement) was studied. Noradrenaline and isoprenaline fully antagonized the CCh effect after either intracerebroventricular, intraseptal or intrapreoptic application. Phenylephrine inhibited CCh polydipsia after intracerebroventricular or intrahypothalamic but not after intraseptal injection. Previous experiments had shown that adrenergic agonists also exerted a marked modulatory influen

Effects of intracerebroventricular administration of aliphatic diamines on ingestive behavior in the rat.

We examined the pharmacological effects of intracerebroventricularly administered aliphatic diamines on ingestive behavior in male rats adapted to a 4 hr per day feeding and drinking schedule. 1,2-Ethanediamine (ETD), 1,3-propanediamine (PRD), 1,4-butanediamine (putrescine, PUT), 1,5-pentanediamine (cadaverine, CAD) and 1,6-hexanediamine (HED) suppressed feeding and drinking behavior in a dose-dependent manner, but not unless a relatively high dose (over 80 micrograms) was given. The approximate anorectic potency was HED greater than CAD divided by PUT greater than ETD greater than PRD. A seda

Cefazolin: a valid model of experimental epilepsy?

In various animal species (chicks, rats and rabbits) the intravenous injection of high doses of cefazolin, a cephalosporin not hitherto reported to produce epileptic seizures, was found to produce epileptiform electrocortrical changes similar to those evoked by intravenous benzylpenicillin. Similar phenomena as those reported after systemic injection were also observed after microinjection of cefazolin into the III cerebral ventricle. In fact, both in chicks and rats cefazolin given by the latter route produced wild running crisis, myoclonic jerks of the limbs and in some instances generalized

Effects on blood pressure of noradrenaline and isoprenaline administered into the third ventricle of the brain of anaesthetized and conscious cats.

1 Noradrenaline administered into the third ventricle of the brain (IIIv) of both conscious and anaesthetized cats induced increases in blood pressure accompanied by small and variable heart rate effects. The pressor responses were reduced after autonomic ganglion blockade indicating their likely central origin. 2 Noradrenaline when administered either into a lateral cerebral ventricle (i.c.v.) in conscious and anaesthetized cats or into the cisterna magna (i.c.) in anaesthetized cats induced falls in blood pressure accompanied by bradycardia. 3 Pressor responses to IIIv noradrenaline in consc

CCK-octapeptide injected in CSF decreases meal size and daily food intake in sheep.

Cholecystokinin octapeptide (CCK-OP) is a potent and specific suppressor of feeding when administered as a continuous lateral cerebral ventricular injection in fasted sheep, and we have proposed that endogenous CCK-OP in the brain is released during meals and acts to terminate feeding. In previous studies, however, only relatively short-term effects of CCK-OP on feeding were examined. In the first experiment of the present series sheep were adapted to a 6-hr feeding period per day. CCK-OP injected continuously for 6 hr into the lateral ventricles reduced feeding during the entire feeding perio

Effects of chronic intraventricular administration of angiotensin II on drinking behavior and blood pressure.

Angiotensin II was continuously infused into the lateral cerebral ventricle of rats, and the effects on daily food and water consumption, urine volume, and aortic blood pressure were studied. All was infused at a rate of 10 ng/hr for seven days, using subcutaneously implanted osmotic minipumps. An intraventricular (IVT) control group was infused with only the saline vehicle, while a third group received AII subcutaneously. IVT AII rats showed a four-fold increase in water consumption, to a mean of 171 ml/day during Days 2-4 of infusion, whereas water intake of the other groups did not change f

Effects on fluid balance induced by non-febrile intracerebroventricular infusions of PGE2, PGF2alpha, and arachidonic acid in the goat.

Prostaglandins (PG) E2, F2 alpha (30 ng/kg . min-1) and arachidonic acid (150 and 300 ng/kg . min-1) were infused for 30 min into the lateral cerebral ventricle of conscious hydrated and non-hydrated goats. Like previously shown as concerns PGE1 PGE2 was found to inhibit the water diuresis and cause some increase in the renal sodium excretion in the hydrated animal, and to elicit thirst in the non-hydrated goat. The effects of PGE2 were enhanced when hypertonic (0.25 M) NaCl was simultaneously infused into the ventricle. The antidiuretic effect of PGF2 alpha was less pronounced, and drinking o

Respiratory and circulatory effects of saxitoxin in the cerebrospinal fluid.

1 In cats anaesthetized with pentobarbitone, saxitoxin and, on a few occasions, tetrodotoxin were injected into a lateral cerebral ventricle or into the subarachnoid space of the lower brain stem. Observations were made on frequency and tidal volume of breathing, on CO(2) responsiveness and on electrical responsiveness of the respiratory centre. Effects on the blood pressure were observed simultaneously.2 A single large dose of toxin, e.g., 250 ng, produced within minutes apneustic breathing and a rise in blood pressure which were converted rapidly to respiratory failure and hypotension. In co

Mucor cerebral abscess associated with intravenous drug abuse.

We report on a case of a 26-year-old intravenous narcotic abuser with a primary cerebral mucormycotic abscess caused by Rhizopus oryzae. He was treated with a combination of intravenous and intraventricular amphotericin B and surgical drainage with a successful outcome. There was no evidence that his infection was acquired by the rhinocerebral route, it seems likely that he injected himself with a contaminated batch of narcotic or amphetamine. Mucormycosis presenting in this way has been described previously but this is only the second such case to survive. Early diagnosis and treatment is ess

Polymyxins for CNS infections: Pharmacology and neurotoxicity.

Central nervous system (CNS) infections caused by multi-drug resistant (MDR) Gram-negative bacteria present a major health and economic burden worldwide. Due to the nearly empty antibiotic discovery pipeline, polymyxins (i.e. polymyxin B and colistin) are used as the last-line therapy against Gram-negative 'superbugs' when all other treatment modalities have failed. The treatment of CNS infections due to multi-drug resistant Gram-negative bacteria is problematic and associated with high mortality rates. Colistin shows significant efficacy for the treatment of CNS infections caused by MDR Gram-
