Fusarium wilt of cotton, caused by the pathogenic fungal Fusarium oxysporum f. sp. vasinfectum (Fov), is a devastating disease of cotton, dramatically affecting cotton production and quality. With the increase of pathogen resistance, controlling Fusarium wilt disease has become a significant challenge. Biocontrol agents (BCAs) can be used as an additional solution to traditional crop breeding and chemical control. In this study, an actinomycete with high inhibitory activity against Fov was isolated from rhizosphere soil and identified as Streptomyces alfalfae based on phylogenetic analyses. Ne

Multifunctional autoprocessing repeats-in-toxin (MARTX) toxins are pore-forming bacterial toxins that translocate multiple functionally independent effector domains into a target eukaryotic cell. Vibrio cholerae colonizes intestinal epithelial cells (IECs) and uses a MARTX toxin with three effector domains-an actin cross-linking domain (ACD), a Rho inactivation domain (RID), and an α/β hydrolase domain (ABH)-to suppress innate immunity and enhance colonization. We investigated whether these multiple catalytic enzymes delivered from a single toxin functioned in a coordinated manner to suppress

Glutathione S-transferases (GSTs) is a family of multifunctional enzymes that are involved in detoxification of poisonous compounds. In the present paper, the Leptinotarsa decemlineata genome and transcriptome dataset were mined and 30 GST genes were identified. These GSTs belonged to cytosolic (29 genes) and microsomal (1 gene) classes. Among them 3 GSTs (LdGSTe2, LdGSTs4, and LdGSTo3) possessed splice variants. Of the 29 cytosolic LdGSTs, 3, 10, 5, 4, 4, and 1 members were classified as delta, epsilon, omega, sigma, theta, and zeta subclasses respectively, along with 2 unclassified genes. Ph…

Kaposi's sarcoma associated herpesvirus (KSHV) or human herpesvirus 8 (HHV8) is a major etiological agent for multiple severe malignancies in immune-compromised patients. KSHV establishes lifetime persistence in the infected individuals and displays two distinct life cycles, generally a prolonged passive latent, and a short productive or lytic cycle. During latent phase, the viral episome is tethered to the host chromosome and replicates once during every cell division. Latency-associated nuclear antigen (LANA) is a predominant multifunctional nuclear protein expressed during latency, which pl

Tardigrades are microinvertebrates known for their extreme resilience to ionizing radiation, desiccation, and other environmental stresses. This tolerance is partly attributed to efficient DNA repair mechanisms. Here, we biochemically characterized the apurinic/apyrimidinic endonuclease 1 from Hypsibius exemplaris (hexAPE1), a multifunctional enzyme in the base excision repair (BER) pathway. Sequence analysis confirmed that hexAPE1 belongs to the ExoIII family and shares conserved catalytic residues with human APE1. Using synthetic DNA and RNA substrates, we demonstrated that hexAPE1 exhibits

Cancer remains a global health challenge, calling for integrated strategies that combine precise diagnosis with synergistic treatment. Although glucose oxidase (GOx)-mediated starvation therapy disrupts tumor metabolism by converting glucose into cytotoxic hydrogen peroxide (H2O2), its clinical translation is hindered by inefficient delivery and insufficient monotherapy efficacy. To address these issues, we developed a multifunctional nanoplatform by encapsulating GOx within hollow porous superparamagnetic Fe3O4 nanoparticles (Fe3O4-GOx NPs) to preserve its catalytic activity and enable high l

Copper-doped mesoporous silica nanoparticles (Cu-MSNs) were developed as multifunctional carriers to construct a single-particle enzyme-nanozyme cascade system for colorimetric glucose detection. The mesoporous framework provided abundant sites for glucose oxidase (GOx) immobilization and substrate diffusion, while the incorporated Cu⁺/Cu2+ species acted as catalytic active sites that endowed Cu-MSNs with peroxidase-like activity, enabling hydrogen peroxide (H2O2) activation and 3,3',5,5'-tetramethylbenzidine (TMB) oxidation. GOx was immobilized onto amino-functionalized Cu-MSNs through electr

Spent mushroom substrate (SMS) is a major agricultural byproduct whose complex lignocellulosic matrix hinders direct reuse and poses environmental risks when stockpiled. This study evaluated whether pretreatment with plant growth-promoting rhizobacteria (PGPR) could enhance SMS as a soil amendment for blueberry cultivation. Two PGPR-treated SMS formulations, along with raw SMS and a blank control, were applied to blueberry seedlings in a 10-month greenhouse experiment. Plant height, rhizosphere soil nutrients, enzyme activities, lignocellulose fractions, and the microbial communities were moni

As a class of multifunctional biocatalysts, halohydrin dehalogenases are of great interest for the synthesis of chiral β-substituted alcohols and epoxides. There are less than 40 halohydrin dehalogenases with relatively clear catalytic functions, and most of them do not meet the requirements of scientific research and practical applications. Therefore, it is of great significance to excavate and identify more halohydrin dehalogenases. In the present study, a putative halohydrin dehalogenase (HHDH-Ra) from Rhodospirillaceae bacterium was expressed and its enzymatic properties were investigated.

