Does physical activity and inflammation mediate the job stress-headache relationship? A sequential mediation analysis in the ELSA-Brasil study.

BACKGROUND: Evidence indicates that physical activity reduces stress and promote a myriad of health-enhancing effects through anti-inflammatory mechanisms. However, it is unknown whether these mechanisms interfere in the association between psychosocial job stress and headache disorders. OBJECTIVE: To test whether physical activity and its interplay with the systemic inflammation biomarkers high-sensitivity C-reactive protein (hs-CRP) and acute phase glycoproteins (GlycA) would mediate the associations between job stress and headache disorders. METHODS: We cross-sectionally evaluated the bas

Association between high emotional demand at work, burnout symptoms, and sleep disturbance among Korean workers: a cross-sectional mediation analysis.

We explored the mediating role of burnout on the association between EDW and sleep disturbances. Our study included 18,744 Korean workers. Respondents were grouped into four levels (none, low, moderate, and high) based on their exposure to two EDW factors: handling angry clients (HAC) and hiding emotion (HE). Mediation models were used to estimate odds ratios (OR) at a 95% confidence interval (CI). Compared to those not exposed, low, moderate, and high HAC increased odds of sleep disturbance by 1.52 (95% CI 1.30-1.78), 3.20 (95% CI 2.57-3.98), and 3.28 (95% CI 2.77-3.88) times, respectively. T

The renoprotective effect of esaxerenone independent of blood pressure lowering: a post hoc mediation analysis of the ESAX-DN trial.

Angiotensin-converting enzyme (ACE) inhibitors or angiotensin II receptor blockers (ARBs) are recommended as first-line drugs for hypertension with diabetic nephropathy owing to their renoprotective effect; however, their effect beyond lowering blood pressure (BP) has not been confirmed. Recent studies have shown that aldosterone plays a key role in causing renal injury; therefore, it is likely that mineralocorticoid receptor (MR) blockers inhibit aldosterone-induced renal damage in different ways from ACE inhibitors and ARBs. Therefore, we investigated the mechanism of the effect of an MR blo

Does asset ownership influence sexual risk-taking behaviors among women engaged in sex work in Southern Uganda? A mediation analysis.

BACKGROUND: Economic vulnerability influences women engaged in commercial sex work (WESW) to further engage in sexual risk behaviors, as they often have multiple customers and engage in unprotected sex for financial gains. This study examined asset ownership's direct and indirect impact on sexual risk-taking behaviors among WESW in Southern Uganda, a very vulnerable group of women at high risk for contracting HIV and other STIs. METHODOLOGY: We used baseline data from the Kyaterekera study, an NIH-funded study among WESW aged 18-55 across 19 HIV hotspots in Southern Uganda. Structural equatio

Counterfactual mediation analysis in the multistate model framework for surrogate and clinical time-to-event outcomes in randomized controlled trials.

In cancer randomized controlled trials, surrogate endpoints are frequently time-to-event endpoints, subject to the competing risk from the time-to-event clinical outcome. In this context, we introduce a counterfactual-based mediation analysis for a causal assessment of surrogacy. We use a multistate model for risk prediction to account for both direct transitions towards the clinical outcome and indirect transitions through the surrogate outcome. Within the counterfactual framework, we define natural direct and indirect effects with a causal interpretation. Based on these measures, we define t…

Itch as Major Mediator of Effect of Tofacitinib on Health-Related Quality of Life in Psoriatic Arthritis: A Mediation Analysis.

Patients with psoriatic arthritis (PsA) experience impaired health-related quality of life (HRQoL). Tofacitinib is an oral Janus kinase inhibitor for the treatment of PsA, which has been associated with improvements in dermatologic endpoints in patients with PsA. To assess the extent to which tofacitinib affects patient HRQoL via improvements in dermatologic symptoms, including itch, data were pooled from patients with PsA who received tofacitinib in phase III studies (NCT01866668 and NCT01882439). Mediation modeling assessed the indirect effects (via Itch Severity Item [ISI] and Physician's G

Bayesian Methods for Exposure Misclassification Adjustment in a Mediation Analysis: Hemoglobin and Malnutrition in the Association Between Ascaris and IQ.

