Co-ordination of iron acquisition, iron porphyrin chelation and iron-protoporphyrin export via the cytochrome c biogenesis protein CcmC in Pseudomonas fluorescens.

The cytoplasmic membrane protein CcmC is, together with other Ccm proteins, a component for the maturation of c-type cytochromes in Gram-negative bacteria. A Pseudomonas fluorescens ATCC 17400 ccmC mutant is cytochrome c-deficient and shows considerably reduced production of the two siderophores pyoverdine and quinolobactin, paralleled by a general inability to utilize various iron sources, with the exception of haem. The ccmC mutant accumulates in a 5-aminolevulinic acid-dependent synthesis a reddish, fluorescent pigment identified as protoporphyrin IX. As a consequence a visA phenotype simil…

Time-of-day difference in iron supplementation in iron-deficient pregnant model mice.

During pregnancy, iron supplementation is commonly recommended once daily, partly because hepatic hepcidin secretion following iron intake suppresses subsequent iron absorption. However, the optimal timing of iron administration during maternal iron deficiency remains unknown. We established a pregnant mouse model of iron deficiency and compared the effects of iron (FeSO4, 1 mg/kg) supplementation at the beginning (ZT12) and end (ZT0) of the daily active phase of pregnant mice on maternal, placental, and fetal outcomes. Supplementation at ZT12 significantly enhanced placental iron transport-re…

Comparative study of oral iron and intravenous iron therapy in pregnancy induced anemia.

Pregnancy-induced anemia, primarily caused by iron deficiency, poses significant risks to maternal and fetal health, especially in developing countries. Oral iron therapy is commonly used but has limitations due to poor compliance and gastrointestinal side effects. Therefore, it is of interest to compare the efficacy and tolerability of oral versus intravenous iron therapy in pregnant women with iron deficiency anemia. Results show that intravenous iron significantly improves hemoglobin levels more effectively and with fewer adverse effects than oral iron. This study advances knowledge by high…

Iron-borane catalyzed carbonyl hydroboration and isolation of an iron(I)-ketyl radical.

Hydroboration of carbonyl compounds has proven a pivotal route to access alcohols and other C1 moieties in recent years. Despite this, iron-based catalyst systems are somewhat limited with very little mechanistic understanding of these systems developed. Here we show that an iron metalloborane complex [{(iPrDPBPh)Fe}2(μ-1,2-N2)] (A) is an efficient pre-catalyst for hydroboration of ketones, cyclic esters and CO2 with mild conditions. Mechanistic insights reveal a previously unreported direct iron(0)-mediated ligand-to-ligand hydride transfer (LLHT) process is in operation with B-H bond breakin…

Polycatechol-based iron predators disrupt fungal iron homeostasis to drive selective antifungal action.

Fungal infections pose a growing global health challenge, exacerbated by a scarcity of effective treatments and rising drug resistance. Although cationic polymers emerge as promising antifungal candidates owing to structural tunability, design flexibility, and resistance to proteolytic degradation, their clinical utility has been hampered by nonselective membrane-disruption mechanisms. Herein, we develop a class of polycatechols- termed fungal iron predators (FIPs), exhibit exceptional fungicidal activity and markedly low cytotoxicity. These FIPs can efficiently infiltrate fungal cells, select…

Spatiotemporal iron-hijacking hydrogel reprograms host-pathogen iron homeostasis for fungal keratitis therapy.

Severe infections caused by opportunistic fungal pathogens like Candida albicans remain a major clinical challenge, exemplified by vision-threatening fungal keratitis (FK). Targeting host-pathogen iron competition offers a promising antifungal strategy. Here, we identify a vicious cycle in human FK, where iron overload-driven host ferroptosis promotes fungal proliferation and inflammation. To disrupt this, we develop a spatiotemporally controlled gallium-deferiprone (Ga-DFP) composite hydrogel (PGaDH) that reprograms host-pathogen iron homeostasis through an iron-hijacking strategy. Specifical…

Iron Deficiency and Iron Deficiency Anemia in Chronic Disease-Common, Important, and Treatable.

Iron has many important functions related to energy metabolism. However, hemoglobin synthesis is always a priority. Iron deficiency can be caused by increased loss, insufficient intake, or decreased absorption from the intestine and reduced release from depots in systemic inflammation. Anemia appears when stores are depleted or when utilization of iron from the stores is impaired. Treatment with oral iron is the first choice when the intestine is healthy, and the patient is free of inflammation. Intravenous iron is indicated when oral iron is ineffective or not tolerated and if more rapid corr…

Nano-nutraceuticals from spinach-iron oxide as a green remedy for iron deficiency.

