Polychlorinated biphenyls: a new type of inducer of cytochrome P-448 in the liver.

The CO-difference spectrum of microsomes from rats treated with the polychlorinated biphenyls mixture, Aroclor 1254, has an absorption maximum at 448 nm. With ethylisocyananide as the ligand for reduced microsomes, Aroclor 1254 treatment causes a shift in the 455-nm peak to 453 nm and increases the ratio of absorbance of 455 nm to that at 430 nm from 0.53, obtained with untreated rats, to 1.24. These findings are similar to those seen in rats treated with the polycyclic hydrocarbon, 3-methylcholanthrene, but differ from those that characterize cytochrome P-450 in control or phenobarbital-treat

Neuroprotective effects of sinomenine and metformin in diabetic stroke: Role of NLRP3/caspase-1 and mitophagy.

Type 2 diabetes mellitus (T2DM) significantly increases the risk and severity of cerebral ischaemia-reperfusion (IR) injury, yet effective neuroprotective treatments for diabetic stroke remain limited. This study explored whether a combination of sinomenine and metformin could offer enhanced neuroprotection in diabetic rats subjected to cerebral IR injury, and investigated the molecular pathways involved. Male rats were rendered diabetic using a high-fat diet and low-dose streptozotocin. After establishing diabetes, rats underwent transient middle cerebral artery occlusion followed by 24 h of

Protective effect of sinomenine against CCl4-induced acute liver injury through regulation of mitochondrial biogenesis.

Carbon tetrachloride (CCl4)-provoked acute liver injury (ALI) is typified by intensified apoptotic, inflammatory, and oxidative changes besides mitochondrial dysfunction. Sinomenine is an active constituent in the medicinal plant Sinomenium acutum. The main objective of this study was to determine sinomenine-induced hepatoprotection following CCl4 challenge with an emphasis on unraveling the contribution of mitochondrial biogenesis-related factors. To induce ALI, CCl4 was injected i.p. and sinomenine was orally administered at 10, 25, and 50 mg/kg. Serum factors in relation to liver dysfunctio

The impact of naloxegol on anal sphincter function - Using a human experimental model of opioid-induced bowel dysfunction.

BACKGROUND AND AIMS: Opioid treatment interferes with anal sphincter function and its regulation during defecation. This may result in straining, incomplete evacuation, and contribute to opioid-induced bowel dysfunction (OIBD). Employing an experimental model of oxycodone-induced OIBD, we hypothesized that co-administration of the peripherally acting μ-opioid antagonist naloxegol would improve anal sphincter function in comparison to placebo. METHODS: In a double-blind randomized crossover trial, 24 healthy males were assigned to a six-day treatment of oral oxycodone 15 mg twice daily in comb

Influence of sinomenine upon mesenchymal stem cells in osteoclastogenesis.

With the advent of aging, the morbidity rates of such diseases as osteoarthritis, rheumatoid arthritis, and osteoporosis has witnessed a significant increase. As a common rattan drug, sinomenine (SIN) has been widely applied for the treatment of various arthritic diseases in traditional Chinese medicine (TCM) clinics. Given that SIN has been reported to inhibit the expression of Prostaglandin E2 (PGE2) in several types of cells, in this study, the influence of SIN treatment on PGE2 expression in mesenchymal stem cells (MSCs), thereby changing the osteoprotegerin (OPG) receptor/activator for th

Mechanism of sinomenine-induced apoptosis of human liver cancer cells via GRP78 membrane translocation.

BACKGROUND: Sinomenine (SIN), the main active ingredient of Sinomenium acutum (Thunb.) Rehd. et Wils. Rhizomes, exhibits sensitive pharmacological activity against liver cancer. To elucidate the underlying mechanism, in vitro studies were conducted on human hepatocellular carcinoma (HCC) cell lines SMMC-7721 and HepG-2. METHODS AND RESULTS: SIN's effects on cell viability, apoptosis, and expression of apoptosis-related genes and proteins were assessed. Immunofluorescence staining and Western blotting evaluated GRP78 cell-surface translocation and its role in apoptosis. SMMC-7721 cells were tr

Nalfurafine reverses fentanyl-induced muscle rigidity and respiratory depression without affecting sedation in rats: decoupling respiration from sedation.

This study examines whether nalfurafine, a κ-opioid receptor (κOR) agonist, reverses fentanyl-induced muscle rigidity and respiratory depression without impacting sedation. Adult, nonsedated Sprague-Dawley rats received intravenous fentanyl bolus injection (150 μg/kg), which induced immediate coma and muscle rigidity along with a transient central apnea, followed by sustained respiratory depression. Five minutes postfentanyl, rats were treated with saline, nalfurafine (0.1 or 1 mg/kg), or the κOR agonist U50488 (1 or 10 mg/kg). Muscle rigidity, sedation, and response to painful stimuli were as

A sinomenine derivative alleviates bone destruction in collagen-induced arthritis mice by suppressing mitochondrial dysfunction and oxidative stress via the NRF2/HO-1/NQO1 signaling pathway.

Bone destruction in rheumatoid arthritis (RA) leads to significant disability, yet effective treatments are limited. Sinomenine (Sino) demonstrates anti-arthritic and bone-protective effects but requires high doses. In this study, we developed a Sino derivative, SINX, and evaluated its efficacy in RA. Safety assessments in mice confirmed its suitability for further study. In vitro, SINX inhibited osteoclast differentiation by reducing TRAP-positive cells, disrupting F-actin ring formation, and suppressing bone resorption pits, alongside downregulating osteoclast-specific genes. It also showed

Mu- and delta-opioid receptor antagonists reduce levodopa-induced dyskinesia in the MPTP-lesioned primate model of Parkinson's disease.

Long-term treatment of Parkinson's disease with levodopa is complicated by the emergence of involuntary movements, known as levodopa-induced dyskinesia. It has been hypothesized that increased opioid transmission in striatal output pathways may be responsible for the generation of dyskinesia. In this study, we have investigated the effect of blockade of opioid peptide transmission on levodopa-induced dyskinesia in a primate model of Parkinson's disease-the MPTP-lesioned marmoset. Coadministration of nonselective and mu- or delta-subtype-selective opioid receptor antagonists with levodopa resul