BACKGROUND: Soil-transmitted helminth infections have been found to be associated with child development. The objective was to investigate hemoglobin levels and malnutrition as mediators of the association between Ascaris infection and intelligence quotient (IQ) scores in children. METHODS: We conducted a longitudinal cohort study in Iquitos, Peru, between September 2011 and July 2016. A total of 1760 children were recruited at 1 year of age and followed up annually to 5 years. We measured Ascaris infection and malnutrition at each study visit, and hemoglobin levels were measured as of age 3.

Does parental mediation of media influence child outcomes? A meta-analysis on media time, aggression, substance use, and sexual behavior.

The current study examined how parental mediation of media (restrictive mediation, active mediation, and coviewing) influenced child outcomes. Three meta-analyses, 1 for each type of mediation, were conducted on a total of 57 studies. Each analysis assessed the effectiveness of parental mediation on 4 pertinent child outcomes: media use, aggression, substance use, and sexual behavior. The overall results indicated small, but significant relationships between child outcomes and restrictive mediation (r+ = -.06), and coviewing (r+ = .09). Overall active mediation was nonsignificant, though activ

The structural analysis of adhesions mediated by Ep-CAM.

The epithelial cell adhesion molecule Ep-CAM is capable of mediating Ca2+-independent homotypic cell-cell adhesion when introduced into cells lacking their own means of cell-cell interactions. We used (confocal) immunofluorescent and (immuno-) electron microscopy to investigate the structural organization of Ep-CAM-mediated adhesions and their relation to other types of intercellular adhesions. Ep-CAM-transfected cell lines, cells of epithelial origin, and epithelial tissues were analyzed. In transfected L cells Ep-CAM brings the opposing intercellular membranes into a close proximity (approxi

Social Anxiety Symptoms and Body-Focused Repetitive Behaviour Severity in Adolescents: A Serial Mediation Analysis of Shame and Emotion Regulation Difficulties.

Body-focused repetitive behaviour disorders (BFRBDs), including trichotillomania (TTM) and skin-picking disorder (SPD), typically emerge during adolescence. These disorders have been conceptualized within emotion regulation models, in which body-focused repetitive behaviours (BFRBs) may function as attempts to regulate aversive emotional states. Although anxiety frequently co-occurs with BFRBDs, its association with BFRB severity is generally modest, highlighting the need to examine affective and regulatory processes. This cross-sectional study examined the associations among social anxiety, s

Exploring the pathways between green space and blue space, and cognitive function in later life: a mediation analysis in Northern Ireland.

INTRODUCTION: The pathways by which green space and blue space exposure influence cognitive function are unclear. METHODS: To investigate the association between green space and blue space exposure and cognitive function and potential mediating pathways through which this association may act, we used multilevel structural equation models and longitudinal data from Waves 1 and 2 of the Northern Ireland Cohort for the Longitudinal Study of Ageing study. Cognitive function was assessed by the Mini-Mental State Examination (MMSE). We assessed green space and blue space exposure using: (1) NDVI an

Guilt Proneness and Shame Proneness Differentially Relate to Sex Disparity in Depression Among Adolescents in China: A Longitudinal Serial Mediation Analysis.

While sex disparities in adolescent depression are well-documented, how distinct self-conscious emotions divergently impact this gap remains incompletely understood. Using a three-wave longitudinal design, Chinese adolescents (N = 2,915; Mage = 16.99, SD = 1.33; 40.07% female) completed measures of shame, guilt, maladaptive cognitive emotion regulation, and depression over 12 months. Females exhibited higher shame, guilt, maladaptive cognitive emotion regulation, and depressive symptoms. Crucially, shame mediated the sex disparity by increasing depressive risk, whereas guilt was not a signific

Effects of Transdiagnostic Cognitive Behavioural Therapy on Long-Term Quality of Life: A Causal Mediation Analysis Across Anxiety and Depressive Symptoms.

INTRODUCTION: Anxiety and depression significantly impair quality of life (QoL) in primary care (PC) patients. While transdiagnostic cognitive behavioural therapy (TD-CBT) is effective, the mechanisms underlying its long-term impact on QoL remain unclear. This study examined whether changes in anxiety and depressive symptoms mediate the effect of TD-CBT on QoL at 12-month follow-up. METHODS: Data were used from the "Psychology in Primary Care" trial (PsicAP, from its Spanish acronym), which included 1061 PC patients with anxiety and depression, randomised to TD-CBT plus treatment-as-usual (TA