The Gram-negative, rod-shaped bacterium Aeromonas hydrophila has two multifunctional 5'-methylthioadenosine/S-adenosylhomocysteine nucleosidase (MTAN) enzymes, MtaN-1 and MtaN-2, that differ from those in other bacteria. These proteins are essential for several metabolic pathways, including biological methylation, polyamine biosynthesis, methionine recycling, and bacterial quorum sensing. To gain insight into how these two proteins function, we determined four high-resolution crystal structures of MtaN-1 in its apo form and in complex with the substrates S-adenosyl-l-homocysteine, 5'-methylthi

Congenital hypothyroidism with goiter (CHG) occurring as an autosomal recessive disorder is typically due to a defect of thyroid hormone synthesis (aka dyshormonogenesis). Thyroid peroxidase (TPO) is a multifunctional, heme-containing enzyme whose activity is required, and several inactivating TPO mutations causing CHG in humans and dogs have been described. Recently, two half-sib Spanish water dog (SWD) pups were diagnosed with CHG based on clinical signs, endocrine testing, and thyroid histology. TPO enzyme activity was absent, and immuno-cross-reactive TPO was undetectable in affected-dog t

Developing multifunctional ingredients for fat reduction without quality loss is challenging. This study developed a whey protein concentrate-linoleic acid-epigallocatechin gallate (WPC-LA-EGCG) ternary complex as a butter substitute in processed cheese. Multi-spectroscopy experiments and molecular docking simulations suggested that hydrogen bonding, hydrophobic, and electrostatic interactions reconfigured protein secondary structure. At optimal EGCG levels (<0.1%), the complex showed enhanced molecular flexibility, interfacial adsorption, emulsifying activity, and stability. In vitro, the com

Lipase-catalyzed hydrolysis is generally hampered by a narrow substrate scope, poor catalytic efficiency, and surfactants are often required to form emulsions to improve enzyme-substrate contact. Developing lipase immobilization supports that achieve efficient emulsifier-free hydrolysis while retaining high loading capacity and operational stability remains a critical challenge. Herein, we report a tbo covalent organic framework (COF) as a robust support for immobilizing a commercial CalA/CalB lipase blend. Through van der Waals interactions, the COF enables an ultrahigh lipase loading of 1900

Acetylcholinesterase (AChE) has emerged not only as a cholinergic enzyme but also as a modulator of β-amyloid (Aβ) aggregation via its peripheral anionic site (PAS), making it a dual-purpose target in Alzheimer's disease. While classical AChE inhibitors provide symptomatic relief, they lack efficacy against the amyloidogenic cascade. This review highlights recent advances in multifunctional AChE pharmacophores that inhibit enzymatic activity while simultaneously interfering with Aβ aggregation, oxidative stress, metal dyshomeostasis, and neuroinflammation. Particular emphasis is placed on dual

Patatin, a multifunctional glycoprotein from potatoes, is a promising lipase for industrial applications due to its emulsifying, antioxidant, and lipid-modifying properties. However, its low expression efficiency and preference for short-chain substrates limit practical utility. Here, we addressed these challenges by heterologously expressing patatin in Pichia pastoris X-33, achieving a yield of 121 mg/L through optimized fermentation. The enzyme showed optimal activity at 35 °C and pH 10.0, with methanol enhancing activity, while Fe2+/Fe3+ inhibited it. Rational design of the D286A mutant sig

Energy restriction is closely related to cellular senescence and species longevity. Here, based on the structure and function of ATP synthase, a key enzyme for energy generation, we develop energy metabolism-engaged nanomedicines (EM-eNMs) to rejuvenate aged stromal/stem cells, and help to prevent skeletal ageing. We show that EM-eNMs infiltrate the mitochondria of aged bone marrow mesenchymal stromal/stem cells (BMMSCs), driving mitochondrial fission, mitophagy, glycolysis and maintaining BMMSC stemness and multifunction. The EM-eNMs directly bind to the ATP synthase and promote mitophagy thr

CD38 is a multifunctional receptor and enzyme present on the surface of B lymphocytes, which can induce B lymphocytes proliferation and apoptosis by crosslinking related cytokines to affect the function of B cells, thus affecting immune regulation in humans and promoting tumorigenesis. The level of CD38 expression in B cells has become an important factor in the clinical diagnosis, treatment, and prognosis of malignant tumors and other related diseases. Therefore, studying the relationship between CD38 expression on the surface of B cells and the occurrence of the disease is of great significa

During viral evolution and adaptation, many viruses have utilized host cellular factors and machinery as their partners. HBx, as a multifunctional viral protein encoded by the hepatitis B virus (HBV), promotes HBV replication and greatly contributes to the development of HBV-associated hepatocellular carcinoma (HCC). HBx interacts with several host factors in order to regulate HBV replication and evolve carcinogenesis. The cellular FADD-like IL-1β-converting enzyme (FLICE)-like inhibitory protein (c-FLIP) is a major factor that functions in a variety of cellular pathways and specifically in ap
