We investigate the interaction between dipalmitoylphosphatidylcholine (DPPC) and a nitroxide spin label in order to understand its influences on lipid structure and dynamics using molecular dynamics simulations. The system was modified by covalently attaching nitroxide spin labels to the headgroups of two DPPC molecules. (S-(2,2,5,5-tetramethyl-2,5-dihydro-1H-pyrrol-3-yl)methyl methanesulfonothioate) (MTSL) was used as the spin label. The label position and dynamics were analyzed as was the impact of the modified DPPC on the structure of the surrounding lipids. The modified DPPC molecules loca

Bacillus anthracis secretes three distinct proteins which interact in binary combinations to produce two toxins. The two effector moieties, edema factor (EF) and lethal factor (LF), interact competitively with the cell receptor-binding moiety, protective antigen (PA), to produce biologically distinct effects. The passage of the toxins through an acidified endosomal compartment is an essential step in the intoxication process, and it has been shown that low pH triggers the insertion of the activated form of PA, PA63, into model lipid bilayers. In this study, we have examined the effects of pH o…

Liquid-liquid phase separation of proteins has been observed to occur on biological membranes, where it is thought to play a role in diverse cellular behaviors. Recent work has demonstrated colocalization between protein condensates on opposing leaflets of the bilayer, suggesting that protein phase separation may be coupled across the bilayer. However, the mechanism behind this coupling phenomenon remains poorly understood. Here we seek to understand the protein-protein and protein-membrane interactions that give rise to transbilayer coupling of protein condensates. We perform coarse-grained m

The design of vehicles for transdermal gene delivery is at the forefront of molecular medicine, facilitating targeted therapies. Reports suggest that flexible liposomes can be a good alternative for transdermal delivery, and asymmetric liposomes may enhance gene delivery efficiency. This study aims to create flexible asymmetric-type liposomes with high encapsulation of DNA and high deformability rates. The synthesis of asymmetric liposomes was standardized using the inverse emulsion method, with lipids DOTMA (1,2-di-O-octadecenyl-3-trimethylammonium propane) and DOPE (1,2-dioleoyl-sn-glycero-3

Rift Valley fever virus (RVFV) is a member of the genus Phlebovirus within the family Bunyaviridae. It is a mosquito-borne zoonotic agent that can cause hemorrhagic fever in humans. The enveloped RVFV virions are known to be covered by capsomers of the glycoproteins G(N) and G(C), organized on a T=12 icosahedral lattice. However, the structural units forming the RVFV capsomers have not been determined. Conflicting biochemical results for another phlebovirus (Uukuniemi virus) have indicated the existence of either G(N) and G(C) homodimers or G(N)-G(C) heterodimers in virions. Here, we have stud

The interactions of irbesartan (IRB) and irbesartan-2-hydroxypropyl-β-cyclodextrin (HP-β-CD) complex with dipalmitoyl phosphatidylcholine (DPPC) bilayers have been explored utilizing an array of biophysical techniques ranging from differential scanning calorimetry (DSC), small angle X-ray scattering (SAXS), ESI mass spectrometry (ESI-MS) and solid state nuclear magnetic resonance (ssNMR). Molecular dynamics (MD) calculations have been also conducted to complement the experimental results. Irbesartan was found to be embedded in the lipid membrane core and to affect the phase transition properti

Lipid membrane permeability is fundamental for cellular homeostasis, signaling, and drug transport. Experimental methods exist to characterize it, from cell-based assays to more reproducible artificial, filter-supported systems. Although they closely mimic physiological environments, these assays pose challenges for quantitative analysis. Simpler models, such as supported lipid bilayers and liposomes, offer better control of experimental variables and membrane composition, making them suitable for detailed physicochemical investigations. The dynamic response of unilamellar lipid vesicles to os…

α-Synuclein (αSyn) is an intrinsically disordered protein whose interactions with lipid membranes are central to both its physiological function and its role in synucleopathies. While membrane charge, phase, and curvature are each known to influence αSyn binding, these properties are typically examined independently, leaving their combined effects on both equilibrium and dynamic membrane association unresolved. Here, we systematically investigate how membrane phase and charge jointly regulate αSyn binding, curvature sensitivity, and exchange dynamics using fluorescence microscopy, circular dic

α-Synuclein (αSyn) remodels cellular membranes through interactions that involve both its structured, membrane-binding N-terminal domain (NTD) and intrinsically disordered C-terminal domain (CTD). While the amphipathic NTD helix is known to insert into lipid bilayers and generate curvature, the contribution of the acidic CTD remains unclear. Here, we dissect the individual and complementary roles of these domains using Supported Bilayers with Excess Membrane Reservoir (SUPER) templates to quantify membrane remodeling via membrane fission and membrane morphological deformations (i.e., membrane …

