Cardiovascular effects of intrathecally administered bradykinin in the rat: characterization of receptors with antagonists.

1. The effects of intrathecal (i.t.) pretreatment with selective B1 or B2 kinin receptor antagonists were studied on the cardiovascular response to i.t. injection of bradykinin (BK) in conscious freely moving rats. 2. BK (81 pmol) produced an increase in mean arterial pressure (MAP: 9-13 mmHg) and decrease in heart rate (HR: 20-30 beats min-1) that reached a maximum 2 min after injection. 3. The BK-induced cardiovascular responses were dose-dependently and reversibly reduced by four antagonists with the following rank order of potency: Tyr, D-Arg[Hyp3,D-Phe7,Leu8]-BK = D-Arg[Tyr3,D-Phe7,Leu8]-

Characterization of serine proteinases isolated from rat submaxillary gland: with special reference to the degradation of rat kininogens by these enzymes.

From the homogenate of rat submaxillary gland, two kinds of serine proteinases, named tentatively proteinases A and B, were isolated and their chemical properties and activities toward rat kininogens were examined, in comparison with those of submaxillary kallikrein. Proteinase A with Mr of 28,200 rapidly cleaved high-molecular-weight (HMW) kininogen into a protein of 67 kDa, which retained thiol-proteinase inhibitory activity, but had lost the correcting activity of HMW kininogen on the prolonged clotting time of Fitzgerald trait plasma. It liberated bradykinin from HMW kininogen but did not

"Two-step" pemphigus induction by ACE-inhibitors.

Pemphigus lesions appeared in a 58-year-old man who was taking captopril for his hypertension. Drug withdrawal resulted in complete remission of the eruption. The subsequent use of enalapril as an antihypertensive agent caused a recurrence of pemphigus lesions along with onset of itching and dermographism. Intercellular antibodies were not found. Discontinuance of enalapril therapy had no effect on the clinical course. Steroid treatment was needed to resolve the eruption. Recently repeated immunofluorescent studies disclosed intercellular IgG antibodies in the serum at a low titer. Pemphigus i

Evidence for new spasmogenic substances released by trypsin from plasma of normotensive and hypertensive rats.

We have extended our previous study of increased levels of new spasmogenic substances released by trypsin from the plasma of chronic one-kidney, one-clip hypertensive rats by describing preparative procedures utilizing Sephadex G-25, CM-cellulose and C18 reverse-phase HPLC for their partial purification. We demonstrate the existence of at least 6 HPLC components, distinguishable from bradykinin, bradykinin homologues having Lys before Arg1 of bradykinin and T-kinin (Ile-Ser-bradykinin) on the basis of their chromatographic properties. Several of these spasmogenic substances induce a very slow

[Initial manifestation of eruptive exanthematous psoriasis vulgaris caused by captopril medication].

Angiotensin-converting enzyme (ACE) is identical to the dipeptidyl carboxypeptidase kininase II, which inactivates bradykinin. That is why captopril, an orally active ACE inhibitor, alters the kallikrein-kinin-arachidonic acid system. It is suggested that this mechanism could be responsible for the dosage-dependent incidence of polymorph cutaneous eruptions during therapy with captopril. A 42-year-old female patient with no previous personal or family history of psoriasis developed an eruptive-exanthematous type of psoriasis vulgaris 1-2 weeks after the beginning of captopril therapy. It is su

Bartter's syndrome - a dilemma of cause and effect.

Six interrelated abnormalities of Bartter's syndrome are analyzed-juxtaglomerular hyperplasia, angiotensin resistance, altered kallikrein-kinin system, hyperprostaglandinuria, hypokalemia, and chloride-losing nephropathy. Arguments are advanced that any one of these could be the proximate cause and result in all the others. By the same token, each abnormality could be a consequence of any of the others and, furthermore, modulate the others by negative or positive feedback. Despite many recent insights, available data do not permit a definitive conclusion as to the locus of the primary abnormal

Normalization of C1 Inhibitor in a Patient with Hereditary Angioedema.

Hereditary angioedema is a potentially life-threatening autosomal dominant condition, causing attacks of angioedema due to failure to regulate bradykinin. Nearly all cases of hereditary angioedema are caused by mutations in the gene encoding C1 inhibitor, SERPING1. C1 inhibitor is a multifunctional protein produced in the liver that regulates the kallikrein-kinin system at multiple points. An infant with genetically confirmed hereditary angioedema and low C1 inhibitor levels (but without previous episodes of angioedema) underwent liver transplantation for biliary atresia, an unrelated conditio

[Characteristics of the kallikreins in healthy and malignant (adenocarcinoma) mucous membrane of the human large intestine].

Total kininogenase activity was higher by 50% in supernatant of malignant mucosa homogenate as compared with normal tissue. But specific kininogenase and N-acetyl-arginine esterase activity were distinctly similar both in normal and tumoral tissues; N-ac-arg esterase activity tended to decrease in malignant mucosa. Partially purified preparations of kallikrein, isolated from adenocarcinoma and normal mucosa of human large intestine, were distinctly identical in their properties: both these enzymes were not inhibited by soybean trypsin inhibitor, they were highly sensitive to main pancreatic in

Plasma kallikrein: the bradykinin-producing enzyme.