Iron deficiency anemia (IDA) is a nutritional metabolic condition that effects infants, children, even adults, both men and women. Individuals with IDA have inadequate iron intake or impaired absorption of iron. The main cause of IDA is the low bioavailability of iron from conventional means, diet and supplementation, even if present in an ambient amount. The major objective of this study was to increase the bioavailability of iron using green technology. For this purpose, iron oxide nanoparticles were fabricated using iron-rich spinach leaves extract. The characterization of the particles was…

Advancing Iron Therapy in Maternal Health: Evolving Strategies for Treating Iron Deficiency Anemia.

IMPORTANCE: Iron deficiency anemia (IDA) affects approximately one-third of pregnant women worldwide and is a significant contributor to adverse maternal and fetal outcomes, including preterm delivery, low birth weight, and cognitive impairment in children. Despite advancements in iron therapy, IDA remains prevalent, affecting 29.9% of women globally. This highlights a critical need for more effective management strategies during pregnancy. OBJECTIVE: To provide a comprehensive review of the mechanisms governing iron absorption, current guidelines for iron therapy, and the efficacy and safety…

Role of Iron Metabolic Disturbances and Inflammatory Iron Biomarkers in Liver Transplant Prognosis.

Iron metabolism plays a pivotal role in liver transplantation, significantly impacting outcomes for both donors and recipients. The liver, central to iron homeostasis, is often impaired in chronic liver diseases leading to metabolic disorders that exacerbate liver damage. Liver transplantation (LT) is a critical treatment for end-stage liver diseases, with iron status in both donors and recipients influencing post-transplant outcomes. Studies indicate that pre-transplant iron overload in recipients is associated with poor liver function recovery, increased graft rejection risk, and reduced pat…

Mammalian SLC39A13 promotes ER/Golgi iron transport and iron homeostasis in multiple compartments.

Iron is a potent biochemical, and accurate homeostatic control is orchestrated by a network of interacting players at multiple levels. Although our understanding of organismal iron homeostasis has advanced, intracellular iron homeostasis is poorly understood, including coordination between organelles and iron export into the ER/Golgi. Here, we show that SLC39A13 (ZIP13), previously identified as a zinc transporter, promotes intracellular iron transport and reduces intracellular iron levels. ZIP13 loss causes an iron deficiency in the ER/Golgi and other intracellular compartments, such as lysos…

Fetal iron uptake from recent maternal diet and the maternal RBC iron pool.

BACKGROUND: During pregnancy iron can be obtained from the diet, body iron stores, or iron released from RBC catabolism. Little is known about the relative use of these sources to support fetal iron acquisition. OBJECTIVES: To describe longitudinal change in iron absorption and enrichment across gestation and partitioning of RBC iron to the fetus. METHODS: Fifteen pregnant women ingested an oral stable iron isotope (57Fe) in the second trimester (T2) of pregnancy (weeks 14-16) to label the RBC pool, and a second oral stable isotope (58Fe) in the third trimester (T3) (weeks 32-35). Absorption…

Erythroferrone contributes to iron mobilization for embryo erythropoiesis in iron-deficient mouse pregnancies.

Erythroferrone (ERFE) is an erythroblast-secreted regulator of iron metabolism. The production of ERFE increases during stress erythropoiesis, leading to decreased hepcidin expression and mobilization of iron. Pregnancy requires a substantial increase in iron availability to sustain maternal erythropoietic expansion and fetal development and is commonly affected by iron deficiency. To define the role of ERFE during iron-replete or iron-deficient pregnancy, we utilized mouse models expressing a range of ERFE levels: transgenic (TG) mice overexpressing ERFE, wild-type (WT), and ERFE knockout (KO…

TMPRSS6 gene mutations in six Saudi families with iron refractory iron deficiency anemia.

Iron-refractory iron deficiency anemia (IRIDA) is considered an autosomal recessive iron deficiency anemia due to mutations in the transmembrane protease serine 6 (TMPRSS6) gene. Variations in iron parameters and a higher risk of iron deficiency have been linked to the TMPRSS6 mutations. Furthermore, human genome-wide association studies (GWAS) identified a common mutation (rs855791) linked to abnormal hematological parameters, highlighting the importance of the TMPRSS6 gene in the regulation of iron homeostasis. This is the first study to investigate TMPRSS6 gene mutation in six Saudi familie…