A critical step in how malaria parasites invade red blood cells (RBCs) is the wrapping of the membrane around the egg-shaped merozoites. Recent experiments have revealed that RBCs can be protected from malaria invasion by high membrane tension. While cellular and biochemical aspects of parasite actomyosin motor forces during the malaria invasion have been well studied, the important role of the biophysical forces induced by the RBC membrane-cytoskeleton composite has not yet been fully understood. In this study, we use a theoretical model for lipid bilayer mechanics, cytoskeleton deformation,

We performed an NMR and EPR study of the interaction of four [Ga(3,4-HPO)3] chelates with liposomes derived from a soybean extract (SEL) and simpler formulations using POPC (100%) and POPE:POPC (50%). Parent [Fe(3,4-HPO)3] chelates are eligible to prevent Iron Deficiency Chlorosis and we took advantage of the likenesses of the ions Fe (III) and Ga (III), and the fact their metal ion complexes are isostructural, to perform a combined NMR and EPR study to get information about the permeation properties of the complexes. The results demonstrate the presence of liposomes loaded with Ga-chelates an

HYPOTHESIS: Enveloped viruses are pivotal in causing various illnesses, including influenza and COVID-19. The antimicrobial peptide LL-37, a critical part of the human innate immune system, exhibits potential as an antiviral agent capable of thwarting these viral threats. Its mode of action involves versatile and non-specific interactions that culminate in dismantling the viral envelope, ultimately rendering the viruses inert. However, the exact mechanism of action is not yet understood.
EXPERIMENTS: Here, the mechanism of LL-37 triggered changes in the structure and function of an enveloped

A majority of biomimetic membranes used for current biophysical studies rely on planar structures such as supported lipid bilayer (SLB) and self-assembled monolayers (SAMs). While they have facilitated key information collection, the lack of curvature makes these models less effective for the investigation of curvature-dependent protein binding. Here, we report the development and characterization of curved membrane mimics on a solid substrate with tunable curvature and ease in incorporation of cellular membrane components for the study of protein-membrane interactions. The curved membranes we

Curcumin, the main ingredient in turmeric, has attracted attention due to its potential anti-inflammatory, anticancer, wound-healing, and antioxidant properties. Though curcumin efficacy is related to its interaction with biomembranes, there are few reports on the effects of curcumin on the lateral motion of lipids, a fundamental process in the cell membrane. Employing the quasielastic neutron scattering technique, we explore the effects of curcumin on the lateral diffusion of the dipalmotylphosphatidylcholine (DPPC) membrane. Our investigation is also supported by Fourier transform infrared s

We have studied the impact of the lipid ratio on the membrane permeability of mixed phosphatidylcholine (POPC)/phosphatidylglycerol (POPG) membranes induced by 1-dodecyl-3-methylimidazolium bromide ([C12MIM]+Br-) ionic liquid by evaluating the role of affinity and architecture of the phospholipid bilayer. Nine different model membranes composed of negatively charged POPG and zwitterionic POPC lipids mixed in molar ratios of 9:1, 8:2, 7:3, 6:4, 5:5, 4:6, 3:7, 2:8, and 1:9 have been studied. The membrane permeability of each composition has been evaluated using fluorescence-based dye leakage ass

Gallstone disease and in particular its cholesterol type is one of the most common diseases of the digestive system. More than 10% of the population is affected by this disease and in the case of Native Indians of Peru and Chile this rate reaches as much as 100%. While the disease is well-researched from a medical point of view, the two-step mechanism of formation of gallstones is not well studied. In this work the first stage gallstone formation namely cholesterol crystallization in the bilayer of the phospholipid vesicles has been described as space confined aggregation process. The formula

The ability to observe lethal anthrax exotoxins translocating through size-constricting nanopores in vitro, combined with detailed sequence and structural data, has aided in elucidated mechanisms of exotoxin cell entry and toxicity. However, due to limited observations of anthrax exotoxins translocating through protective antigen nanopores in vitro and the instability of protective antigen-functionalized suspended lipid bilayers, questions remain regarding the native mechanisms of cell entry. Nanoporous hydrogel membranes offer a robust tool for studying protein translocation with ensemble mea…

Hanatoxin (HaTx), a 35-residue polypeptide from spider venom, functions as an inhibitor of Kv2.1 channels by interacting with phospholipids prior to affecting the voltage-sensor. However, how this water-soluble peptide modifies the gating remains poorly understood, as the voltage-sensor is deeply embedded within the bilayer. To determine how HaTx interacts with phospholipid bilayers, in this study, we examined the toxin-induced partitioning of liposomal membranes. HPLC-results from high-speed spin-down vesicles with HaTx demonstrated direct binding. Dynamic light scattering (DLS) and leakage a…