Plasma prekallikrein is the liver-derived precursor of the trypsin-like serine protease plasma kallikrein (PK) and circulates in plasma bound to high molecular weight kininogen. The zymogen is converted to PK by activated factor XII. PK drives multiple proteolytic reaction cascades in the cardiovascular system such as the intrinsic pathway of coagulation, the kallikrein-kinin system, the fibrinolytic system, the renin-angiotensin system and the alternative complement pathway. Here, we review the biochemistry and cell biology of PK and focus on recent in vivo studies that have established impor

Septic shock (Part 1).

Septic shock is a distinct clinical entity with an overall mortality of 30% to 40%. Gram-negative organisms are the most frequently identified causal agents. The shock syndrome appears to result from activation of complement, coagulation, kinin, endorphin, and other hormonal systems. Physiologic abnormalities include hypotension, metabolic acidosis, increased cardiac index, and decreased tissue extraction of oxygen. Clinicians must entertain the diagnosis of septic shock in a variety of settings, as prognosis is affected by early institution of therapy. Treatment is directed toward control of

[Physiologically active peptides and the regulation of arterial pressure in normal and pathological states].

Relation between kallikrein-kinin and renin-angiotensin polypeptide blood systems, main regulators of arterial pressure, is considered. Activities of kallikrein, renin and angiotensin-converting enzyme were studied in healthy persons and in patients with various forms of renin-dependent arterial hypertension under conditions of immobility and after physical loading. In dynamics of hereditary-determined hypertension of rats there were studied the effects of bradykinin and angiotensin II on alterations in reactivity of the vascular bed as well as on alterations in lung metabolic functions. Participation of kallikrein, coagulation/fibrinolysis parameters in the development of glomerulonephritis.

Masugi nephritis was induced in dogs in which platelet count, fibrinogen, antithrombin activity, plasma prekallikrein and immediate plasmin inhibitors were coincidentally decreased immediately after the injection of nephrotoxin serum. It was found that the grade of decrease of urinary kallikrein excretion following these immediate reactions were parallel with the grade of renal damages. By the pretreatment with heparin or the defibrination with snake venom, however, the histological findings of Masugi nephritis showed rather severe damage. Based on the consumption of coagulation factors, kalli

Possible relationship between salivary kallikrein and water-Na-K secretion stimulated by tityustoxin (TsTx).

Intravenous injection of purified scorpion toxin (TsTx) brings about the appearance of salivary flow with high levels of Na and K and kallikrein in the saliva of the rats. In experiments performed in vivo a positive correlation between Na and partially with K and kallikrein was observed in parotid and submaxillary saliva. After ouabaine, saliva K was increasing, whereas Na decreased, and a parallel decrease of kallikrein was observed. Adrenergic and cholinergic blocking drugs also showed a marked reduction in the output of Na (atropine), and partially of K (atropine or phentolamine), with a co

Studies on the prekallikrein-bradykininogen system in liver disease.

The plasma prekallikrein-bradykininogen system was studied in 45 patients with chronic liver disease since its activation with increased liberation of kinin into the plasma could account for some of the clinical manifestations of cirrhosis, namely, vasodilatation, hypotension, and increased capillary permeability.A significant reduction in plasma bradykininogen was found in the cirrhotic patients as compared with control groups of normal subjects and hospital inpatients. The mean plasma prekallikrein was not significantly different and only five patients with liver disease had reduced levels.

Mast cell degranulation and bradykinin-induced angioedema - searching for the missing link.

Initiation of the bradykinin generation cascade is responsible for the occurrence of attacks in some types of angioedema without wheals. Hereditary angioedema due to C1 inhibitor deficiency (HAE-C1-INH) is one such clinical entity. In this paper, we explore the existing evidence that mast cells (MCs) degranulation may contribute to the activation of the kallikrein-kinin system cascade, followed by bradykinin formation and angioedema. We present the multidirectional effects of MC-derived heparin and other polyanions on the major components of the kinin-kallikrein system, particularly on the fac

Unraveling Complexities: Rhabdomyolysis, Acute Renal Injury, and Compartment Syndrome Following a Wasp's Sting.

This study delves into the rare occurrence of rhabdomyolysis induced by wasp stings, emphasizing its toxic systemic repercussions. Drawing parallels with documented instances of insect bites worldwide, including those by honey bees and Africanized bees, the research explores the correlation between multiple wasp stings and acute renal failure associated with rhabdomyolysis. The venom's active components, such as amines, kinins, and histamine-releasing peptides, underpin toxic systemic reactions, leading to hemolysis, coagulopathy, and severe cytotoxicity-induced acute renal failure. The basis of topical superoxide dismutase antipruritic activity.

In humans, as in all mammals and most chordates, three forms of superoxide dismutase (SOD) are present: SOD1 is located in the cytoplasm, SOD2 in the mitochondria, and SOD3 is extracellular. SOD is used in cosmetic products to reduce free radical damage to the skin, for example, to reduce fibrosis following radiation for breast cancer. Pruritus is one of the most common symptoms of skin diseases, but can also be a major symptom of systemic diseases (e.g., malignancy, infection or metabolic disorders). There are various antihistaminics used as antipruritogenic substances. In the genesis of prur
